The effect of temperature on the rate of ADP-stimulated respiration of mitochondria from dog, rabbit, pig, and human kidney cortex mitochondria was plotted according to the Arrhenius relationship. The temperature at which the plot demonstrated a break was at 15 °C for mitochondria from dog, pig, and human kidneys. The discontinuity occurred at 10 °C or less for mitochondria from rabbit kidneys. This difference suggests that mitochondria from rabbit kidneys undergo a lipid-phase transition at lower temperatures than for other species commonly used in experimental renal preservation. The implications of this difference suggest caution in using results obtained with rabbit kidneys for comparison to results obtained from hypothermic renal preservation of other species kidneys. Apparent fluidization of dog kidney mitochondrial membranes with adamantine abolished the discontinuity in the Arrhenius plot.  相似文献

Antigenic expression of B-cell chronic lymphocytic leukemic lymphocytes   总被引:2,自引:0,他引:2  
A flow cytometric analysis of lymphocytes in B-cell chronic lymphocytic leukemia (B-CLL) samples and in monocyte-depleted and T-cell-depleted normal peripheral blood (B-PBL) samples was undertaken using 129 reagents from the blind panel (BP) and 72 reagents from the cluster designation (CD) panel obtained from the Fourth International Leucocyte Differentiation Conference and Workshop, B-Cell Section. After determining the average mean channel fluorescence and the average percentage of positive cells for the B-CLL and the normal B-PBL preparations, a combined ratio and difference analysis was performed for each monoclonal antibody reactivity. This analysis confirmed the intense expression of class II antigens on B-CLL and the preferential expression of CD19, CD20, CD23 and CD24 antigens. In addition, three new clustered and three new unclustered antigens were also preferentially expressed on B-CLL lymphocytes. Cluster analysis of these differences suggests the existence of at least three overlapping immunophenotypic subpopulations, composed of CD19, CD20, CD21, CD22, CD23, CD24, CD75, CD76 and CDw78.  相似文献

A number of screws commonly used for internal fixation in scaphoid bone fractures and nonunions are compared regarding biomechanical properties and clinical applicability. The experiments were carried out on models made of ash-wood, representing a reconstruction and fixation as is performed in a cortico-cancellous inlay bone graft for scaphoid non-union. For fixation use was made of 2.7 and 3.5 AO/ASIF cortical screws respectively, 4.0 AO/ASIF cancellous screws, Herbert screws, and a newly designed screw called the three components screw (D.K.S.). The models with implanted screws were tested for bending strength, tensile strength and torsion stability. No large differences between the various screws were found regarding the measured parameters, so that a small intra-osteal implant such as the Herbert screw and the D.K.S., which can be inserted easily and which gives a certain amount of interfragmentary compression, will be sufficient for osteosynthesis of the scaphoid bone. In case an intra-osteal implant is not available a single 3.5 AO/ASIF cortical screw, inserted following lag-screw principles, is recommended.  相似文献

Directly upstream of the Lactococcus lactis subsp. cremoris Wg2 proteinase gene is an oppositely directed open reading frame (ORF1). The complete nucleotide sequence of ORF1, encoding a 33-kilodalton protein, was determined. A protein of approximately 32 kilodaltons was synthesized when ORF1 was expressed in Escherichia coli by using a T7 RNA polymerase-specific promoter. L. lactis subsp. lactis MG1363 transformants carrying the proteinase gene but lacking ORF1 were phenotypically proteinase deficient, unlike transformants carrying both the proteinase gene and ORF1. Synthesis and secretion of proteinase antigen by L. lactis could be detected with proteinase-directed monoclonal antibodies regardless of whether ORF1 was present. The requirement of ORF1 for proteinase activation was reflected in a reduction in the molecular weight of the secreted proteinase. Furthermore, deletion of the 130 C-terminal amino acids of the Wg2 proteinase prevented attachment of the enzyme to lactococcal cells.  相似文献

Large DNA inversions caused by an intrachromosomal recombination between homologous regions located in intron 22 and 5 of the factor VIII gene have recently been identified in patients with severe haemophilia A. To evaluate better the prevalence of this large inversion and to estimate the overall sensitivity of the Southern blot/hybridization method we analysed the factor VIII gene of 49 unrelated patients with severe haemophilia A. All patients were screened for the inversion mutation, TaqI site mutations, and deletions. Mutations were identified in 31 (63%) patients, and comprised 24 large inversions, 4 partial deletions, and 3 point mutations. Three different haplotypes were characterised in the patients presenting the inversion mutation, confirming its independent origin. Two novel deletions are reported: a large one spanning from intron 14 to intron 22 and a deletion of 86 bp comprising the 3 region of exon 1 and 39–41 bp of intron 1. DNA sequencing of the deletion junction showed no significant homology between normal 5 and 3 sequences around the breakpoints. A novel missense mutation is also reported: CGAGGA, Arg-2209 to Gly. These results confirm that the inversion mutation is the most common cause of severe haemophilia A and indicate that the Southern blot/hybridization assay should be used as the first method for screening of mutations in severe haemophilia A.  相似文献

The symbioses between plants of the Rubiaceae and Primulaceae families with Burkholderia bacteria represent unique and intimate plant–bacterial relationships. Many of these interactions have been identified through PCR-dependent typing methods, but there is little information available about their functional and ecological roles. We assembled 17 new endophyte genomes representing endophytes from 13 plant species, including those of two previously unknown associations. Genomes of leaf endophytes belonging to Burkholderia s.l. show extensive signs of genome reduction, albeit to varying degrees. Except for one endophyte, none of the bacterial symbionts could be isolated on standard microbiological media. Despite their taxonomic diversity, all endophyte genomes contained gene clusters linked to the production of specialized metabolites, including genes linked to cyclitol sugar analog metabolism and in one instance non-ribosomal peptide synthesis. These genes and gene clusters are unique within Burkholderia s.l. and are likely horizontally acquired. We propose that the acquisition of secondary metabolite gene clusters through horizontal gene transfer is a prerequisite for the evolution of a stable association between these endophytes and their hosts.  相似文献

The amount of rain which falls sporadically in the relatively dry season at the time that the white rice borer is diapausing as a full grown larva in the rice stubble, has a very important influence on the size of population of the borers in the following wet season when rice is grown. If this dry season is wet or very wet (according to our standard, based on the figures of rainfall and borer damage over 26 years in five regions of Eastern Java), no damage can be expected in the following planting season. If the dry season is really dry or very dry, outbreaks of borers may occur (in the period under survey this occurred in nine out of fourteen years). We have tried to give an outline of a method of prediction of borer damage in the planting season, based on the rainfall dates of the foregoing dry season.
Zusammenfassung Der Weisse Reisbohrer (Scirpophaga innotata Wlk.) kommt nur in Gebieten vor, wo jährlich eine ausgeprägte Regen- und Trockenzeit (Monsun) auftritt. Am Ende der Regenzeit gehen die vollerwachsenen Raupen, die im Stengel der reifenden Reispflanze leben, in Diapause. Während der folgenden relativ trockenen Jahreszeit, wenn nur gelegentlich Regenschauer fallen, ruhen die Raupen in der Stoppel. Bald nachdem die ersten heftigen Regenfälle der neuen Regenzeit einsetzen, verpuppen sich die Larven und alle Falter schlüpfen gemeinsam zum Stoppelflug.Dieser Zünsler ist in gewissen Gebieten Javas einer der Hauptschädlinge des Reises, obwohl sein Auftreten sehr unregelmäßig ist. Jahrelang war man allgemein der Auffassung, daß relativ reichlicher Regenfall während der Trockenzeit die Reisbohrerpopulation erheblich vermindere und daß in diesem Falle keine schweren Schäden durch Reisbohrerbefall erwartet werden können.Während eines Zeitraums von 26 Jahren (1915–1940) wurden in fünf Gebieten Ost-Javas sowolhl der Niederschlag wie der Bohrerschaden untersucht und Berechnungen der jährlichen Niederschlagsmenge in der Trockenzeit und des Bohrerschadens verglichen. Dies erhärtete die alte Auffassung, daß nach einer relativ feuchten oder sehr feuchten Trockenzeit kein Bohrerbefall von Bedeutung während des Beobachtungszeitraumes auftritt. Dagegen folgten in neun von 14 Jahren mit einer trockenen oder sehr trockenen Trockenzeit schwere oder ziemlich schwere Ausbrüche der Weißen Reisstengelbohrer während der folgenden feuchten Jahreszeiten. Obwohl auch andere Faktoren von einiger Bedeutung sein dürften, wird geschlossen, daß die Niederschlagsmenge während der Trockenzeit einen sehr entscheidenden Einfluß auf die Möglichkeit des Auftrétens einer Reisbohrer Kalamität im darauffolgenden Jahre hat. Diesen Daten entsprechend wird auf der Grundlage der Niederschlagszahlen in der vorangegangenen Trockenzeit ein Prognose-Schema entwickelt, wann Bohrerschaden zu erwarten ist.
