A medium composed of (in g/100 ml) Tryptose broth with thiamine (Difco), 2.6; cysteine-HCl, 0.12; glucose, 1; FeSO4, 7H2O, 0.005; KCl, 0.02; histidine, 0.1; tris(hydroxymethyl)aminomethane (tris) buffer, 0.3; and agar, 1; will support rapid growth of the fully virulent SCHU-S4 strain of Pasteurella tularensis. Although the test organism grew rapidly on medium from which KCl and tris buffer were omitted, these two components increased the stability of the medium upon storage at 4 C. It was necessary to (i) control carefully the relative concentration of the ferrous iron and cysteine-HCl, (ii) incubate the prepared medium overnight prior to use, and (iii) incubate the inoculated plates in an atmosphere of high relative humidity. Rapid growth of the organism was obtained also from very small inocula in the liquid form of the medium. Biochemical studies designed to elucidate the mechanisms involved in the enhancement of growth of P. tularensis in this relatively simple blood-free medium were initiated. 相似文献

Summary Cell-attached patch clamp recordings from unfertilized oocytes of the ascidianBoltenia villosa reveal an ion channel which is activated by mechanical deformation of the membrane. These channels are seen when suction is applied to the patch pipette, but not in the absence of suction or during voltage steps. The estimated density of these stretch-activated channels is about 1.5/m2, a figure equal to or greater than the density of known voltage-dependent channels in the oocyte. Ion substitution experiments done with combined whole-cell and attached patch recording, so absolute potentials are known, indicate that the channel passes Na+, Ca2+ and K+, but not Cl–. The channel has at least two open and two closed states, with the rate constant that leaves the longer-lived closed state being the primary site of stretch sensitivity. External Ca2+ concentration affects channel kinetics: at low calcium levels, long openings predominate, whereas at high calcium virtually all openings are to the short-lived open state. In multiple channel patches, the response to a step change in suction is highly phasic, with channel open probability decreasing over several hundred milliseconds to a nonzero steady-state level after an initial rapid increase. This channel may play a role in the physiological response of cells of the early embryo to the membrane strains associated with morphogenetic events. 相似文献

Abstract: A polyclonal antibody for the NMDA receptor subunit NR2D has been developed that identifies an ∼160-kDa band on immunoblots from NR2D transfected cells and CNS tissues. No cross-reactivity is seen with other NMDA receptor subunits. The NR2D receptor subunit is N -glycosylated in both brain and transfected cells. Transfected cells expressing NR2D are immunofluorescently labeled, whereas untransfected cells or cells transfected with other NMDA receptor subunit cDNAs are not. Similarly, the NR2D subunit is selectively and quantitatively immunoprecipitated, whereas the NR1, NR2A, or NR2B subunit is not. The relative densities of the NR2D subunit in nine areas of postnatal day 7 and adult rat brains have been determined by quantitative immunoblotting. NR2D was expressed at highest levels in the thalamus, midbrain, medulla, and spinal cord, whereas intermediate levels of this subunit were found in the cortex and hippocampus. Low or undetectable levels were seen in the olfactory bulb, striatum, and cerebellum. Following a peak after the first week of birth, NR2D protein levels decreased by about twofold in adulthood in all rat brain regions examined. More complete ontogenic profiles were determined for the diencephalon, telencephalon, and spinal cord where similar ontogenic patterns were seen. NR2D protein is present at high levels at embryonic stages of development, rises to a peak at postnatal day 7, and decreases but remains measurable during late postnatal life. This study demonstrates the generation and characterization of an antibody selective for the NR2D NMDA receptor subunit as well as a determination of the distribution and ontogenic profile of this subunit in rat brain. We have cloned the first bifunctional gene dihydrofolate reductase-thymidylate synthase (DHFR-TS) from a free-living, ciliated protozoan,Paramecium tetraurelia, and determined its macronuclear sequence using a modified ligation-mediated polymerase chain reaction (PCR) that can be of general use in cloning strategies, especially where cDNA libraries are limiting. While bifunctional enzyme sequences are known from parasitic protozoa, none had previously been found in free-living protozoa. The AT-rich (68%) coding region spanning 1386 bp appears to lack introns. DHFR-TS localizes to a 500 kb macronuclear chromosome and is transcribed as an mRNA of 1.66 kb, predicted to encode a 53 kDa protein of 462 residues. The N-terminal one-third of the protein is encoded by DHFR, which is joined by a short junctional peptide of 12 amino acids to the highly conserved C-terminal TS domain. Among known DHFR-TS sequences, theP. tetraurelia gene is most similar to that fromToxoplasma gondii, based on primary sequence and parsimony analyses. Variation in male dispersal and behavior patterns are components of intraspecific differences in social systems. A comparison of male behavior in different social settings can be useful for determining which behavioral mechanisms contribute to variability in social systems. Two heterosexual multimale groups and one all-male group of mountain gorillas (Gorilla gorilla beringei) were observed for over 1100 h at the Karisoke Research Centre, Rwanda. Data on proximity patterns, dominance relationships, aggression, agonistic interventions, affiliation, and homosexual behavior were compared among the males in these groups to examine the influence of female presence, sex ratio, group size, and kinship on male—male interactions. Males in the all-male group stayed closer together, affiliated more, exhibited more homosexual behavior, and were more aggressive toward each other than males in heterosexual groups. However, the males in heterosexual groups showed more wounding and more consistent dominance relationships. Kinship did not influence male-male relationships in the all-male group. The males in the heterosexual groups rarely interacted with one another; they may actively avoid close proximity to reduce aggression. Results suggest that the variable social system of mountain gorillas may be more strongly influenced by demographic factors, male-female social relationships, and male-male competition for mates than by any benefits of male-male relationships. 相似文献

The distribution of two repetitive DNA probes Sat-121 and PB6-4, specific for the section Procumbentes of the genus Beta, was tested in 16 B. patellaris monosomic addition families using a dot-blot hybridization procedure. All monosomic additions were accurately distinguished from diploid sib plants with both DNA probes. The probe PB6-4, with the strongest signal after hybridization, was selected for rapid screening of an extensive number of putative monosomic additions in B. patellaris or B. procumbens addition families using a squash-blot hybridization procedure. The probe PB6-4 detected 118 monosomic additions in 640 plants (18.4%) in eight different B. procumbens addition families. The addition family with chromosome 4 of B. procumbens was semi-lethal and could not be tested. The distribution of PB6-4 in B. patellaris addition families was confirmed in 63 addition families using the squash-blot procedure. In 4580 plants of these addition families, 628 individual monosomic additions (13.7%) were found. The relationship of the morphological characteristics of monosomic addition plants to the results of the squash-blot hybridization (plants with signal) using probe PB6-4 is quite rigorous but not complete. The correlation between plants with a signal and chromosome number (2n=19) is complete. These results indicate that sequences present on PB6-4 are probably present on all chromosomes of B. patellaris and B. procumbens. The possibility of utilizing the sequence information of Sat-121 for a PCR-based assay to screen for putative monosomic addition plants was also investigated as an alternative to chromosome counting. The DNA-amplification profiles using the primers REP and REP.INV clearly distinguished monosomic addition plants from their diploid sibs. 相似文献

The characterization of the human antiserum designated MAN has led to the identification of a subset of non-lamin proteins that are exclusively located at the nuclear periphery in all vertebrate cell types examined, from human to fish. Immunoreactive protein species were whown to comprise three major polypeptides of M
r 78000, 58000 and 40000. These antigens co-partitioned with the nuclear lamina during in situ isolation of nuclear matrices from lamin A/C-positive and-negative mammlian cells. Using double immunofluorescence, the spatial relationship of MAN antigens to type-A and type-B lamins was further examined throughout the cell cycle of lamin A/C-positive mammalian cells. In interphase HeLa and 3t3 cells, MAN antigens colocalized with both types of lamins at the periphery of the nucleus, but were absent from intranuclear foci of lamin B. As HeLa cells proceeded into mitosis, MAN antigens were seen to segregate from lamins A/C and coredistribute with lamin B. Lamins A/C disassembled during late prophase/early prometaphase and reassociated with chromatin in telophase/cytokinesis. In contrast, MAN antigens and lamin B dispersed late during prometaphase and reassembled on chromosomes in anaphase. Altogether, our data suggest that MAN antigens may play key functions in the maintenance of the structural integrity of the nuclear compartment in vertebrate cells. 相似文献

Our objective is to test an optimality model of human fertility that specifies the behavioral requirements for fitness maximization
in order (a) to determine whether current behavior does maximize fitness and, if not, (b) to use the specific nature of the behavioral deviations from fitness maximization towards the development of models of evolved
proximate mechanisms that may have maximized fitness in the past but lead to deviations under present conditions. To test
the model we use data from a representative sample of 7,107 men living in Albuquerque, New Mexico, between 1990 and 1993.
The model we test proposes that low fertility in modern settings maximizes number of grandchildren as a result of a trade-off
between parental fertility and next generation fertility. Results do not show the optimization, although the data do reveal
a trade-off between parental fertility and offspring education and income.
We propose that two characteristics of modern economies have led to a period of sustained fertility reduction and to a corresponding
lack of association between income and fertility. The first is the direct link between costs of investment and wage rates
due to the forces of supply and demand for labor in competitive economies. The second is the increasing emphasis on cumulative
knowledge, skills, and technologies in the production of resources. Together they produce historically novel conditions. These
two features of modern economies may interact with evolved psychological and physiological mechanisms governing fertility
and parental investment to produce behavior that maximizes the economic productivity of lineages at the expense of fitness.
If cognitive processes evolved to track diminishing returns to parental investment and if physiological processes evolved
to regulate fertility in response to nutritional state and patterns of breast feeding, we might expect non-adaptive responses
when returns from parental investment do not diminish until extremely high levels are reached. With high economic payoffs
from parental investment, people have begun to exercise cognitive regulation of fertility through contraception and family
planning practices. Those cognitive processes maynot have evolved to handle fitness trade-offs between fertility and parental investment.

Hillard S. Kaplan is an Associate Professor of Anthropology at the University of New Mexico. His earlier research and publications
focused on food sharing, time allocation, parental investment, and reproductive strategies among Ache hunter-gatherers in
Paraguay, Machiguenga and Piro forager-horticulturalists in Peru, and villagers of several ethnicities in Botswana. New research
and theory concern fertility, parental investment, and mating strategies in developed and developing nations. This research
formulates a new theory of reproductive decision-making and the demographic transition, integrating human capital and parental
investment theory in a synthesis of economic and evolutionary approaches.
Jane B. Lancaster is a Professor of Anthropology at the University of New Mexico. Her research and publications are on human
reproductive biology and behavior, especially human parental investment; women’s reproductive biology of pregnancy, lactation,
and child-spacing; and male fertility and investment in children. Current research with Hillard S. Kaplan is on male life
history strategies among a large sample of men in New Mexico. She has coedited three books on human parental investment:School-Age Pregnancy and Parenthood (with B. Hamburg),Parenting across the Life Span (with J. Altmann, A. Rossi, and L. Sherrod), andOffspring Abuse and Neglect (with R. Gelles). She is scientific editor of a quarterly journal,Human Nature: An Interdisciplinary, Biosocial Perspective published by Aldine de Gruyter. She is also a council member of the newly formed Human Behavior and Evolution Society.
John A. Bock is Andrew W. Mellon Post-Doctoral Fellow in Epidemiology and Population Health at the National Centre for Epidemiology
and Population Health, The Australian National University. His research focuses on the allocation of parental investment and
the determinants of children’s activities, integrating aspects of economic and evolutionary theory. He has ongoing field research
with Bantu and Bushmen agro-pastoralists and forager-horticulturalists in the Okavango Delta, Botswana. He is also collaborating
with Lancaster and Kaplan on the determinants of progeny distribution and homosexuality among New Mexican men.
Sara E. Johnson is a Ph.D. candidate at the University of New Mexico. Her major research trajectory focuses on trade-offs
in life history characters. Her research experience includes participation in a study of variation in growth and development
among children in a multi-ethnic community in the Okavango Delta, Botswana, in addition to her dissertation work on individual
variation in growth and mortality among juvenile baboons. She is collaborating with Lancaster and Kaplan on the association
