Sterile Mediterranean fruit fly, Ceratitis capitata (Wied.), males were evaluated as vectors to spread Beauveria bassiana (Bals) conidia to wild C. capitata populations under field conditions. The inoculated sterile males were released by air, using the chilled adult technique over 7000 ha of coffee growing in Chimaltenango, Guatemala, Central America. The impact of releases was determined using dry traps baited with a food attractant. The effects of these releases on Apis mellifera, Linnaeus (honey bee), Hypothenemus hampei, Ferrari (coffee berry borer) and the parasitic mite Varroa destructor (Oudeman) were also evaluated. Inoculated sterile males were able to transmit fungal spores to 44% of the wild C. capitata flies captured in traps, which likely were infected through intra- and intersexual interactions during leks, mating or mating attempts. There was no transmission of the fungal spores to non-target insect species such as coffee berry borer, honey bees or varroa. We conclude that sterile males of Mediterranean fruit fly inoculated with B. bassiana can act as effective vectors of conidia to wild populations, constituting a safe, environmentally friendly and selective alternative for suppressing the medfly under a Sterile Insect Technique-based IPM approach. 相似文献

The genus Urochloa P. Beauv. presents a prominent role in the tropical agricultural scenario being composed of species with different ploidy levels. Studies on the genomic relationship within this genus as well as specific analysis involving epigenetic marks are limited. The aim of the present study was to identify the cytosine methylation (5-mCyt) and histone H3 lysine 9 dimethylation (H3k9me2) in the different modulations of 45S ribosomal DNA (rDNA) sites in interphase nuclei and to associate these results with gene expression analysis in Urochloa ruziziensis (2n = 4x = 36), Urochloa brizantha cv. Marandu (2n = 4x = 36), and their respective interspecific hybrid H1863 (2n = 4x = 36). Immunolocalization techniques were performed in combination with Fluorescence in situ hybridization (FISH) for the location of the 45S rDNA sites. Predominantly, we observed intra- and perinucleolar sites, mostly hypomethylated and/or hyper/hypomethylated, decondensed or partially condensed. The gene expression analysis was performed qualitatively through the conventional PCR using complementary DNA and confirmed by the RT-qPCR technique and primers designed for the ITS-1 region of U. brizantha and U. ruziziensis. The molecular analyses performed on leaves showed that there is dominance of U. brizantha 45S rDNA gene expression on U. ruziziensis in the H1863 hybrid. In roots, the analyses showed that the 45S rDNA genes of the two parents are expressed in the hybrid genome. Thus, it is plausible to infer a tissue-specific nuclear dominance model in which the pattern of hypermethylated cytosine sites with heterochromatic marks and, therefore, silenced were mostly inherited from U. ruziziensis, whereas the rDNA originated from U. brizantha was characterized by cytosine and H3k9 hypomethylation.
The environmental unsustainability of African oil palm crops is a growing worldwide concern, and the macauba palm in tropical Southern America surges as the most viable substitute for the production of first-generation biodiesel as a renewable fuel. Large-scale cultivation of the macauba palm, however, is bound to suffer a major setback due to the fast emergence of a florivorous pest threat, Cyclocephala forsteri, a scarab beetle species that can severely reduce fruit yield. Like other congenerics, female and male C. forsteri are night-foraging and driven in large numbers to macauba palm inflorescences by specific volatile organic compounds. In the present study, we assessed the kairomonal cues involved in this plant–florivore interaction and their potential application in selective pest management strategies. Headspace samples of the floral fragrance of macauba palms at our study site in Central Brazil were largely dominated by 4-methylanisole (>97% relative amount), which along with the minor constituents 2-isopropyl-3-methoxypyrazine and 2-sec-butyl-3-methoxypyrazine triggered electroantennographic responses from both female and male C. forsteri. Field behavioural assays evidenced that beetles of this species were exclusively attracted to scented traps baited with 4-methylanisole. Increased total scent discharge attained with an ultrasonic piezoelectric diffuser has positively influenced attractiveness efficiency of the trapping setup. 4-Methylanisole is hereby identified as yet another volatile kairomone involved in highly selective attraction of potential pest florivorous cyclocephaline scarabs associated with economically exploitable palms and should be viewed as a key element in short-term integrated management plans for the cultivation of the macauba palm in South America. 相似文献
In this work, porous scaffolds obtained from the freeze-drying of pectin/chitosan polyelectrolyte complexes were prepared and characterized by FTIR, SEM and weight loss studies. Additionally, the cytotoxicity of the prepared scaffolds was evaluated in vitro, using human osteoblast cells. The results obtained showed that cells adhered to scaffolds and proliferated. The study also confirmed that the degradation by-products of pectin/chitosan scaffold are noncytotoxic. 相似文献
The ancient renin-angiotensin system (RAS) was discovered more than a hundred years ago by identifying the rate-limiting enzyme of the system and its relevance to blood pressure regulation. Forty years ago, Detlev Ganten et al. postulated the existence of a tissue RAS. In these forty years, he kept developing the knowledge of these systems either directly or by training or attracting the interest of many researchers. Through the present review, we try to highlight recent advancements that originated from the postulation of local brain RAS. Intraperitoneal injection of CCK reduces food intake and triggers a behavioral pattern similar to natural satiation. Reduction of food intake by CCK is mediated by vagal afferents that innervate the stomach and small intestine. These afferents synapse in the hindbrain nucleus of the solitary tract (NTS) where gastrointestinal satiation signals are processed. Previously, we demonstrated that intraperitoneal (IP) administration of either competitive or noncompetitive N-methyl-d-aspartate (NMDA) receptor antagonists attenuates reduction of food intake by CCK. However, because vagal afferents themselves express NMDA receptors at both central and peripheral endings, our results did not speak to the question of whether NMDA receptors in the brain play an essential role in reduction of feeding by CCK. We hypothesized that activation of NMDA receptors in the NTS is necessary for reduction of food intake by CCK. To test this hypothesis, we measured food intake following IP CCK, subsequent to NMDA receptor antagonist injections into the fourth ventricle, directly into the NTS or subcutaneously. We found that either fourth-ventricle or NTS injection of the noncompetitive NMDA receptor antagonist MK-801 was sufficient to inhibit CCK-induced reduction of feeding, while the same antagonist doses injected subcutaneously did not. Similarly fourth ventricle injection of d-3-(2-carboxypiperazin-4-yl)-1-propenyl-1-phosphoric acid (d-CPPene), a competitive NMDA receptor antagonist, also blocked reduction of food intake following IP CCK. Finally, d-CPPene injected into the fourth ventricle attenuated CCK-induced expression of nuclear c-Fos immunoreactivity in the dorsal vagal complex. We conclude that activation of NMDA receptors in the hindbrain is necessary for the reduction of food intake by CCK. Hindbrain NMDA receptors could comprise a critical avenue for control and modulation of satiation signals to influence food intake and energy balance. 相似文献
Testis-mediated gene transfer (TMGT) has been used as in vivo gene transfer technology to introduce foreign DNA directly into testes, allowing mass gene transfer to offspring via mating. In this study, we used plasmid DNA (pEGFP-N1) mixed with dimethylsulfoxide (DMSO), N,N-dimethylacetamide (DMA) or liposome (Lipofectin) in an attempt to improve TMGT. Males receiving consecutive DNA complex injections were mated to normal females to obtain F0 progeny. In vivo evaluation of EGFP expression, RT-PCR and PCR were used to detect the expression and the presence of exogenous DNA in the progeny. We also evaluated possible testicular damage by histological procedures. PC R and RT-PCR analyses revealed that liposome and DMSO increased the rate of TMGT. Histological analyses demonstrated that repeated (4 times) injections of DNA complexes can affect spermatogenesis. DMSO was the most deleterious among the reagents tested. In this study, we detected the presence of transgene in the progeny, and its expression in blood cells. Consecutive injections of DNA complexes were associated with impaired spermatogenesis, suggesting requirement of optimal conditions for DNA delivery through TMGT. 相似文献
