Summary Experiments were undertaken on the fresh water algaChara corallina to determine the form of inorganic carbon (CO2 or HCO 3 ) which enters the cell during photosynthesis at alkaline pH. Recent proposals have centered on the possibility that proton efflux in alkaline solution is able to generate, in the immediate vicinity of the cell, a sufficiently low pH to raise the partial pressure of CO2, and hence facilitate its passive permeation into the cell. Predictions have been made by modelling this situation (N.A. Walker, F.A. Smith & I.R. Cathers, 1980,J. Membrane Biol. 5751–58, J.M. Ferrier, 1980,Plant Physiol. 661198–1199), and these were tested by placing recessed-tip pH microelectrodes in the unstirred layer surrounding cells in stagnant solution (bulk pH 8.2, buffered only with 1mm HCO 3 ). Even as close as 2 m from the cell wall, the pH was typically 7.2 to 7.6 in the acid band center — over 1 pH unitgreater than that suggested by the models for CO2 entry at the necessary rate for C-fixation. Further evidence for the entry of HCO 3 , rather than CO2, at high solution pH was obtained from experiments in which the radial pH gradient in the unstirred layer was reduced. Buffer solutions containing 5mm phosphate or 5mm HEPES, raised the pH at the cell surface in the acid regions from around 7.2 to 7.8 or higher. This pH increase (reduction in acid gradient) would have greatly reduced the CO2 level at the cell surface and should, therefore, have greatly reduced the CO2-related14C-influx. However,14C-fixation was reduced by only 31% (phosphate) or 15% (HEPES), compared with buffer-free controls. Reduction of the unstirred layer thickness by fast solution flow resulted in a stimulation, and not a reduction, of14C-fixation. The similarity of our radial pH profiles near the wall with that predicted by the model (Walker et al., 1980) assuming H+–HCO 3 cotransport, together with the effects of buffer, and the results of increased solution flow rate, lead to the conclusion that cotransport of HCO 3 with H+ is the likely method of entry of inorganic carbon. Longitudinal pH profiles of theChara cell were obtained at a distance of 25 m from the wall. These revealed much sharper delineation of the acid and alkaline bands than has previously been possible with miniature pH electrodes. Profiles of local electric field, obtained with a vibrating probe, were in excellent agreement with the high resolution pH profiles. This supports the hypothesis that membrane proton transport has a role (direct) in the generation of the extracellular currents.  相似文献

A dramatic difference is observed in the intracellular distribution of the high mobility group (HMG) proteins when chicken embryo fibroblasts are fractionated into nucleus and cytoplasm by either mass enucleation of cytochalasin-B-treated cells or by differential centrifugation of mechanically disrupted cells. Nuclei (karyoplasts) obtained by cytochalasin B treatment of cells contain more than 90 percent of the HMG 1, while enucleated cytoplasts contain the remainder. A similar distribution between karyoplasts and cytoplasts is observed for the H1 histones and the nucleosomal core histones as anticipated. The presence of these proteins, in low amounts, in the cytoplast preparation can be accounted for by the small percentage of unenucleated cells present. In contrast, the nuclei isolated from mechanically disrupted cells contain only 30-40 percent of the total HMGs 1 and 2, the remainder being recovered in the cytosol fraction. No histone is observed in the cytosol fraction. Unike the higher molecular weight HMGs, most of the HMGs 14 and 17 sediment with the nuclei after cell lysis by mechanical disruption. The distribution of HMGs is unaffected by incubating cells with cytochalasin B and mechanically fractionating rather than enucleating them. Therefore, the dramatic difference in HMG 1 distribution observed using the two fractionation techniques cannot be explained by a cytochalasin-B-induced redistribution. On reextraction and sedimentation of isolated nuclei obtained by mechanical cell disruption, only 8 percent of the HMG 1 is released to the supernate. Thus, the majority of the HMG 1 originally isolated with these nuclei, representing 35 percent of the total HMG 1, is stably bound, as is all the HMGs 14 and 17. The remaining 65 percent of the HMGs 1 and 2 is unstably bound and leaks to the cytosol fraction under the conditions of mechanical disruption. It is suggested that the unstably bound HMGs form a protein pool capable of equilibrating between cytoplasm and stably bound HMGs.  相似文献

Viable interspecies cytoplasmic-nuclear hybrid cells were constructed by fusion of karyoplasts prepared from the highly tumorigenic A9 mouse fibroblast cell line and cytoplasts prepared from the Detroit 532 normal human diploid cell strain. The identity of the hybrid cells was ascertained using a variety of morphological, immunological, and genetic criteria, including: nuclear pattern of staining with the fluorochrome Hoechst 33258, appearance of the actin-myosin containing cytoskeleton, presence of fibronectin, and resistance to azaguanine and diphtheria toxin. About 90% of the hybrid cells were viable, that is, capable of division. Changes in the morphology of the hybrid cells, apparently nuclear directed, were observed before cell division occurred. Using the techniques described here, large numbers of interspecies hybrid cells suitable for many types of biochemical analyses can be routinely produced.  相似文献

Electroconvulsive shock (ECS) in rats produced a generalized seizure which was followed by an opiate-like catalepsy and an increase in hot-plate escape latencies. Preinjection of naloxone, at doses of 3.0 and 10.0 mg/Kg, significantly diminished the ECS-induced increase in hot-plate latencies. Paradoxically, simultaneous measurement of tail-flick latencies in these same rats demonstrated opiate-agonist effects of naloxone. The cataleptic effects of ECS were demonstrated to be opiate-like by evaluating righting reflexes, grid responses, and haloperidol effects. Colonic temperatures were also measured in all animals. These data, collectively discussed relative to affective and reflexive components of nociceptive behaviors, support the hypothesis that selective endorphin systems are activated by ECS. Moreover, these observations suggest consideration of a role for endorphin systems in the therapeutic mechanisms of electroconvulsive therapy (ECT) in man.  相似文献

The cyanobacterium (blue-green alga) Synechococcus 6301 incorporated a large amount of isotope from [1-14C] and [2-14C]acetate into phaeophorbide a obtained from chlorophyll a and into glutamatein cell protein; very little radioactivity was present in aspartate in cell protein. This distribution of isotope indicates that aspartate and the tetrapyrrole of chlorophyll a are not derived from a common C4, precursor. The ratios of the specific radioactivities of phaeophorbide a to glutamate for organisms grown in the presence of 1-14C] and [2-4C ] acetate were 2.5:1 and 10:1 respectively. These are close to the theoretical values for the C5, route to δ-aminolaevulinate which indicates that this is the only pathway to the tetrapyrrole precursor in Synechococcus 6301.  相似文献

Summary We report on an unusual phenomenon which occurs in some characean algae as a normal plasma membrane activity and also in association with charasome formation. The phenomenon of formation of coated invaginations of the plasma membrane was observed in twoChara and 6Nitella species. These invaginations are coated on their cytoplasmic surface, are 50–60 nm in diameter and rarely exceed 60 nm in length. They are abundant in the young cells ofChara andNitella and also occur in mature cells, but at a lower frequency.N. translucent is an exception in that coated invaginations were few in the young cells and absent in mature cells. Coated vesicles (50–60 nm diameter) were closely associated with these invaginations. Our observations suggest the vesicles may be derived from the invaginations by endocytosis.A close relationship was noted between the development of charasomes (plasmalemma modifications) and coated invaginations. Numerous coated invaginations are seen along the membranes of young charasomes; these invaginations appear to be associated with growth of the charasomes. Coated vesicles were not associated with the coated invaginations of the charasome membrane. The tubular network of cytoplasm and wall space seen in the mature charasome may be formed by fusion of coated invaginations of the developing charasomes, leaving cytoplasmic strands between the fused portions. Coated invaginations were not present along charasomes of the mature cells.  相似文献

Stain precipitates resulting from the use of lead or uranyl salts, or both, on ultrathin sections can be classified as belonging to one of three morphological types: I) extremely electron-dense particles caused by prolonged use of lead salts only, II) amorphous networks formed following double staining with either aqueous or alcoholic uranyl and lead salts, and III) crystalline needles sometimes resulting from double staining with alcoholic uranyl and lead salts. It has been found, however, that either acetic acid or aqueous uranyl acetate can be used to remove type I and type II precipitates from sections, and that oxalic acid and alcoholic uranyl solution will remove type II precipitates. Unfortunately, type III precipitates are unaffected by any agents tested so far.  相似文献

At a mean age of 93 +/- 5 days and a mean weight of 29 +/- 5 kg, 44 crossbred ram lambs were castrated to evaluate the accuracy of the estimation of testis weight from in situ testis measures (scrotal circumference and testis diameter). Means and standard deviations for testis weight (both testes), scrotal circumference and average in situ testis diameter were 134 +/- 57 g, 20.2 +/- 3.1 cm and 3.7 +/- .7 cm, respectively. Testis weight (W) was predicted from scrotal circumference (C) and average in situ diameter (D(o)) as W=.131C(1.90) D(o)(.88) (R(2)=.949). When adjusted to the same scrotal circumference and in situ diameter, testes of 3/4-Finnish Landrace rams were heavier (P<.05) than testes of 7/8-Dorset rams. However, the additional accuracy obtained by using equations specific to each crossbred group was small.  相似文献

Discs of tissue excised from ripe nonflue-cured and flue-cured tobacco leaves, harvested on six dates and cultured on three selective media, yielded 21 and 24 genera of fungi, respectively. Of 5,094 fungi isolated from 3,240 pieces of nonfluecured leaf tissue, 89.5% comprised five genera, Alternaria, Cladosporium, Epicoccum, Trichoderma, and Nigrospora. Of 2,494 fungi isolated from 3,240 pieces of flue-cured leaf tissue, 70.9% were Alternaria, Cladosporium Epicoccum, Aspergillus, and Nigrospora. Flue-cured and nonflue-cured brown-spot lesions harvested at two locations yielded 12 and 14 genera of fungi, respectively. Alternaria, Penicillium, Phoma, and Stemphyllium comprised 91.5% of the 2,245 fungi isolated from noncured and 87.1% of the 1,118 fungi isolated from the cured lesions. The number and kinds of fungi obtained from diseased and healthy tissue were reduced but not eliminated by flue-curing.  相似文献
