Persistent DNA damage triggers cellular senescence, which may play an important role in the pathogenesis of cigarette smoke (CS)-induced lung diseases. Both p21CDKN1A (p21) and poly(ADP-ribose) polymerase-1 (PARP-1) are involved in DNA damage and repair. However, the role of p21-PARP-1 axis in regulating CS-induced lung DNA damage and cellular senescence remains unknown. We hypothesized that CS causes DNA damage and cellular senescence through a p21-PARP-1 axis. To test this hypothesis, we determined the levels of γH2AX (a marker for DNA double-strand breaks) as well as non-homologous end joining proteins (Ku70 and Ku80) in lungs of mice exposed to CS. We found that the level of γH2AX was increased, whereas the level of Ku70 was reduced in lungs of CS-exposed mice. Furthermore, p21 deletion reduced the level of γH2AX, but augmented the levels of Ku70, Ku80, and PAR in lungs by CS. Administration of PARP-1 inhibitor 3-aminobenzamide increased CS-induced DNA damage, but lowered the levels of Ku70 and Ku80, in lungs of p21 knockout mice. Moreover, 3-aminobenzamide increased senescence-associated β-galactosidase activity, but decreased the expression of proliferating cell nuclear antigen in mouse lungs in response to CS. Interestingly, 3-aminobenzamide treatment had no effect on neutrophil influx into bronchoalveolar lavage fluid by CS. These results demonstrate that the p21-PARP-1 pathway is involved in CS-induced DNA damage and cellular senescence.  相似文献

(NIn)‐Formyl protective group of tryptophan has been introduced as a base/nucleophile‐labile protective group. It has long been known that a free ‐amino group of the peptide can serve as a nucleophile: an irreversible formyl NIn → NH2 transfer is consistently observed when deformylation is performed last on an otherwise deprotected peptide that possesses free ‐amino group. Obviously, this particular side reaction should be expected any time free amino group is exposed to Trp(For), but, at the best of our knowledge, has never been reported in the course of Boc‐SPPS. In the present communication, we describe a set of appropriately designed model experiments that permitted to detect the title side reaction both in solution and in solid‐phase reactions. We observed intermolecular formyl group transfer with a model compound, Trp(For)‐NH2. Importantly, we also observed this migration on solid support with the rate roughly estimated to be up to 1% of residues per minute. We also observed that the formyl‐group transfer reaction occurred in a sequence‐dependent manner and was suppressed to a non‐detectable level using ‘in situ neutralization’ technique. Because this side reaction is sequence dependent, there might be situations when the rate of the formation of Nα‐formyl termination by‐products is significant. In other cases, the Nα‐For truncated by‐products would not contaminate the final peptide significantly but still could be a source of microheterogeneity. The CYP2E1 gene polymorphism has been studied in Yakut women with ovarian cancer and without cancer. The two groups have been found to substantially differ in the frequency of the CYP2E1* 1D allele (with a 96-bp insertion in the promoter region of the gene): it is more frequent in healthy women (16.3 versus 7.4%, P = 0.007).  相似文献

It has been demonstrated that SPPS with Boc amino group protection can be monitored spectrophotometrically if it is performed in a continuous-flow reactor of variable volume. It is shown that this approach provides useful and adequate evidence on the beginning/end-point of most steps of the SPPS cycle. At the deprotection step the spectrophotometric monitoring enables real-time recording of the initial and final moments of the process. When synthesizing a 'difficult' polyalanine sequence, we were able to monitor variation in the deprotection dynamics associated with the aggregation phenomena. The time actually necessary for the Boc protecting group removal appeared to be significantly smaller than that usually preset in the available Boc-SPPS protocols.  相似文献

Restriction polymorphism at the monoamine oxidase A (MAO A) gene was typed in eight male populations of the Volga– Ural region (Bashkirs, Chuvashes, Tatars, Udmurts, Maris, Mordovians, Komis, and Russians inhabiting the Republic of Bashkortostan). Analysis of the MAO A alleles frequency distribution patterns did not reveal statistically significant differences between the Volga–Ural populations examined. The results obtained suggest genetic homogeneity of the populations described in respect of the polymorphic locus examined.  相似文献

The influence of leptin and ghrelin, as well as their combined effects, on the expression of membrane molecules and cytokine production by NK cells from peripheral blood was studied in vitro. The effects of hormones were assayed at the concentrations corresponding to their peripheral blood levels in the course of physiological pregnancy. It was established that the investigated hormones exerted significant effects only at the concentrations typical of the II–III trimester of pregnancy. In particular, leptin and ghrelin and their combination increased the number of CD56brightNKp46+NK cells in the suspension of mononuclear cells and inhibited the expression of homing molecules CCR7 and inhibitor molecules LILRB in NKp46+NK cells. Leptin and its combination with ghrelin increased the expression of L-selectin in CD56brightNKp46+NK cells but inhibited the secretion of IL-10 by NKp46+NK cells. Leptin reduced the production of IL-4 by NKp46+ cells, while ghrelin eliminated this effect. The hormones did not influence the expression of inhibitory molecules NKG2A in NKp46+ cells and the production of TGF-β1, IL-17A, and IFN-γ by these cells. Thus, the investigated hormones at the concentrations typical of the II–III trimester of pregnancy effectively regulate the expression of membrane molecules and cytokine production by NK cells of the peripheral blood.  相似文献

Distribution of dissolved organic carbon in Lake Baikal and its watershed   总被引:2,自引:0,他引:2  
 Concentrations of dissolved organic carbon (DOC) in Lake Baikal ranged from 90 to 110 μM C, considerably higher than those in oceanic environments. The DOC concentrations in the epilimnion were higher than those in the hypolimnion. Since particulate organic carbon (POC) concentrations in the pelagic waters of Lake Baikal were <10–40 μM C in the epilimnion and 2–5 μM C in the hypolimnion, DOC constitutes a major component of the organic carbon pool in Lake Baikal, especially in the deep layers. The DOC concentrations downstream of the Barguzin and Selenga Rivers were quite high (400–500 μM C). Probably because of the high concentrations of DOC in these rivers, the DOC levels in Barguzin Bay and offshore at the mouth of the Selenga River were higher than those in the pelagic regions of the central and south basins of Lake Baikal. The relationship between DOC and electric conductivity revealed the transport of DOC from rivers to the pelagic area in Lake Baikal. The spatial distribution of DOC suggested that a major part of DOC in the lake was allochthonous (land-derived).
