Continuous production of L-phenylalanine by transamination 总被引:2,自引:0,他引:2
L-Phenylalanine was produced continuously from L-as-partate and phenylpyruvate by transaminase from a newly screened Pseudomonas putida strain. The process was carried out with an isolated enzyme in homogeneous phase in an enzyme membrane reactor and with immobilized whole cells in a stirred tank reactor, respectively. Due to the difference in transport resistance, the productivity of the free enzyme in homogeneous phase (72 mmol/L h) was about 3 times higher than the productivity achieved using immobilized cells. 本文讨论了将光质应用于一品红(Euphorbia pulcherrima)大规模细胞繁殖及形态发生调控的可行性,并测定了一些形态及生化指标作为这种调控的参数。 以一品红试管苗的叶片作为外植体,置于MS培养基中并分别培养于白(W)、红(R)、黄(Y)、蓝(B)、绿(G)色光及黑暗(D)下,一个月后进行测定。蓝光和黄光明显促进出苗。而愈伤组织形成的情况则正相反,红光利于愈伤组织的形成,黄光效果最差。过氧化物酶、过氧化氢酶活力及蛋白质、RNA、碳水化合物的含量与愈伤组织形成有较为一致的关系,但与分化不直接相关。 实验中还测定了叶绿素a、叶绿素b及类胡萝卜素的含量。白光与蓝光均能促进叶绿素及类胡萝卜素的合成,但蓝光中chl b/chl a的比率较高。此外,本文尚涉及暗处理中的晶质化现象(vitrification)及其成因。 相似文献

Cells of the cyanobacterium, Synechococcus PCC7942, grown under high inorganic carbon (Ci) conditions (1% CO2; pH 8) were found to be photosynthetically dependent on exogenous CO2. This was judged by the fact that they had a similar photosynthetic affinity for CO2 (K0.5[CO2] of 3.4-5.4 micromolar) over the pH range 7 to 9 and that the low photosynthetic affinity for Ci measured in dense cell suspensions was improved by the addition of exogenous carbonic anhydrase (CA). The CA inhibitor, ethoxyzolamide (EZ), was shown to reduce photosynthetic affinity for CO2 in high Ci cells. The addition of 200 micromolar EZ to high Ci cells increased K0.5(CO2) from 4.6 micromolar to more than 155 micromolar at pH 8.0, whereas low Ci cells (grown at 30 microliters CO2 per liter of air) were less sensitive to EZ. EZ inhibition in high and low Ci cells was largely relieved by increasing exogenous Ci up to 100 millimolar. Lipid soluble CA inhibitors such as EZ and chlorazolamide were shown to be the most effective inhibitors of CO2 usage, whereas water soluble CA inhibitors such as methazolamide and acetazolamide had little or no effect. EZ was found to cause a small drop in photosystem II activity, but this level of inhibition was not sufficient to explain the large effect that EZ had on CO2 usage. High Ci cells of Anabaena variabilis M3 and Synechocystis PCC6803 were also found to be sensitive to 200 micromolar EZ. We discuss the possibility that the inhibitory effect of EZ on CO2 usage in high Ci cells of Synechococcus PCC7942 may be due to inhibition of a `CA-like' function associated with the CO2 utilizing Ci pump or due to inhibition of an internal CA activity, thus affecting CO2 supply to ribulose bisphosphate carboxylase-oxygenase. 相似文献

An enriched IgG serum fraction obtained from rabbits immunized against pea chloroplast fructose-1,6-bisphosphatase (FBPase) was used, coupled to colloidal gold (15 nanometer particles) goat anti-rabbit IgG, to analyze by electron microscopy the location of photosynthetic FBPase in pea (Pisum sativum L.) leaf ultrathin sections. In accordance with earlier biochemical studies on distribution of FBPase activity, the enzyme was visualized both in the stromal space and bound to the chloroplast membranes. Some gold particles also appear in the cytoplasm, which can be related to the presence in the cytosol of a high molecular weight precursor of this nuclear coded enzyme. 相似文献

Leghemoglobin was localized by immunogold techniques in nodules of Lupinus albus cv Multolupa inoculated with Bradyrhizobium sp. (Lupinus) strain ISLU 16. The protein localization was performed in nodules embedded in Spurr's and Araldite epoxy resins and Lowycryl K4M. A very good preservation of both the ultrastructure and antigenicity was obtained with the tissues embedded in Araldite following glutaraldehyde fixation and unpostfixed in osmium tetroxide. Lupin leghemoglobin is a stable and abundant protein which allows a conventional method to be safely used for localization of leghemoglobin. Labeling of leghemoglobin was specifically confined to the cytosol matrix and nuclei. Gold particles were never observed in the peribacteroidal spaces nor in the cytoplasmic organelles of the infected cells. Decrease of leghemoglobin was observed when the plants were grown with 10.7 micromolar and 21.4 micromolar of nitrate. 相似文献

The total glucose metabolism of 48-h spherical trophoblastic vesicles, Day-60 trophoblastic vesicles sections and Day-14 porcine blastocyst sections was measured by the method of O'Fallon and Wright (1). Trophoblastic vesicles were formed by enzyme dispersal in Day-14 porcine blastocysts. Glucose was based on DNA content of the tissue measured by diamino benzoic acid reaction with DNA (2). Slope of the lines (PMoles glucose utilized/4 h x DNA content) was different between Day-14 blastocyst sections and 48 h trophoblastic vesicles (P = 0.062) and between Day-14 blastocyst sections and Day-60 trophoblastic vesicles sections (P >/= 0.05). Slopes of the lines were identical between 48-h trophoblastic vesicles and Day-60 trophoblastic vesicles sections (P >/= 0.87). Average glucose utilization on a per ng DNA basis was calculated. Day-14 blastocyst sections utilized 0.67 Pmoles glucose/4 h per ng DNA, Day-60 trophoblastic vesicles sections; 0.57; and 48-h sperical trophoblastic vesicles used 0.29. It is hypothesized that the change in glucose utilization between the Day-14 porcine blastocyst and enzymatically formed trophoblastic vesicles may be due to a decrease in metabolism as a consequence of in vitro culture. Further, it is theorized that Day-60 trophoblastic vesicles sections used higher quantities of glucose than 48-h sperical trophoblastic vesicles on a per ng DNA basis due to the increased availability of glucose to the cells of the inner layers, caused by the sectioning of the tissue. The results of this study identify changes in glucose metabolism of enzymatically formed porcine trophoblastic vesicles during culture. It is proposed that enzymatically-formed trophoblastic vesicles be used as a model system for the study of embyro metabolism. 相似文献

Eighty Holstein dairy cows were treated with 25 mg of prostaglandin F2alpha (PGF2alpha) on Days 14 to 16 post calving. Eighty-four herd mates served as saline controls in the double blind study. The reproductive parameters used to measure fertility were mean days to first service on all cows and mean days open, first service conception rates and services per pregnancy on cows that became pregnant. Mean days to first service was similar in both groups (71.8 +/- 27, treated; 68.5 +/- 28.6, controls; P = 0.5352). The mean days open was 98.6 +/- 52.0 d for treated cows compared to 118.8 +/- 71.2 d for controls (P = 0.0680). First service pregnancy rates (treated, 41.3%; control, 35.7%) were not significantly different (P = 0.0630); however, the mean services per pregnancy (treated, 1.64; control, 2.33) were significantly (P = 0.0021) improved in the treated group. Ten cows in the treated group and twelve cows in the control group were diagnosed as having retained fetal membranes and/or metritis. For treated and control cows mean days to first service were 82.2 +/- 34.8 d and 82.8 +/- 58.7 d (P = 0.5652). Days open were 97.0 +/- 32.5 d and 133.4 +/-58.4 d (P = 0.3636); services per conception were 1.83 and 2.50 (P = 0.3178), respectively. In all treated cows reproductive parameters were similar whether cows had high serum progesterone ( > 1 ng/ml) or low serum progesterone ( < 1 ng/ml) on the treatment day. This study suggests that early postpartum treatment of lactating dairy cows with prostaglandin produces an improvement in fertility. 相似文献

Nuclear magnetic resonance (NMR) has become a valuable tool for the study of metabolism in a wide variety of biological systems. Its inherent advantages are that it is non-destructive and non-invasive. Observations can be carried out not only on extracts and media but also on whole cells and whole tissues under varying conditions and over varying times. The information gained gives considerable insight into cellular metabolism. There has been, to date, relatively little literature on the application o f NMR to the biochemistry of parasites, presumably reflecting the paucity of interfaces between parasitologists and NMR practitioners as well as the inherent difficulties in obtaining sufficient parasite material for NMR experiments. These difficulties are being overcome and William O'Sullivan, Michael Edwards and Raymond Norton believe that NMR has a great deal to offer those interested in parasite metabolism. In particular, it has the capacity to turn up the unexpected, on important factor as so many parasites appear to have developed their own variations on orthodox metabolic pathways. 相似文献
