T cell Ig- and mucin-domain-containing molecules (TIMs) comprise a recently described family of molecules expressed on T cells. TIM-3 has been shown to be expressed on murine Th1 cell clones and has been implicated in the pathogenesis of Th1-driven experimental autoimmune encephalomyelitis. In contrast, association of TIM-1 polymorphisms to Th2-related airway hyperreactivity has been suggested in mice. The TIM molecules have not been investigated in human Th1- or Th2-mediated diseases. Using real-time (TaqMan) RT-PCR, we show that human Th1 lines expressed higher TIM-3 mRNA levels, while Th2 lines demonstrated a higher expression of TIM-1. Analysis of cerebrospinal fluid mononuclear cells obtained from patients with multiple sclerosis revealed significantly higher mRNA expression of TIM-1 compared with controls. Moreover, higher TIM-1 expression was associated with clinical remissions and low expression of IFN-gamma mRNA in cerebrospinal fluid mononuclear cells. In contrast, expression of TIM-3 correlated well with high expression of IFN-gamma and TNF-alpha. These data imply the differential expression of human TIM molecules by Th1 and Th2 cells and may suggest their differential involvement in different phases of a human autoimmune disease.  相似文献

Carbon cycling in ecosystems, and especially in forests, is intensively studied to predict the effects of global climate change, and the role which forests may play in 'changing climate change'. One of the questions is whether the carbon balance of forests will be affected by increasing atmospheric CO2 concentrations. Regarding this question, effects of elevated [CO2] on woody-tissue respiration have frequently been neglected. Stem respiration of three Populus species ( P. alba L. (Clone 2AS-11), P. nigra L. (Clone Jean Pourtet), and P.  ×  euramericana (Clone I-214)) was measured in a managed, high-density forest plantation exposed to free-air CO2 enrichment (POPFACE). During the period of measurements, in May of the third year, stem respiration rates were not affected by the FACE treatment. Moreover, FACE did not influence the relationships between respiration rate and both stem temperature and relative growth rate. The results were supported by the reported absence of a FACE-effect on growth and stem wood density.  相似文献

For the moment the agents that are used against Botrytis cinerea, in glasshouses were tomatoes are cultivated, are from chemical origin. For reducing the use of chemical agents in the future it is important to search for effective biological control agents against the fight of Botrytis cinerae. The following biological products Vital pasta, Vital gel and Elot-Vis were examined in there possibility to control Botrytis cinerea. Elot Vis was tested out in experiments that were carried out in climate chambers were leafs of 3 week old tomato plants were artificially infected with Botrytis cinerea spores. Also the biological products of Vital were first investigated in experiments that were carried out in climate chambers. In stead off leafs of tomato plants it were stem wounds of tomato plants who were treated with the pasta or the gel that was spread over the wounded surface after this has been inoculated with a suspension of conidia of Botrytis cinerae. The results of these first tests that were executed in the climate chambers were the circumstances for Botrytis cinerea were ideal seemed promising. In the next step these products were tested out on large scale in glasshouses. For each plant 5 wounds were created by removing the leafs, these wounds were or first treated with the Biological product and thereafter artificially infected with Botrytis cinerea spores to check out if these products can be used as a preventive agent or the wounds were first inoculated with a suspension of Botrytis cinerea spores and thereafter treated with the product. For the product Elot-Vis a few plants were totally sprayed with an Elot-Vis suspension before leafs were removed and the wounds were inoculated with conidia of Botrytis cinerea to check out if this product was able to activated the induced systemic resistance pathway. The experiments that were executed in glasshouses showed different percentages of succeeded Botrytis cinerea infections. This is probable due to the different weather conditions during both days that the experiments were executed. For the wounds that were treated with pasta it was difficult to distinguish wounds were Botrytis cinerea succeeded to infect the plant, because these wounds frequently didn't show any sign of infection on the surface but when the wounds with the pasta were cut open it was possible to see Botrytis cinerea infections inside the stem.  相似文献

The present paper describes a geometrically and physically nonlinear continuum model to study the mechanical behaviour of passive and active skeletal muscle. The contraction is described with a Huxley type model. A Distributed Moments approach is used to convert the Huxley partial differential equation in a set of ordinary differential equations. An isoparametric brick element is developed to solve the field equations numerically. Special arrangements are made to deal with the combination of highly nonlinear effects and the nearly incompressible behaviour of the muscle. For this a Natural Penalty Method (NPM) and an Enhanced Stiffness Method (ESM) are tested. Finally an example of an analysis of a contracting tibialis anterior muscle of a rat is given. The DM-method proved to be an efficient tool in the numerical solution process. The ESM showed the best performance in describing the incompressible behaviour.  相似文献

Video-rate confocal microscopy of Indo-1-loaded human skeletal myotubes was used to assess the relationship between the changes in sarcoplasmic ([Ca(2+)](S)) and nuclear ([Ca(2+)](N)) Ca(2+) concentration during low- and high-frequency electrostimulation. A single stimulus of 10 ms duration transiently increased [Ca(2+)] in both compartments with the same time of onset. Rate and amplitude of the [Ca(2+)] rise were significantly lower in the nucleus (4.0- and 2.5-fold, respectively). Similarly, [Ca(2+)](N) decayed more slowly than [Ca(2+)](S) (mono-exponential time constants of 6.1 and 2.5 s, respectively). After return of [Ca(2+)] to the prestimulatory level, a train of 10 stimuli was applied at a frequency of 1 Hz. The amplitude of the first [Ca(2+)](S) transient was 25% lower than that of the preceding single transient. Thereafter, [Ca(2+)](S) increased stepwise to a maximum that equalled that of the single transient. Similarly, the amplitude of the first [Ca(2+)](N) transient was 20% lower than that of the preceding single transient. In contrast to [Ca(2+)](S), [Ca(2+)](N) then increased to a maximum that was 2.3-fold higher than that of the single transient and equalled that of [Ca(2+)](S). In the nucleus, and to a lesser extent in the sarcoplasm, [Ca(2+)] decreased faster at the end of the stimulus train than after the preceding single stimulus (time constants of 3.3 and 2.1 s, respectively). To gain insight into the molecular principles underlying the shaping of the nuclear Ca(2+) signal, a 3-D mathematical model was constructed. Intriguingly, quantitative modelling required the inclusion of a satiable nuclear Ca(2+) buffer. Alterations in the concentration of this putative buffer had dramatic effects on the kinetics of the nuclear Ca(2+) signal. This finding unveils a possible mechanism by which the skeletal muscle can adapt to changes in physiological demand.  相似文献

An analytical model is developed that describes oxygen transport and oxygen consumption for small biological structures without a circulatory system. Oxygen inside the organism is transported by diffusion alone. Oxygen transfer towards the organism is retarded by a thin static fluid film at the surface of the organism. The thickness of this film models the outward water conditions, which may range from completely stagnant water conditions to so-called well-stirred water conditions. Oxygen consumption is concentration-independent above a specified threshold concentration (regulator behaviour) and is proportional to the oxygen concentration below this threshold (conformer behaviour). The model takes into account shape and size of the organism and predicts the transition from (pure) regulator behaviour to (pure) conformer behaviour, as well as the mean oxygen consumption rate. Thereby the model facilitates a proper analysis of the physical constraints set on shape and size of organisms without an active internal oxygen transport mechanism. This analysis is carried out in some detail for six characteristic shapes (infinite sheet, cylinder and beam; finite cylinder, sphere and block). In a well-stirred external medium, a flattened shape appears to be the most favourable for oxygen supply, while a compact shape (cube) is more favourable if the external medium is nearly stagnant. The theoretical framework is applied to oxygen consumption data of eight teleost embryos. This reveals relative insensitivity to external flow conditions in some species (e.g., winter flounder, herring), while others appear to rely on external stirring for a proper oxygen supply (e.g., largemouth bass). Interestingly, largemouth bass is the only species in our analysis that exhibits ‘fin-fanning’.  相似文献

Abstract: This article analyzes the possibilities for reducing carbon dioxide (CO2) emissions in the life cycle of Japanese petrochemicals, focusing primarily on the nonenergy use of fossil fuels. For this purpose a linear programming model called CHEAP (CHemical industry Environmental strategy Analysis Program) has been developed. The results show a moderate autonomous growth of emissions by 5% in the period 2000 to 2020, if it is assumed that no new technology is introduced and demand (measured in physical units) increases 1% per year, on average. However, if it is assumed that ongoing technology development succeeds, emissions in 2020 may decrease by 5% from 2000 levels (a decrease of 10% compared to the case that assumes no new technology). This is a significant contribution to emission reduction. According to this model, a further emission reduction by 10% in 2020 is possible but costly as it requires emission reduction incentives of up to 10,000 yen per ton CO2 (approximately 100 US/ton). The use of biomass feed-stocks, waste recycling, energy recovery from waste and gas-based co-generation are the main strategies for achieving this emission reduction.  相似文献

Materials use is an important factor influencing carbon dioxide (CO2) emissions because significant amounts of carbon dioxide are released during the production of materials from natural resources, and because products and wastes can function as important sinks for CO2. This article analyzes the impact of Western European materials use on CO2 emissions. The material flows for steel, cement, petrochemicals, and wood products are analyzed in more detail. The analysis shows that particular characteristics of the materials system must be considered in the development of emission reduction strategies. It is important to select a relatively closed system for policymaking, as in Western Europe, in order to prevent unwanted transboundary effects. The materials stored in the form of products, and the net exports of materials, products, and waste limit the potential of a recycling strategy. Carbon storage in products and waste disposal sites is significant both for synthetic and natural organic materials, but is not accounted for in natural organic materials in current emissions statistics. Accordingly the emissions accounting practices should be modified to reflect the storage of such materials.  相似文献

A cDNA clone for a hormonally regulated beta-glucanase from Nicotiana plumbaginifolia has been isolated by using an oligodeoxynucleotide probe, synthesized to match the previously determined N-terminal amino acid sequence. The cDNA has the complete sequence of the mature protein and contains at least part of a hydrophobic signal peptide. At the amino acid level, the beta-glucanase of N. plumbaginifolia is 73% homologous to a beta(1,3)-glucanase from tobacco and 52% homologous to a beta(1,3;1,4)-glucanase from barley. Southern-blot analysis clearly demonstrated that N. plumbaginifolia contains at least two related genes encoding beta-glucanase. The extent of the complete signal peptide of the cloned beta-glucanase was determined by sequencing part of the corresponding gene. Northern analysis showed that the expression of the beta-glucanase gene is influenced by auxins and cytokinins.  相似文献
