Integration between ecology and biogeography provides insights into how niche specialization affects the geographical distribution of species. Given that rivers are not effective barriers to dispersal in three parapatric species of squirrel monkeys (Saimiri vanzolinii, S. cassiquiarensis and S. macrodon) inhabiting floodplain forests of Central Amazonia, we tested whether forest structure and tree diversity may explain species differences in niche specialization and spatial segregation. We sampled 6617 trees of 326 species in three habitats (high várzea, low várzea and chavascal) used by three Saimiri species, and estimated tree species richness in each of them. For each tree, we measured variables known to influence habitat use in primates, such as crown area and presence of lianas, epiphytes and hemi-epiphytes. We used ANOVA to compare these variables and performed multivariate analyses (NMDS, ANOSIM and SIMPER) to evaluate dissimilarities in forest structure among each habitat inhabited by the three Saimiri species. We identified differences in the tree species richness, crown area and presence of lianas, epiphytes and hemi-epiphytes between the three habitats for all Saimiri species. NMDS demonstrated that areas of high and low várzeas occupied by S. vanzolinii were clearly separated from the other species. We also found that different plant species contributed to dissimilarity among Saimiri ranges. Our findings support the hypothesis that tree community structure may promote niche specialization and spatial segregation among primates. We discuss how these patterns could have been favored by historical changes in forest flood patterns, the evolutionary history of Saimiri spp., and past competition.  相似文献

Mycosporine‐like amino acids (MAA) are ultraviolet screen substances synthesized by marine algae. The physiological function of these substances is related to cellular protection against UV radiation and as a protective mechanism against oxidative stress. These substances can be found mainly in the ocean, among red seaweeds. Its concentration in organisms has been related to ultraviolet radiation and availability of inorganic nitrogen in the environment. We start our study of MAA content in different species to understand if environmental conditions influence the concentration of MAAs in red seaweeds. The Brazilian coast presents abiotic factors that interact to create different physical‐chemical features in the environment. We collected 441 samples from 39 species of red seaweed easily found in the intertidal zone, in low tide, during the summer of 2015. The sampling encompassed a latitudinal gradient (3° S to 28°5′ S) at 23 points along the coast. We quantified and identified the content of MAAs in species through the method of high performance liquid chromatography. We detected for the first time the occurrence of MAAs in certain species of red algae that have not been reported to contain MAAs before. We confirmed that some environmental factors influenced the content of MAAs. Enhanced MAA contents, for example, were found in environments with a basic pH, a high ultraviolet index, and high concentrations of phosphate and nitrate. Salinity, dissolved oxygen and variations of sea surface temperature also influenced, in a secondary way, MAA content in algae in their natural environments.  相似文献

Background

The fluorescent dye 10-N-nonyl acridine orange (NAO) is widely used as a mitochondrial marker. NAO was reported to have cytotoxic effects in cultured eukaryotic cells when incubated at high concentrations. Although the biochemical response of NAO-induced toxicity has been well identified, the underlying molecular mechanism has not yet been explored in detail.

Methods

We use optical techniques, including fluorescence confocal microscopy and lifetime imaging microscopy (FLIM) both in model membranes built up as giant unilamellar vesicles (GUVs) and cultured cells. These experiments are complemented with computational studies to unravel the molecular mechanism that makes NAO cytotoxic.

Results

We have obtained direct evidence that NAO promotes strong membrane adhesion of negatively charged vesicles. The attractive forces are derived from van der Waals interactions between anti-parallel H-dimers of NAO molecules from opposing bilayers. Semi-empirical calculations have confirmed the supramolecular scenario by which anti-parallel NAO molecules form a zipper of bonds at the contact region. The membrane remodeling effect of NAO, as well as the formation of H-dimers, was also confirmed in cultured fibroblasts, as shown by the ultrastructure alteration of the mitochondrial cristae.

Conclusions

We conclude that membrane adhesion induced by NAO stacking accounts for the supramolecular basis of its cytotoxicity.

General significance

Mitochondria are a potential target for cancer and gene therapies. The alteration of the mitochondrial structure by membrane remodeling agents able to form supramolecular assemblies via adhesion properties could be envisaged as a new therapeutic strategy.  相似文献

Infectious diseases are one of the main causes of human morbidity and mortality. In the last few decades, pathogenic microorganisms' resistance to conventional drugs has been increasing, and it is now pinpointed as a major worldwide health concern. The need to search for new therapeutic options, as well as improved treatment outcomes, has therefore increased significantly, with biologically active peptides representing a new alternative. A substantial research effort is being dedicated towards their development, especially due to improved biocompatibility and target selectivity. However, the inherent limitations of peptide drugs are restricting their application. In this review, we summarize the current status of peptide drug development, focusing on antiviral and antimicrobial peptide activities, highlighting the design improvements needed, and those already being used, to overcome the drawbacks of the therapeutic application of biologically active peptides.  相似文献

Sphex ingens is one of 30 species in the family Sphecidae that occur in the state of Rio de Janeiro. However, details of the behavior and sexual selection of natural populations of this wasp species have only recently been unveiled. In addition, the knowledge of its ecology is still poor. This is the first study on the feeding behavior interactions between S. ingens and prey captured to feed its larvae. Paralyzed prey were collected manually at the sites of wasp nests on Aventureiro Beach, Ilha Grande, Brazil during the provisioning activity of marked female S. ingens. All prey were preserved, their sex and sexual maturity were determined, and they were identified to the lowest possible taxonomic level. The body mass and size of the prey and female wasps were measured. Sphex ingens females captured only Pleminia vicina and Meroncidius sp. The body masses of wasps and katydids were positively correlated. The body mass of captured katydids was significantly dependent on the wasp’s wing length. Most of the prey were adult females, but the differences did not confirm possible preferences, as those values can be related to differences in the distribution and fluctuation in the population density of prey species and to the individual foraging strategies of female wasps. However, the predisposition to predatory specialization exhibited by S. ingens populations in Ilha Grande and elsewhere suggests that this interaction can be an important source of mortality for populations of pseudophylline katydid species.  相似文献

In the state of São Paulo in southeastern Brazil, a dangerous decree allows the culture of hybrids and non-native extralimital fish species in the Paraná and East Atlantic River basins. We recorded the illegal importation and sale of the non-native striped catfish Pangasianodon hypophthalmus in northeastern, central-western, southeastern, and southern areas of the country. This species is already consumed across Brazil and is cultivated for ornamental purposes. Besides escapes from fish farms and ‘fish and pays’, P. hypophthalmus can reach natural environments through aquarium dumping and ornamental ponds, motivated by its large size that reach after being sold at small size. The species has been introduced in many countries, and if established in Brazil, could put native Brazilian biodiversity and ecosystems at risk. Although importing specimens of P. hypophthalmus is prohibited by law, Decree 62.243/2016 in the state of São Paulo creates a new opportunity for invasion. In fact, once a novel fish species is moved to a new continent, it is irrational to supposed that it will stay static just in the area for which it was intended. In general, the fish disperse in the novel area on their own, by escapes from captivity or deliberate and illicit releases by persons or associations. The unwise attitude of Brazilian decision-makers and lay people in their attempt to develop aquaculture with non-native species goes against the objectives of responsible aquaculture based in native species.  相似文献

New treatment modalities for glioblastoma multiforme (GBM) are urgently needed. Proton therapy is considered one of the most effective forms of radiation therapy for GBM. DNA alkylating agents such as temozolomide (TMZ) are known to increase the radiosensitivity of GBM to photon radiation. TMZ is a fairly impotent agent, while duocarmycin SA (DSA) is an extremely potent cytotoxic agent capable of inducing a sequence-selective alkylation of duplex DNA. Here, the effects of sub-nM concentrations of DSA on the radiosensitivity of a human GBM cell line (U-138) to proton irradiation were examined. Radiation sensitivity was determined by viability, apoptosis, necrosis and clonogenic assays. DSA concentrations as low as 0.001?nM significantly sensitized U-138 cells to proton irradiation. DSA demonstrates synergistic cytotoxicity against GBM cells treated with proton radiation in vitro, which may represent a novel therapeutic alternative for the treatment of GBM.  相似文献

Coastal forested wetlands provide important ecosystem services such as carbon sequestration, nutrient retention, and flood protection, but they are also important sources of greenhouse gas emissions. Human appropriation of surface water and extensive ditching and draining of coastal plain landscapes are interacting with rising sea levels to increase the frequency and magnitude of saltwater incursion into formerly freshwater coastal wetlands. Both hydrologic change and saltwater incursion are expected to alter carbon and nutrient cycling in coastal forested wetlands. We performed a full factorial experiment in which we exposed intact soil cores from a coastal forested wetland to experimental marine salt treatments and two hydrologic treatments. We measured the resulting treatment effects on the emissions of carbon dioxide (CO2), methane (CH4), and nitrous oxide (N2O) over 112 days. Salinity effects were compared across four treatments to isolate the effects of increases in ionic strength from the impact of adding a terminal electron acceptor (SO42?). We compared control treatments (DI addition), to artificial saltwater (ASW, target salinity of 5 parts per thousand) and to two treatments that added sulfate alone (SO42?, at the concentration found in 5 ppt saltwater) and saltwater with the sulfate removed (ASW-SO42?, with the 5 ppt target salinity maintained by adding additional NaCl). We found that all salt treatments suppressed CO2 production, in both drought and flooded treatments. Contrary to our expectations, CH4 fluxes from our flooded cores increased between 300 and 1200% relative to controls in the ASW and ASW-SO42? treatments respectively. In the drought treatments, we saw virtually no CH4 release from any core, while artificial seawater with sulfate increased N2O fluxes by 160% above DI control. In contrast, salt and sulfate decreased N2O fluxes by 72% in our flooded treatments. Our results indicate that salinization of forested wetlands of the coastal plain may have important climate feedbacks resulting from enhanced greenhouse gas emissions and that the magnitude and direction of these emissions are contingent upon wetland hydrology.  相似文献

The Cerrado Biome is the second largest in Brazil covering roughly 2 million km2, with varying features throughout its area. The Biome is endangered but it is also source of animal species for rural, green urban and degraded rainforest areas. Ticks are among Cerrado species that establish at anthropogenic sites and although information about them is steadily increasing, several features are unknown. We herein report tick species, abundance and some ecological relationships within natural areas of the Cerrado at higher altitudes (800–1500 m) within and around Serra da Canastra National Park, in Minas Gerais State Brazil. In total of 1196 ticks were collected in the environment along 10 campaigns held in 3 years (2007–2009). Amblyomma sculptum was the most numerous species followed by Amblyomma dubitatum and Amblyomma brasiliense. Distribution of these species was very uneven and an established population of A. brasiliense in the Cerrado is reported for the first time. Other tick species (Amblyomma ovale, Amblyomma nodosum, Amblyomma parvum, Ixodes schulzei and Haemaphysalis leporispalustris) were found in lesser numbers. Domestic animals displayed tick infestations of both rural and urban origin as well as from natural areas (Rhipicephalus sanguineus sensu lato, Rhipicephalus microplus, Dermacentor nitens, A. sculptum, A. ovale, Amblyomma tigrinum, Argas miniatus). Amblyomma sculptum had the widest domestic host spectrum among all tick species. DNA of only one Rickettsia species, R. bellii, was found in an A. dubitatum tick. Several biological and ecological features of ticks of the studied areas are discussed.  相似文献

Phosphorus and nitrogen fertilizers represent a source of cadmium (Cd) which may be leached into aquatic systems. Macrophytes accumulate contaminants, and Egeria densa has been shown to grow in aquatic environments polluted with trace elements. In this study, Cd accumulation by E. densa exposed to two Cd treatments (3 and 5 mg L?1) was evaluated under increasing nutrient levels (NP as N–NO3 ?, N–NH4 +, and P–PO4 3?, in concentrations 5-, 10- and 100-fold higher (NP5, NP10 and NP100) than in the sampling site) to simulate different levels of eutrophication. Bioaccumulation factors and Cd recovery were calculated and effects on plants were evaluated based on chloroplastidic pigment concentrations (chlorophylls a and b, and carotenoids). We conclude that Cd accumulation by Egeria densa is positively influenced by increasing availability of N and P at the level of around NP10 and probably at a broader concentration range not defined in this study. A further increase in N and P, however, does not generate a significant increase in Cd accumulation. Chloroplastidic pigment concentrations were not linearly correlated with Cd accumulation and the NP10 experiment produced less damage to macrophyte when compared to NP5 and NP100 experiments. Under controlled conditions, it was possible to satisfactorily model Cd bioaccumulation over time, in order to provide essential data for E. densa use in phytoremediation processes. The Cd residence in the macrophyte tissue is increased in eutrophic environments, which puts at risk the whole food chain of the aquatic ecosystem, mainly the primary consumers.  相似文献
