1. A soluble D-alanine carboxypeptidase from Escherichia coli strain B was purified on a p-aminobenzylpenicillin-Sepharose column. This one-step chromatography followed by an (NH4)2SO4 precipitation yielded an enzyme purified 1200-fold and some of its properties are reported. 2. The pure D-alanine carboxypeptidase was devoid of D-alanine carboxypeptidase II activity and migrated as a single protein band on analytical disc gel electrophoresis. 3. Triton X-100 in the purification procedure is an absolute requirement for obtaining a stable enzyme. 4. The enzymic activity of D-alanine carboxypeptidase was greatly affected in solution of high salt concentrations and varied somewhat with the nature of the cation tested.  相似文献

Among a number of mutants unable to utilize non-fermentable carbon substrates, scoring for membrane ATPase and for ATP-driven transhydrogenase activity permitted to distinguish two phenotypes: (A) mutants lacking ATPase and ATPdriven transhydrogenase; (B) one mutant with an ATPase which behaved according to several criteria as released into solution instead of being membrane bound, a.o it exhibited no ATP-driven transhydrogenase activity. All A and B mutants exhibited a common nutritional pattern.The ATPase-deficient group, when scored for ATPase-binding sites on its membrane particles revealed three different subgroups: (1) mutants having free ATPase-binding sites, (2) mutants with ATPase-binding sites made available by the procedure which releases ATPase from wild-type membrane, and (3) mutants with no detectable ATPase-binding sites.Membranes of the mutant B with unbound ATPase also exhibited a deficiency in ATPase-binding sites, but its soluble ATPase was also found unable to bind to ATPase-binding sites of wild type membranes.The double alteration, namely abnormal or inactive ATPase and absence of ATPase-binding sites on the membrane is compatible with a single mutational defect.  相似文献

Procedures are described for the isolation of lipoproteins from human serum by precipitation with polyanions and divalent cations. A mixture of low and very low density lipoproteins can be prepared without ultracentrifugation by precipitation with heparin and either MnCl(2) alone or MgCl(2) plus sucrose. In both cases the precipitation is reversible, selective, and complete. The highly concentrated isolated lipoproteins are free of other plasma proteins as judged by immunological and electrophoretic methods. The low density and very low density lipoproteins can then be separated from each other by ultracentrifugation. The advantage of the method is that large amounts of lipoproteins can be prepared with only a single preparative ultracentrifugation. Polyanions other than heparin may also be used; when the precipitation of the low and very low density lipoproteins is achieved with dextran sulfate and MnCl(2), or sodium phosphotungstate and MgCl(2), the high density lipoproteins can subsequently be precipitated by increasing the concentrations of the reagents. These lipoproteins, containing small amounts of protein contaminants, are further purified by ultracentrifugation at d 1.22. With a single preparative ultracentrifugation, immunologically pure high density lipoproteins can be isolated from large volumes of serum.  相似文献

Antibodies to the F metabolite 9α, 11α-dihydro-15-keto-prostanoic acid (I), produced in the rabbit, do not cross react with any of the primary PGs. There is a 50% cross reaction with the metabolite 9α, 11α-dihydroxy-15-keto-prost-5-enoic acid (III), and a 23% cross reaction with 9α,11α,15-trihydroxy prostanoic acid (F0α). No cross reactivity resulted with this antiserum when tested against 9α,11α,15-trihydroxy-5-enoic acid (VII) or with 9α,11α-dihydroxy-15-ketoprost-5,13-dienoic acid (VIII). Utilizing this antibody in a radioimmunoassay, some preliminary data are presented on levels of these F metabolites (I and III) for human adult male samples of plasma, urine and seminal plasma.  相似文献

Addition of alkali metal ions (Li+, Na+, K+, Rb+) to the solution of isolated component A of the viper neurotoxic complex was found to pronouncedly change the protein fluorescence properties. The maximal effect takes place at the addition of potassium salts which induce the fluorescence spectral shift toward shorter wavelengths by 13.5 nm. This effect is dependent on the protein concentration and is evidence that alkali ions induce the oligomerization of component A. The oligomerization equilibrium constants are strongly dependent on both the concentration and the kind of alkali ions inducing it. The presence in the solution of Tris strongly inhibits the oligomerization. The process is temperature dependent and the oligomerization is maximal at 35–50 °C. An attempt was made to check the assumption that component A can increase the K+ permeability of phospholipid membranes (liposomes). The results confirmed qualitatively this assumption. Possible functional significance of these findings is discussed.  相似文献

The response of murine megakaryocytopoiesis was studied under conditions of varying platelet demand. Twenty-four hours after mice were given a single injection of rabbit anti-platelet serum, megakaryocyte number and volume were increased, becoming maximal at 65 and 40 hr, respectively. Total body megakaryocytic colony-forming unit (CFU-M) numbers did not change until 90 hr, when a 35% increase in the experimental group was noted. The percentage of CFU-M in DNA synthesis in the experimental group was 38 ± 2% at 24 hr, 49 ± 1% at 40 hr, and returned to normal (11 ± 3%) at 90 hr. When mice were made thrombocytotic by platelet transfusions, both megakaryocyte number and volume were decreased compared to controls, while no difference was noted in the number and percentage of CFU-M in DNA synthesis. Finally, experiments were performed to examine the effect of platelet transfusions on regenerating marrow. Experimental mice were given platelet transfusions while control animals received platelet buffer solution. At sacrifice the number and volume of megakaryocytes in the experimental group (platelet count 2.568 × 106/μl) were less than controls (platelet count 0.363 × 106/μl), while the number and percentage of CFU-M in DNA synthesis were similar in both groups. These results demonstrate that CFU-M are not immediately responsive to acute changes in platelet demand. The data suggest that megakaryo-cytopoiesis is structured on at least two levels which are independently regulated.  相似文献

Rare cases of extrapulmonary Pneumocystis carinii (EPPC) have been seen in patients with acquired immunodeficiency syndrome (AIDS). We report seven such diagnoses of nonpulmonary P carinii (PC) from four AIDS patients between 1986 and 1989. The specimens included fine needle aspirate of liver, spleen, periarticular tissue and pleura as well as ankle fluid, pleural fluid and ascites. In some, but not all, cases the patients had concurrent or previous episodes of PC pneumonia. In all cases the typical granular, eosinophilic aggregates of PC cysts were noted on routine Papanicolaou staining, leading to the definitive detection of PC cysts with Grocott silver stain. In most cases, evidence for granulomalike and neovascularized tissue reaction was present in cytologic material. One specimen demonstrated concurrent acid fast bacilli. In the setting of AIDS, cytology of effusions and masses should include an evaluation for EPPC.  相似文献

We used indirect immunofluorescence to examine the factors determining the intranuclear location of herpes simplex virus (HSV) DNA polymerase (Pol) in infected cells. In the absence of viral DNA replication, HSV Pol colocalized with the HSV DNA-binding protein ICP8 in nuclear framework-associated structures called prereplicative sites. In the presence of viral DNA replication, HSV Pol colocalized with ICP8 in globular intranuclear structures called replication compartments. In cells infected with mutant viruses encoding defective ICP8 molecules, Pol localized within the cell nucleus but showed a general diffuse intranuclear distribution. In uninfected cells transfected with a plasmid expressing Pol, Pol similarly showed a diffuse intranuclear distribution. Therefore, Pol can localize to the cell nucleus without other viral proteins, but functional ICP8 is required for Pol to localize to prereplicative sites. In cells infected with mutant viruses encoding defective Pol molecules, ICP8 localized to prereplicative sites. Thus, Pol or the portions of Pol not expressed by the mutant viruses are not essential for the formation of prereplicative sites or the localization of ICP8 to these structures. These results demonstrate that a specific nuclear protein can influence the intranuclear location of another nuclear protein.  相似文献
