Direct measurement of the nonapeptide vasopressin has been limited by analyte instability ex vivo and in vivo rapid degradation, low serum concentrations requiring a sensitive assay and inherent secretory pulsatility. Copeptin is a 39 amino acid glycopeptide cleavage product of vasopressin synthesis with high stability, providing a marker of vasopressin secretion. Copeptin measurement has applications in diagnosis of diabetes insipidus and other diseases with altered vasopressin secretion. This review summarises our current understanding of serum copeptin measurement in diabetes insipidus and possible future applications of copeptin assays. As vasopressin is a stress hormone, there is emerging evidence on the use of copeptin for diagnosis and prognostication of disorders such as syndrome of inappropriate anti-diuretic hormone secretion, diabetes mellitus, critical illness, stroke, cardiovascular disease, respiratory disease, renal disease and thermal stress. Copeptin concentration measurement is likely to improve the diagnostic reliability of diabetes insipidus and, as a marker of stress, may have diagnostic or prognostic utility in specific clinical circumstances. Further studies are needed to determine if goal-directed therapy using plasma copeptin concentrations may improve patient outcomes.  相似文献

The problem of exchange of substances between circulating blood and tissue is discussed in terms of a model with continuously distributed tissue characteristics. Consideration is given to the distribution of perfusion rates, the distribution of metabolism coefficients for a substance consumed at a rate proportional to its concentration, and the distribution of permeability coefficients for a slowly penetrating inert substance. Equations are derived for obtaining crude estimates of the moments of these distributions from exchange data.  相似文献

Scanning electron micrographs of the teeth and sections and dissections of the head have been used to describe the functional interrelationships between the dentition and associated cartilages, muscles and ligaments in adults of the southern hemisphere lamprey Geotria australis. These studies, together with manipulation of the piston and oral disc in living specimens, elucidated the probable feeding mechanism in this species. The main cutting action appears to result from a scissoring movement brought about by the rapid interlocking of the three sharp and stout cusps of the transverse lingual lamina with large grooves on the rear face of the supraoral lamina. The movement of excised flesh back through the oral passage to the pharynx would be facilitated by the action of the pair of strongly cuspid longitudinal lingual laminae. The unique oral disc teeth of G. australis are apparently adapted to allow the disc to slide forward over the host and yet restrict any tendency to slip backwards.  相似文献

Hitchhiking effects of advantageous mutations have been invoked to explain reduced polymorphism in regions of low crossing-over in Drosophila. Besides reducing DNA heterozygosity, hitchhiking effects should produce strong linkage disequilibrium and a frequency spectrum skewed toward an excess of rare polymorphisms (compared to the neutral expectation). We measured DNA polymorphism in a Zimbabwe population of D. melanogaster at three loci, yellow, achaete, and suppressor of forked, located in regions of reduced crossing-over. Similar to previously published surveys of these genomic regions in other populations, we observed low levels of nucleotide variability. However, the frequency spectrum was compatible with a neutral model, and there was abundant evidence for recombination in the history of the yellow and ac genes. Thus, some aspects of the data cannot be accounted for by a simple hitchhiking model. An alternative hypothesis, background selection, might be compatible with the observed patterns of linkage disequilibrium and the frequency spectrum. However, this model cannot account for the observed reduction in nucleotide heterozygosity. Thus, there is currently no satisfactory theoretical model for the data from the tip and base of the X chromosome in D. melanogaster.   相似文献

Oligogalacturonic acids (OGAs), derived from plant cell wall pectin, have been implicated in a number of signal transduction pathways involved in growth, development and defense responses of higher plants. This study investigates the size range of OGAs capable of inducing ethylene synthesis in tomato plants, and demonstrates that in contrast with many other effects, only short chain OGAs are active. Oligomers across a range of DP from 2-15 were separated and purified to homogeneity by QAE-Sephadex anion exchange chromatography using a novel elution system. The OGAs were applied to tomato plants and assayed for their ability to induce ethylene gas release and changes in steady state levels of mRNA encoding the ethylene forming enzyme aminocyclopropane-1-carboxylic acid oxidase (ACO). The study demonstrated that only OGAs in the size range of DP4-6 were active both in eliciting ACO expression and in the production of ethylene.   相似文献

Summary Ferritins from liver and spleen of both-thalassaemia/haemoglobin E (HbE) and non-thalassaemic patients were purified by heating a methanol-treated homogenate, followed by molecular exclusion chromatography. The concentrations of ferritins in the-thalassaemia/HbE liver and spleen were calculated as 3.8 and 2.0 mg/g wet tissue. The-thalassaemia/HbE ferritin iron/protein ratios were higher than those of normal ferritins. On PAGE, all ferritins gave a single major monomeric band with only very small differences in their mobility. Ferritins from thalassaemic patients also possessed bands corresponding to oligomers. On SDS/PAGE, all ferritins were resolved into two major subunits: H and L with L subunit predominating. While the isoferritin profiles of ferritins from-thalassaemia/HbE liver and spleen were similar to each other and to those of normal liver and spleen, some extra bands were present in the acidic region. One of the most spectacular motions is the generation of the acrosomal process in the limulus sperm. On contact with the egg, the sperm generates a 60-mum-long process that literally drills its way through the jelly surrounding the egg. This irresversible reaction takes only a few seconds. We suggested earlier that this motion is driven by a change in twist of the actin filaments comprising the acrosomal process. In this paper we analyze the so-called false discharge, a reversible reaction, in which the acrosomal filament bundle extends laterally from the base of the sperm and not anteriorly from the apex. Unlike the true discharge, which is straight, the false discharge is helical. Before extension, the filament bundle is coiled about the base of the sperm. In the coil, the bundle is not smoothly bent but consists of arms (straight segments) and elbows (corners) so that the coil looks like a 14-sided polygon. The extension of the false discharge works as follows: starting at the base of the bundle, the filaments change their twist which concomitantly changes the orientations of the elbows relative to each other; that is, in the coil, the elbows all like in a common plane, but after the change in twist, the plane of each elbow is rotated to be perpendicular to that of its neighbors. This change transforms the bundle from a compact coil into an extended left- handed helix. Because the basal end of the bundle is unconstrained, the extension is lateral. The true discharge works the same way but starts at the apical end of the bundle. The apical end, however, is constrained by its passage through the nuclear canal, which directs the extention anteriorly. Unlike the false discharge, during the true discharge the elbows are melted out, making the reaction irreversible. This study shows that rapid movement can be regenerated by actin without myosin and gives us insight into the molecular mechanism.  相似文献

Hypotension in adult animals recruits brain sites extending from cerebellar cortex to the midbrain and forebrain, suggesting a range of motor and endocrine reactions to maintain perfusion. We hypothesized that comparable neural actions during development rely more extensively on localized medullary processes. We used functional MRI to assess neural responses during sodium nitroprusside challenges in 14 isoflurane-anesthetized kittens, aged 14-25 days, and seven adult cats. Baseline arterial pressure increased with age in kittens, and basal heart rates were higher. The magnitude of depressor responses increased with age, while baroreceptor reflex sensitivity initially increased over those of adults. In contrast to a decline in adult cats, functional MRI signal intensity increased significantly in dorsal and ventrolateral medullary regions and the midline raphe in the kittens during the hypotensive challenges. In addition, significant signal intensity differences emerged in cerebellar cortex and deep nuclei, dorsolateral pons, midbrain tectum, hippocampus, thalamus, and insular cortex. The altered neural responses in medullary baroreceptor reflex sites may have resulted from disinhibitory or facilitatory influences from cerebellar and more rostral structures as a result of inadequately developed myelination or other neural processes. A comparable immaturity of blood pressure control mechanisms in humans would have significant clinical implications.  相似文献

Bulk water transport in reconstituted ghosts is statistically comparable to that in the parent red cells, and is unaffected by incorporation of Ca2+ over the range of 0.01 to 1 mM. Brief exposure of ghosts to p-chloromercuribenzene sulfonate results in a supression of osmotic water flow but leaves K+ permeability unchanged. Incorporation of p-chloromercuribenzene sulfonate provokes extremely rapid K+ loss which can be counteracted by simultaneous inclusion of Ca2+.Erythrocyte ghosts, when prepared with a small amount of Ca2+, demonstrate recovery of normal impermeability to choline, sucrose, Na+ and inulin and have an improved K+ retention over Ca2+-free preparations.The rate of passive transport of K+ from unwashed erythrocyte ghosts was measured during the initial few minutes of efflux. The initial rates vary in a bimodal fashion with the concentration of Ca2+ incorporated at the time of hemolysis. In low concentrations (0.01–0.1 mM), Ca2+ protects the K+ barrier while at higher concentrations (0.1–1.0 mM) it provokes a K+ leakage ranging from 7 to 50 times the normal rate of passive K+ loss. The Ca2+-induced K+ leak is thus a graded response rather than a discrete membrane transport state. The transition from a Ca2+-protected to a Ca2+-damaged membrane occurs upon an increase in Ca2+ concentration of less than 50 μmoles/l.  相似文献

Summary The major iron binding protein (IBP) of larvalM. mordax has an estimated molecular weight (354,000), subunit molecular weight (18,000) and pI (5.1) identical to those recorded previously for larvalG. australis. The IBP in larvalG. australis has also been shown to be relatively heat stable and to react immunologically with antihorse spleen ferritin. The weight of total non-haem iron in the whole body, and both the ferritin and haemosiderin iron components, increased with increasing body weight in larvalG. australis. While the concentration of ferritin iron remained similar throughout larval life, the concentration of total non-haem iron and haemosiderin iron increased rapidly in animals up to a body weight of 0.1–0.2 g, but thereafter rose only slowly throughout the rest of larval life. This implies that any iron in excess of the amount required for the maintenance of a constant ferritin concentration is converted into haemosiderin iron, and that once non-haem iron has reached a particular concentration (c. 500–600 g g–1), the rate of iron accumulation is greatly reduced. While the larvae of bothG. australis andM. mordax had very high plasma iron levels (>19,000 g 100 ml–1), the former had significantly greater concentrations of iron in the whole body (702vs. 267 g g–1) and more particularly in the nephric fold (7382vs. 224 g g–1). A greater reservoir of non-haem iron could facilitate the maintenance of the large amounts of haem and erythrocytic ferritin present in this species as a result of an exceptionally high haemoglobin concentration and red blood cell number. The greater concentration of non-haem iron in the intestine ofM. mordax than ofG. australis (1338vs. 824 g g–1), when considered in conjunction with histological studies, indicates thatMordacia mordax eliminates a larger amount of iron during the extrusion of its intestinal columnar cells.Abbreviation IBP iron binding protein  相似文献
