The TolC-like protein HgdD of the filamentous, heterocyst-forming cyanobacterium Anabaena sp. PCC 7120 is part of multiple three-component “AB-D” systems spanning the inner and outer membranes and is involved in secretion of various compounds, including lipids, metabolites, antibiotics, and proteins. Several components of HgdD-dependent tripartite transport systems have been identified, but the diversity of inner membrane energizing systems is still unknown. Here we identified six putative resistance-nodulation-cell division (RND) type factors. Four of them are expressed during late exponential and stationary growth phase under normal growth conditions, whereas the other two are induced upon incubation with erythromycin or ethidium bromide. The constitutively expressed RND component Alr4267 has an atypical predicted topology, and a mutant strain (I-alr4267) shows a reduction in the content of monogalactosyldiacylglycerol as well as an altered filament shape. An insertion mutant of the ethidium bromide-induced all7631 did not show any significant phenotypic alteration under the conditions tested. Mutants of the constitutively expressed all3143 and alr1656 exhibited a Fox phenotype. The phenotype of the insertion mutant I-all3143 parallels that of the I-hgdD mutant with respect to antibiotic sensitivity, lipid profile, and ethidium efflux. In addition, expression of the RND genes all3143 and all3144 partially complements the capability of Escherichia coli ΔacrAB to transport ethidium. We postulate that the RND transporter All3143 and the predicted membrane fusion protein All3144, as homologs of E. coli AcrB and AcrA, respectively, are major players for antibiotic resistance in Anabaena sp. Post-fire coarse woody debris can represent a valuable nutrient reservoir for a regenerating ecosystem, helping to prevent soil fertility losses after a wildfire. However, there is scarce information on its effect on soil nutrient cycling and availability. We established three study sites along an altitudinal gradient in a burnt pine forest (SE Spain). At each site we determined: (1) decomposition rates and nutrient dynamics in charred logs left on the ground, 2 and 4 years after the fire, and (2) available nutrients in the soil and in the microbial fraction below charred logs and in bare soil areas. Despite the relatively slow decay rates in this Mediterranean climate (ca. 10 % of dry weight lost after 4 years), N and P were progressively released by logs, accounting for ca. 40 and 65 % of the initial content respectively after 4 years. This implies that the total aboveground biomass of the burnt forest released around 20 kg ha?1 of N and 2 kg ha?1 of P during this period. The presence of post fire coarse woody debris consistently increased soil organic matter by around 18 %, total C and N by 42 and 26 %, respectively, dissolved organic C and N by 47 %, available inorganic P by 68 %, and microbial biomass and nitrogen by some 36 and 48 %, respectively. By contrast, soil bulk density decreased by ca. 18 % under logs compared to bare areas. Thus, the fire-killed wood was useful in the recovery of soil fertility and nutrient availability. Leaving the post-fire woody debris on site can enhance the biogeochemical sustainability, microbiological processes and soil ecological functioning. The detrimental effect of post-fire salvage logging on soil fertility should be therefore considered when making management decisions.  相似文献

Detailed analysis of the morphology of Florisphaera profunda from plankton samples collected at three sites in the Atlantic and Pacific Oceans reveals wide variation in this deep ocean-dwelling coccolithophore. In addition to the two varieties described previously, we found a third distinctive form, Florisphaera profunda var. rhinocera var. nov. All three varieties occur at each of the sampling sites. The analysis of monthly samples from different levels in the lower photic zone (LPZ) (100–200?m) at the Hawaii Ocean Time series station suggests that the varieties have similar distributions, which are correlated to primary productivity and the availability of light. The analysis of coccolith and coccosphere size in F. profunda reveals the existence of several size modes in Florisphaera profunda var. profunda and F. profunda var. elongata. The biological significance of these modes, or morphotypes is not known. However, their co-occurrence in single samples from different oceanic areas suggests that they are not ecophenotypes. In the light of recent molecular genetic analyses of intraspecific groups within commonly occurring coccolithophores, the varieties and size morphotypes of F. profunda are of significant interest for the study of marine phytoplankton biodiversity. Coccolithophores inhabiting the LPZ may be adapted to the low light, high nutrient conditions of this layer and hold great potential as a means to reconstruct past oceanographic conditions such as the position of the nutricline. However, coccolithophore biodiversity in the LPZ is poorly documented and the number of species may be much higher than previously thought.  相似文献

Nine O-alkyl and O-prenyl derivatives were synthesized from commercial 2,4-dihydroxybenzophenone, 4,e4,4′-dihydroxybenzophenone and were evaluated for their leishmanicidal activity against promastigote forms of Leishmania amazonensis, as well their toxicity in murine macrophages. All derivatives exhibited better biological activity than their hydroxylated benzophenones precursors, and new compound LFQM-123 (3c) was 250-fold more active than its precursor 4,4′-dihydroxybenzophenone (3). Moreover, some of the results were comparable to the standard drug Amphotericin B, suggesting that the increase in lipophilicity could facilitate protozoa membrane permeation. In this study we confirmed that benzophenone derivatives exhibit leishmanicidal properties, with relatively low toxicity, and thus could be exploited as promise prototypes for the design and development of new drug for the treatment of leishmaniasis.  相似文献

To understand the impact of the northwestern Azores Current Front (NW-AzC/AzF) system on HCO3-and N2-fixation activities and unicellular diazotrophic cyanobacteria (UCYN) distribution, we combined geochemical and biological approaches from the oligotrophic surface to upper mesopelagic waters. N2-fixation was observed to sustain 45–85% of the HCO3-fixation in the picoplanktonic fraction performing 47% of the total C-fixation at the deep chlorophyll maximum north and south of the AzF. N2-fixation rates as high as 10.9 μmol N m-3 d-1 and surface nitrate δ15N as low as 2.7‰ were found in the warm (18–24°C), most saline (36.5–37.0) and least productive waters south of the AzF, where UCYN were the least abundant. However, picoplanktonic UCYN abundances up to 55 cells mL-1 were found at 45–200m depths in the coolest nutrient-rich waters north of the AzF. In this area, N2-fixation rates up to 4.5 μmol N m-3 d-1 were detected, associated with depth-integrated H13CO3-fixation rates at least 50% higher than observed south of the AzF. The numerous eddies generated at the NW-AzC/AzF seem to enhance exchanges of plankton between water masses, as well as vertical and horizontal diapycnal diffusion of nutrients, whose increase probably enhances the growth of diazotrophs and the productivity of C-fixers.  相似文献

Cardiovascular diseases and oxidative stress are related to polycystic ovary syndrome (PCOS) and insulin resistance (IR). We have evaluated the relationship between myeloperoxidase (MPO) and leukocyte activation in PCOS patients according to homeostatic model assessment of IR (HOMA-IR), and have explored a possible correlation between these factors and endocrine and inflammatory parameters. This was a prospective controlled study conducted in an academic medical center. The study population consisted of 101 PCOS subjects and 105 control subjects. We divided PCOS subjects into PCOS non-IR (HOMA-IR<2.5) and PCOS IR (HOMA-IR>2.5). Metabolic and anthropometric parameters, total and mitochondrial reactive oxygen species (ROS) production, MPO levels, interactions between human umbilical vein endothelial cells and leukocytes, adhesion molecules (E-selectin, ICAM-1 and VCAM-1) and proinflammatory cytokines (IL-6 and TNF-α) were evaluated. Oxidative stress was observed in PCOS patients, in whom there was an increase in total and mitochondrial ROS production and MPO levels. Enhanced rolling flux and adhesion, and a decrease in polymorphonuclear cell rolling velocity were also detected in PCOS subjects. Increases in IL-6 and TNF-α and adhesion molecules (E-selectin, ICAM-1 and VCAM-1) were also observed, particularly in the PCOS IR group, providing evidence that inflammation and oxidative stress are related in PCOS patients. HOMA-IR was positively correlated with hsCRP (p<0.001, r = 0.304), ROS production (p<0.01, r = 0.593), leukocyte rolling flux (p<0.05, r = 0.446), E-selectin (p<0.01, r = 0.436) and IL-6 (p<0.001, r = 0.443). The results show an increase in the rate of ROS and MPO levels in PCOS patients in general, and particularly in those with IR. Inflammation in PCOS induces leukocyte-endothelium interactions and a simultaneous increase in IL-6, TNF-α, E-selectin, ICAM-1 and VCAM-1. These conditions are aggravated by the presence of IR.  相似文献
