The present study analysed the concordance among four different molecular diagnostic
methods for tuberculosis (TB) in pulmonary and blood samples from immunocompromised
patients. A total of 165 blood and 194 sputum samples were collected from 181 human
immunodeficiency virus (HIV)-infected patients with upper respiratory complaints,
regardless of suspicious for TB. The samples were submitted for smear microscopy,
culture and molecular tests: a laboratory-developed conventional polymerase chain
reaction (PCR) and real-time quantitative PCR (qPCR) and the Gen-Probe and Detect-TB
Ampligenix kits. The samples were handled blindly by all the technicians involved,
from sample processing to results analysis. For sputum, the sensitivity and
specificity were 100% and 96.7% for qPCR, 81.8% and 94.5% for Gen-Probe and 100% and
66.3% for Detect-TB, respectively. qPCR presented the best concordance with sputum
culture [kappa (k) = 0.864)], followed by Gen-Probe (k = 0.682). For blood samples,
qPCR showed 100% sensitivity and 92.3% specificity, with a substantial correlation
with sputum culture (k = 0.754) and with the qPCR results obtained from sputum of the
corresponding patient (k = 0.630). Conventional PCR demonstrated the worst results
for sputa and blood, with a sensitivity of 100% vs. 88.9% and a specificity of 46.3%
vs. 32%, respectively. Commercial or laboratory-developed molecular assays can
overcome the difficulties in the diagnosis of TB in paucibacillary patients using

The late summer mass occurrences of cyanobacteria in the Baltic Sea are among the largest in the world. These blooms are rarely monotypic and are often composed of a diverse assemblage of cyanobacteria. The toxicity of the blooms is attributed to Nodularia spumigena through the production of the hepatotoxic nodularin. However, the microcystin hepatotoxins have also been reported from the Baltic Sea on a number of occasions. Recent evidence links microcystin production in the Gulf of Finland directly to the genus Anabaena . Here we developed a denaturing gradient gel electrophoresis (DGGE) method based on the mcyE microcystin synthetase gene and ndaF nodularin synthetase gene that allows the culture-independent discrimination of microcystin- and nodularin-producing cyanobacteria directly from environmental samples. We PCR-amplified microcystin and nodularin synthetase genes from environmental samples taken from the Gulf of Finland and separated them on a denaturing gradient gel using optimized conditions. Sequence analyses demonstrate that uncultured microcystin-producing Anabaena strains are genetically more diverse than previously demonstrated from cultured strains. Furthermore, our data show that microcystin-producing Anabaena are widespread in the open Gulf of Finland. Non-parametric statistical analysis suggested that salinity plays an important role in defining the distribution of microcystin-producing Anabaena . Numerous human pathologies result from unrepaired oxidative DNA damage. Base excision repair (BER) is responsible for the repair of oxidative DNA damage that occurs in both nuclei and mitochondria. Despite the importance of BER in maintaining genomic stability, knowledge concerning the regulation of this evolutionarily conserved repair pathway is almost nonexistent. The Saccharomyces cerevisiae BER protein, Ntg1, relocalizes to organelles containing elevated oxidative DNA damage, indicating a novel mechanism of regulation for BER. We propose that dynamic localization of BER proteins is modulated by constituents of stress response pathways. In an effort to mechanistically define these regulatory components, the elements necessary for nuclear and mitochondrial localization of Ntg1 were identified, including a bipartite classical nuclear localization signal, a mitochondrial matrix targeting sequence and the classical nuclear protein import machinery. Our results define a major regulatory system for BER which when compromised, confers a mutator phenotype and sensitizes cells to the cytotoxic effects of DNA damage. 相似文献

The host‐associated microbiome plays a significant role in health. However, the roles of factors such as host genetics and microbial interactions in determining microbiome diversity remain unclear. We examined these factors using amplicon‐based sequencing of 175 Thoropa taophora frog skin swabs collected from a naturally fragmented landscape in southeastern Brazil. Specifically, we examined (1) the effects of geography and host genetics on microbiome diversity and structure; (2) the structure of microbial eukaryotic and bacterial co‐occurrence networks; and (3) co‐occurrence between microeukaryotes with bacterial OTUs known to affect growth of the fungal pathogen Batrachochytrium dendrobatidis (Bd). While bacterial alpha diversity varied by both site type and host MHC IIB genotype, microeukaryotic alpha diversity varied only by site type. However, bacteria and microeukaryote composition showed variation according to both site type and host MHC IIB genotype. Our network analysis showed the highest connectivity when both eukaryotes and bacteria were included, implying that ecological interactions may occur among domains. Lastly, anti‐Bd bacteria were not broadly negatively co‐associated with the fungal microbiome and were positively associated with potential amphibian parasites. Our findings emphasize the importance of considering both domains in microbiome research and suggest that for effective probiotic strategies for amphibian disease management, considering potential interactions among all members of the microbiome is crucial. 相似文献
