Semi-hard cheeses were experimentally elaborated with pasteurized milk from sheep, goat and cow (15: 35: 50) and inoculated to contain 1.9 times 105 Listeria monocytogenes /ml in cheeses 1 and 2 and 4 times 103 L. monocytogenes /ml in cheeses 3 and 4. Counts of L. monocytogenes were determined by direct surface plating of samples on listeria selective agar medium. The results show the substantial survival of L. monocytogenes present in milk during manufacture and ripening of this type of cheese.  相似文献

Seedlings from Euphorbia canariensis and Euphorbia lambii weregrown in the dark at 25 °C. Protein and triglyceride contentas well as levels of sugars and amino acids in the endospermwere determined during endosperm depletion. In the endospermof Euphorbia canariensis, relatively low levels of amino acids(up to 1 µmol.endosperm–1) were found of which glutamine/glutamateaccounted for 40% at the stage of radicle emergence. High levelsof amino acids (up to 4 µmol.endosperm–1) comparedwith sugars (up to 2 µmol sucrose.endosperm–1) weredetected in the endosperm of Euphorbia lambii. Arginine wasthe main component (28 µmol%) of the amino acids in thistissue. In both species amino acid composition changed graduallyduring endosperm depletion. Cotyledons retained their ability to absorb a variety of watersoluble substrates after removal of the endosperm. 14C from[U-14C]sucrose was effectively incorporated into the triterpenesof the laticifers and to a lesser extent into the sterols ofthe seedling. The highest incorporation values were found inyoung seedlings about 2 d after the emergence of the radicle.Seedlings of this age also showed high incorporation rates of14C from labelled alanine, serine, threonine, valine, leucineand isoleucine into both triterpenols and sterols, but no generalconclusions about metabolic channelling in lipid synthesis couldbe made. Phylogenetic information is useful in understanding the evolutionary history of adaptive traits. Here, we present a well-resolved phylogenetic hypothesis for Heliconius butterflies and related genera. We use this tree to investigate the evolution of three traits, pollen feeding, pupal-mating behaviour and larval gregariousness. Phylogenetic relationships among 60 Heliconiina species (86% of the subtribe) were inferred from partial DNA sequences of the mitochondrial genes cytochrome oxidase I , cytochrome oxidase II and 16S rRNA, and fragments of the nuclear genes elongation factor-1α , apterous , decapentaplegic and wingless (3834 bp in total). The results corroborate previous hypotheses based on sequence data in showing that Heliconius is paraphyletic, with Laparus doris and Neruda falling within the genus, demonstrating a single origin for pollen feeding but with a loss of the trait in Neruda . However, different genes are not congruent in their placement of Neruda ; therefore, monophyly of the pollen feeding species cannot be ruled out. There is also a highly supported monophyletic 'pupal-mating clade' suggesting that pupal mating behaviour evolved only once in the Heliconiina. The present study employed basic and molecular cytogenetic methods to characterize three populations of Parauchenipterus galeatus from the basins of the Paraná and São Francisco Rivers, and a region of connection between the two basins. Although the diploid number was equal to 58 chromosomes, variations in karyotype formula were detected among the populations. B chromosomes were detected only in the population from the São Francisco River. Heterochromatin was located in the terminal position in almost all the chromosomes and in the pericentromeric position in some acrocentric chromosomes in the three populations. A single nucleolus organizer region was detected by silver nitrate and 18S rDNA‐fluorescent in situ hybridization in the short arm of one subtelocentric pair in the three populations, varying only in the chromosome pair bearing this site. The 5S rDNA sites were located in two submetacentric chromosome pairs in the three populations, varying only in the chromosome pairs bearing these sequences. Classic and molecular chromosome markers, along with the context of the natural history of the formation of hydrographic basins, ecological aspects, and the geographic isolation of populations between hydrographic basins and within the same basin, were important contributions to the discussion on possible biogeographic relations among the populations of Parauchenipterus galeatus. © 2010 The Linnean Society of London, Biological Journal of the Linnean Society, 2010, 99 , 648–656.  相似文献

Coconut (Cocos nucifera L.) WRKY sequences containing single nucleotide polymorphisms (SNPs) and one microsatellite repeat were used to develop 10 informative markers. These markers were evaluated in 15 genotypes representing six coconut cultivars. SNP‐containing alleles were detected by single‐strand conformation polymorphism (SSCP) analysis. The number of detected alleles ranged from two to four. Five pairs of loci were in linkage disequilibrium in the test population. These markers are currently being evaluated in more individuals/cultivars to determine their value in estimating the genetic diversity of this species.  相似文献

Abstract In this study, we show that the house mouse (Mus musculus) is the most abundant small mammal in the National Reserve of Lachay in central Peru, and that its large population fluctuations are independent of seasonality. Also, we found that M. musculus is the main small mammal prey of burrowing owls (Athene cunicularia) in Lachay, and that owls respond functionally to mouse abundance. In addition, vegetation cover seems to have a strong effect on small mammal predation by burrowing owls, and possibly other predators such as eagles and foxes. We propose the hypothesis that burrowing owls in arid environments can survive several months eating only arthropods, but that their reproduction is synchronized with a highly nutritious diet, provided by small mammal consumption. The principal prediction of this alternative prey hypothesis is now supported by our data from a tropical ecosystem.  相似文献

We compared the breeding phenology and clutch size of Choughs Pyrrhocorax pyrrhocorax in three Spanish areas that differed in feeding habitat, breeding environmental conditions and the distribution and availability of different nesting sites (cliffs or artefacts). The variation in the timing of laying seemed to be related to differences in climatic conditions determined by the altitude. The variation in clutch size was associated with the different use that Choughs made of the feeding areas because of the contrasting availability and distribution of their nesting sites. Within southeastern Madrid, clutch size of the Chough decreased and its variance increased at high nest densities on cliff sites, suggesting that density dependence in fecundity arises from habitat heterogeneity rather than by interference. At high densities, individuals, territories, nest sites or a combination of these may differ in quality, thus promoting differences and increasing the variance in the initial investment of reproductive effort. The syncronization of the onset of laying at increased breeding densities suggests that social influences arising from communal foraging or avoidance of predation by early warning and predator swamping may be acting also. We suggest that nest-site availability and distribution have a major influence on the social organization of Choughs through their breeding and foraging strategies.  相似文献

Thirteen polymorphic microsatellite loci suitable for population genetic studies of Helicoverpa zea were discovered by screening partial genomic libraries enriched for microsatellite sequences. Insects collected (N = 96) in Stoneville, Mississippi were used to characterize these markers. The observed and effective number of alleles per locus ranged from two to nine (average 4.46) and from 1.07 to 2.45 (average 1.81), respectively. Fisher exact tests detected significant deviations from Hardy–Weinberg equilibrium at three loci, probably due to inbreeding, null alleles, or Wahlund's effect. Significant genotypic disequilibrium was not detected between any pair of loci.  相似文献
