Two lines of immortal human fibroblasts were isolated following transfection of TIG-3 cells with plasmid DNA, pMT-1ODtsA, that contained SV40 early gene with a deletion in replication origin and ts mutation in coding sequence for T-antigen. These cells continued proliferation at 34 degrees C, over 565 population doubling level (PDL) which is far over the limited division potential of untransformed normal TIG-3 of 70-80 PDL. When the culture temperature was shifted to 40 degrees C after 70 PDL, they ceased proliferation immediately. One of these immortal clones, SVts8, lost its ts phenotype after retransformation with wtT-antigen gene. These results indicated that the function of intact T-antigen is required for maintenance of immortal proliferation, at least in one of the SV40 transformed immortal clones.  相似文献

The results of the present experiment are shown in terms of the transport of protoheme from mitochondria to apocytochrome b5 when fresh rat liver mitochondria, apocytochrome b5, and cytosol were incubated. The heme transfer protein was purified from rat liver cytosol up to approximately 133-140-fold with a 43% yield by the procedure discussed herein, including Sephadex G-75 and CM-cellulose column chromatography. The final preparation showed apparent homogeneity upon polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate. Its native form was found to be a dimeric protein with a Mr = 45,000 which consists of a subunit with a Mr = 23,000. In the transporting system, the heme transfer depended on the concentration of mitochondria (donor), apocytochrome b5 (acceptor), and purified transfer protein, respectively. Omission of one of these components led to an almost complete loss of the transfer activity. The transport of mitochondrial protoheme was a rapid reaction which showed approximate linearity until 1.5 min and after that it became saturated. When the functional capacity was tested by the NADH-cytochrome c reductase system, the reconstituted cytochrome b5 expressed its complete original catalytic properties, as well as its characteristic absorption spectra for the hemoprotein. Furthermore, the detailed physicochemical and immunological characterization of the transfer protein provided evidence that the protein is identical with soluble glutathione S-transferase, which conjugates glutathione with a variety of electrophilic compounds. At least one of the glutathione S-transferase isozymes observed was identified as GST-C2, which comprises the subunit of Yb'Yb' by the immunoprecipitation reaction using various anti-glutathione S-transferase isozyme antibodies.  相似文献

Acute and chronic effects of γ-butyrolactone-γ-carbonyl-histidyl-prolinamide (DN-1417) were investigated on motor activity, dopamine (DA) metabolites and DA receptors in various brain regions of rats. The motor activity, as measured with Automex recorder, was enhanced after a single injection with DN-1417 (20 mg/kg, IP), and the motor stimulating action persisted during 21 daily injections. Acute DN-1417 elevated both homovanillic acid (HVA) and 3,4-dihydroxyphenylacetic acid (DOPAC) levels in 7 brain regions, prefrontal cortex polar, medial and lateral fields, nucleus accumbens, olfactory tubercles, amygdala and striatum. After chronic treatment for 7 days, the acute effect of DN-1417 on DA metabolites disappeared in all regions except for the striatum in which DN-1417 still increased HVA and DOPAC. The response of striatal DA metabolites was also observed after chronic treatment for 21 days. Chronic DN-1417 produced no significant change in 3H-spiperone binding in the prefrontal cortex, nucleus accumbens, olfactory tubercles and striatum, while striatal 3H-DA binding displaced by 30 nM spiperone was enhanced after chronic treatment. These results indicate that DN-1417 interacts with mesocortical, mesolimbic and nigrostriatal DA systems in the different modes of action. The lack of tolerance to motor hyperactivity, however, raises the question as to whether DN-1417-induced hyperactivity may be mediated by the activation of mesolimbic DA neurons. The involvement of nigrostriatal neurons in DN-1417-induced motor hyperactivity is suggested.  相似文献

Transplantable human oat cell carcinoma cells of the lung with ectopic vasopressin production were incubated with labeled amino acids and immunoreactive neurophysins in cell extracts were analyzed by isoelectric focusing. When the cells were incubated with L-(35S)-cysteine for 20 h, one major peak (isoelectric point; pI=5.3) and several minor peaks (pI=6.1, 5.7, 5.1, 4.9 and 4.7) of labeled proteins were observed. On sodium dodecyl sulfate-polyacrylamide gel electrophoresis, the relative molecular mass (Mr) of the pI 5.7 protein was estimated to be 20,000 and that of the pI 6.1 species to be 19,000, while the remainder had a Mr of approximately 10,000. The result of the pulse-labeling experiment has clearly shown that the pI 5.7 and 6.1 proteins, which have affinity for concanavalin A, are biosynthetic precursors for the smaller form of neurophysin with a pI 5.3. When subjected to limited proteolysis with trypsin, the pI 5.7 protein generated a Mr 10,000 protein and a smaller peptide. The Mr 10,000 protein thus produced was identified as neurophysin on the basis of its pH-dependent affinity for vasopressin and the migration pattern on isoelectric focusing. The smaller peptide coeluted with synthetic arginine vasopressin and bound to neurophysin suggesting that it possesses a cysteine-tyrosyl sequence at its N-terminus. Similarly, the pI 6.1 protein liberated neurophysin and vasopressin-like peptide after incubation with trypsin. These results suggests that the glycosylated protein with a pI of 5.7 and a Mr of 20,000 is the common precursor to vasopressin and neurophysin in human oat cell carcinoma of the lung with ectopic vasopressin production. The pI 6.1 protein may be an intermediate in the conversion of the precursor to vasopressin and neurophysin.  相似文献

Senescent human diploid fibroblasts, TIG-1, had labelling indices of about 0.5-3% when labelled with [3H]thymidine for 3 days in fresh medium containing 10% fetal bovine serum. When these cells were infected with SV40, the percentage of nuclei incorporating [3H]thymidine increased by about 10-fold. The frequency of T-antigen-positive cells and that of [3H]thymidine-incorporating cells were almost the same. About 80% of T-antigen-positive cells were also positive to incorporation of [3H]thymidine, and the same result was obtained in infected young cells. These results indicated that senescent human diploid cells which are brought to synthesize T-antigen always initiate DNA synthesis as young cells do. The characteristics of senescent cells as compared with younger cells was low incidence of T-antigen-positive cells after infection. The basis of low susceptibility of senescent cells to initiate DNA synthesis by SV40 infection thus seems to be concerned with an event after the adsorption of virus, but before the synthesis of a detectable amount of T-antigen.  相似文献

We discovered a new dental mutation causing morphological abnormalities of the teeth in the SHR-SP strain of rats, maintained in the Research institute, Daiichi Seiyaku, Co., Ltd. The incisors of rats with this mutation are white in color, lacked hardness and were easily worn. The incisors broke or became bent with extended growth. Histological examination of the abnormal rats revealed amelogenesis imperfecta due to underdevelopment of tooth ameloblasts. The results of mating tests showed that this characteristic was inherited as an autosomal single recessive trait. We named this gene amelogenesis imperfecta (ami). The rats will prove to be a valuable models for studies on enamel formation.  相似文献

To determine if erythropoietin influences megakaryocytopoiesis, the purified recombinant human hormone (rEpo) was added to serum-free liquid cultures of murine marrow. A dose-related increment in acetylcholinesterase (AchE) production was observed. To assess if increments in this relatively megakaryocyte-specific enzyme marker were mediated by a direct hormone-megakaryocyte interaction rather than via an accessory cell population, rEpo was added to cultures of isolated single megakaryocytes. A significant, dose-related increase in cell size was noted in the presence of the hormone, accompanied by a high probability of an increase in cellular DNA content. The data show that rEpo does directly influence some aspects of megakaryocytic maturation, although the physiologic significance of this effect remains unknown.  相似文献
