The expression of a stress- and salicylic acidinducible protein gene from tobacco, PR1a protein gene, was determined after its Introduction to lettuce (Lactuca sativa L.) plants. The 5 flanking 2.4 Kb fragment from PR1a gene was joined to the bacterial -glucuronidase (GUS) gene (PR-GUS) and introduced into lettuce cotyledons by Agrobacterium-mediated gene transfer using a binary vector containing a kanamycin-resistance gene as a selectable marker. As a control with constitutive expression, the chimeric gene consisting of CaMV 35S RNA promoter and GUS gene (35S-GUS) was used. An improved method for shoot formation directly from lettuce cotyledons was used effectively for transformation, shortening the time for regeneration. In 70% or more of kanamycin-resistant regenerated lettuce plants, into which PR-GUS or 35S-GUS was introduced, high GUS activity and integration of the chimeric gene into the lettuce genome were detected. By treatment with salicylic acid, GUS activity increased 3- to 50-fold in PR-GUS transformants, however, no increase was detected in 35S-GUS plants. A new approach to the origin of the genetic code is proposed based on some regularities in the nucleotide distribution pattern of the code. The relative amounts of various amino acids in primitive proteins were possibly different from those in organisms living today. The primordial ratio was supposed to shift to the modern one guided by the action of primitive nucleotides. Each primitive tRNA had a discriminator site and, distinguished from it, an anticodon site. It is also postulated that primordially each amino acid could correspond to a wide variety of codons. During the course of the evolutionary change, a selective mechanism worked among the protobionts so that less frequent nucleotides became associated with more abundant amino acids in the primordial conditions, thus finally leading to the present codon catalogue.Presented at The International Seminar: The Origin of Life held in Moscow, August 2–7, 1974.  相似文献

A moderately halophilic strain of Bacillus, isolated from unrefined solar salt, was capable of growth in the presence of 4 M NaCl. Maximal growth was obtained in a medium containing 1 to 2 M NaCl. The organism produced protease when cultivated aerobically in media containing 0 to 3 M NaCl or 0 to 2 M KCl. The protease activity was optimal at 0.5 M NaCl and 0.75 M KCl.  相似文献

Summary The Escherichia coli mutator gene mutT, which causes A:TC:G transversion, was cloned in pBR 322. mutT + plasmids carry a 0.9 kb PvuII DNA fragment derived from the E. coli chromosome. Specific labelling of plasmid-encoded proteins by the maxicell method revealed that mutT codes for a polypeptide of about 15,000 daltons. The protein was overproduced when the mutT gene was placed under the control of the lac regulatory region on a multicopy runaway plasmid. Summary An anion channel of sarcoplasmic reticulum vesicle has been incorporated into planar lipid bilayers by means of a fusion method and its basic properties were investigated. Analysis of fusion processes suggested that one SR vesicle contained approximately one anion channel. The conductance of this channel has several substates and shows a flickering behavior. The occupation probability of each substate was voltage dependent, which induced an inward rectification of macroscopic currents. Further, the anion channel was found to have the following properties. (1) The single-channel conductance is about 200 pS at 100mm Cl. (2) The channel does not select among monovalent anions but SO 4 2– hardly permeates through the channel. (3) SO 4 2– added to thecis side (the side to which SR vesicles were added) inhibits Cl current competitively in a voltage-dependent manner. (4) An analysis of this voltage dependence suggests that the binding site of SO 4 2– is located at about 36% of the way across the channel from thecis entrance.  相似文献

The effect of glucose concentration on the in vitro release of histamine (HA) was examined, using two different preparations of the mouse hypothalamus. The HA and tele-methylhistamine released from whole blocks of the hypothalamus into the medium linearly increased during 2-h incubation in normal Krebs-Ringer bicarbonate solution in the absence of external depolarizing stimuli. The release of HA from this preparation depended on the temperature and Ca2+ in the medium and was progressively increased with decrease in the glucose concentration from 11.5 to 1 mM. The rate of the HA release was dependent on the absolute concentration of glucose and not on an abrupt change in the concentration. When slices of the hypothalamus were incubated in high K+ medium, a temperature- and Ca2+-dependent HA release was observed. At low concentrations of glucose, the K+ (20 mM)-induced HA release from the hypothalamic slices was also enhanced. Tetrodotoxin (10 microM) inhibited the enhancing effect of a low glucose concentration (2 mM) on the HA release by 60%, in both preparations of the hypothalamus. The possibility that the release of HA from the mouse hypothalamus is regulated by glucose concentration and that activation of neuronal Na+ channels is involved in the enhancement of the HA release by low glucose concentrations warrants further attention.  相似文献

The Processing of a 57-kDa Precursor Peptide to Subunits of Rice Glutelin   总被引:1,自引:0,他引:1  
The processing of a 57-kDa peptide into 37- and 22-kDa subunitsof glutelin, a major storage protein of rice, was confirmedby the immunological compatibility between the precursor andglutelin subunits. The 57-kDa peptide reacted with the antiseraraised against purified 37-kDa and 22-kDa subunits of glutelin.The processing was further confirmed by alteration of an invivo protein synthesis by monensin, a sodium ionophore whichinhibits the intracellular transport of secretory and membraneproteins. Infusion of monensin into developing rice grains resultedin suppressed formation of mature glutelin subunits with concomitantaccumulation of the 57-kDa peptide. Ulex europaeus agglutinin-I (UEA-I) recognizes the Fuc alpha 1----2 Gal linkage. Receptors for UEA-I were not detected in mouse embryos until the 13th day of embryo-genesis, except for their temporary expression in early trophectoderm cells. In adult mice, UEA-I receptors were detected at various sites, including cells of the digestive tracts, the bronchial epithelium, Hassall's corpuscle of the thymus, and the skin. The fucose-binding protein of Lotus tetragonolobus (FBP) is another lectin that recognizes fucosyl residues. The distribution of FBP receptors was significantly different from that of UEA-I receptors. FBP receptors were first detected in late 8-cell embryos and were expressed in the embryonic ectoderm, visceral endoderm, and trophoblastic giant cells in egg-cylinders. At later stages, the distribution of FBP receptors became restricted to certain parts of the embryo. In the adult, the distribution of FBP receptors was more restricted than that of UEA-I receptors. Particularly in embryos before the 11th day of gestation, the distribution of FBP receptors resembled that of SSEA-1, which is defined by the Gal beta 1----4(Fuc alpha 1----3) GlcNAc linkage. From the specificity of FBP, we inferred that the disappearance of SSEA-1 and FBP receptors during embryogenesis is not the result of alpha 1----2 fucosylation of the terminal galactosyl residue in the determinant. The fact that the expression of two fucose-related cell-surface markers, i.e., UEA-I receptors and SSEA-1 (or FBP receptors), is developmentally regulated in an entirely different fashion is an excellent example illustrating the precise control of differentiation-dependent alterations in cell-surface carbohydrates.  相似文献
