The ability of all 11 variable opacity (Opa) proteins encoded by Neisseria gonorrhoeae MS11 to interact directly with the five CD66 antigens was determined. Transfected HeLa cell lines expressing individual CD66 antigens were infected with recombinant N. gonorrhoeae and Escherichia coli strains expressing defined Opas. Based upon the ability of these bacteria to bind and invade and to isolate specifically CD66 antigens from detergent-soluble extracts of the corresponding cell lines, distinct specificity groups of Opa interaction with CD66 were seen. Defining these specificity groups allowed us to assign a specific function for CD66a in the Opa-mediated interaction of gonococci with two different target cell types, which are both known to co-express multiple CD66 antigens. The competence of individual Opas to interact with CD66a was strictly correlated with their ability to induce an oxidative response by polymorphonuclear neutrophils. The same Opa specificity was observed for the level of gonococcal binding to primary endothelial cells after stimulation with TNFα, which was shown to increase the expression of CD66a rather than CD66e. As CD66e alone is expressed on other target tissues of gonococcal pathogenicity, Opa variation probably contributes to the cell tropism displayed by gonococci.  相似文献

Deliberate killing for use as bait in a regional catfish (Calophysus macropterus) fishery is the primary threat affecting the survival of the Amazon river dolphin, or boto (Inia geoffrensis). Establishing and improving freshwater protected areas has been suggested as a possible course of action to protect the species. However, the ecology of the boto is poorly understood and more information is needed on the species’ habitat use and movement patterns to ensure that spatial protection initiatives meet conservation needs. In this study, mark‐recapture/resight data collected in and near the Mamirauá Sustainable Development Reserve, a várzea in the Brazilian Amazon, was used to examine transition probabilities of botos between various habitat types. Our findings suggest that it is imperative that spatial protection initiatives focused on the boto take into account the importance of várzea habitat and bay systems for individuals in early life stages. Moreover, because botos frequently occupy the main rivers, particularly during the low water period, protecting areas of the main river waters adjacent to várzeas could aid in the protection of hunted populations.  相似文献

To compare the responses of asthmatic and normal subjects to high effective doses of ozone, nine asthmatic and nine normal subjects underwent two randomly assigned 2-h exposures to filtered, purified air and 0.4 ppm ozone with alternating 15-min periods of rest and exercise on a cycle ergometer (minute ventilation = 30 l.min-1.m-2). Before and after each exposure, pulmonary function and bronchial responsiveness to methacholine were measured and symptoms were recorded. Ozone exposure was associated with a statistically significant decrease in forced vital capacity (FVC), forced expired volume in 1 s (FEV1), percent FEV1 (FEV1%), and forced expired flow at 25-75% FVC (FEF25-75) in both normal and asthmatic subjects. However, comparing the response of asthmatic and normal subjects to ozone revealed a significantly greater percent decrease in FEV1, FEV1%, and FEF25-75 in the asthmatic subjects. The effect of ozone on FVC and symptom scores did not differ between the two groups. In both normal and asthmatic subjects, exposure to ozone was accompanied by a significant increase in bronchial responsiveness. We conclude that exposure to a high effective ozone dose produces 1) increased bronchial responsiveness in both normal and asthmatic subjects, 2) greater airways obstruction in asthmatic than in normal subjects, and 3) similar symptoms and changes in lung volumes in the two groups.  相似文献

Viral RNA and proteins in chicken embryo fibroblasts infected with different cloned variants of the Prague strain Rous sarcoma virus (RSV) were analyzed. The ratio of immunoprecipitated pp60src to the gag gene product p27 in Prague A (PrA) and Prague B (PrB) RSV-infected cells was two to three times that in Prague C (PrC) RSV-infected cells. A significant increase in the steady-state ratio of spliced 2.7-kilobase src gene mRNA to unspliced 9.3-kilobase genome-size RNA was observed in PrA- and PrB- compared with PrC-infected cells, consistent with the differences in the ratios of the gag to src gene protein products. Similar results were obtained when hybrid-selected RNA, which had been labeled for 3 h with [3H]uridine, was analyzed on formaldehyde-agarose gels, suggesting that the observed differences were due to splicing rather than RNA stability. Recombinant plasmids from infectious molecular clones of PrA and PrC were constructed to localize the regions responsible for the effects on src gene splicing. The substitution in place of the corresponding PrA region of the 262-base-pair region between the env gene and the src gene coding sequences from the PrC clone into the infectious PrA plasmid conferred the low src splicing efficiency of the PrC strain. The nucleotide sequence of this region of the PrA plasmid was determined and compared with the sequence of the PrC strain. Only four nucleotide differences were found; two changes were within the intron sequence, and two were in the exon sequence. The possible role of these differences in determining the extent of viral RNA splicing is discussed.  相似文献

Circadian rhythmic changes of cell production were found atthe end of a test cycle (LD 14 : 10 h) and the synthetic tendenciesduring the test cycle were examined. The rates but not the durationof syntheses of chlorophyll, DNA and soluble protein rhythmicallychanged with additional waiting time in the dark after a growthcycle (LD 14 : 10). Changes in the rates of RNA and total proteinsynthesis were apparent only during the first 9 h in the lightphase of the test cycle. However, the rate of carbohydrate synthesisduring the test cycle was completely different; an exponentialpattern was observed in cells with 0 and 24 h waiting times,but a sigmoid one in cells with 12 h waiting time. Nuclear divisions(into 16 or even more) followed DNA synthesis. Summary The blue-green alga, Anacystis nidulans was successfully synchronized in continuous light (15,000 lux) by a 14 hr cycle, consisting of 8 hrs at 26°C, and 6 hrs at 32°C, coupled with periodic dilution of the algal suspension to a constant cell number at the end of each cycle. The alga continues to grow at the lower temperature, and a division burst begins 2 hrs after transfer to the higher temperature.

Die Cyclen bestehen aus 8 Std bei 26° C und 6 Std bei 32° C, an ihrem Ende wurde jeweils mit frischer Nährlösung auf die Ausgangszellzahl von 3·107 Zellen/ml verdünnt. Bei beiden Temperaturen kann sich die Alge vermehren, unter den synchronisierenden Bedingungen des Temperaturwechsels beginnt der Teilungsschub (Verdopplung der Zellzahl) bei 32° C nach 2 Std.

The susceptibility of worker honey bees,Apis mellifera L., as a function of age, to infestation by tracheal mites,Acarapis woodi Rennie, was investigated. Bees <24 h old were infested most frequently, and the frequency of infestation declined precipitously thereafter. Bees >4 days old were rarely infested in colonies during active brood rearing. Only 2 of 255 bees >8 days old, and 1 of 246 bees >16 days old, became infested. Most of the eggs found in bees>3 weeks old apparently were produced by the progeny of the original infestation.  相似文献

The relationship between body weight and natural mortality in juvenile and adult fish was analysed for different aquatic ecosystems: lakes, rivers, the ocean, and pond, cage and tank aquaculture systems. Mortality was modelled as a power function of weight, and the parameters b (exponent) and Mu (mortality at the unit weight of 1 g) estimated for fish in the six ecosystems, as well as within selected populations, species and families. At the ecosystem level, no significant differences in parameters were found between lakes, rivers and the ocean and a joint mortality-weight relationship for all natural ecosystems was estimated with parameters b=?0.288 (90% CL[?0.315, ?0.261]) and Mu=3.00 (90% CL[2.70, 3.30]) year?1. Among the culture systems, mortality-weight relationships in ponds and cages were not significantly different and a joint relationship was estimated. The weight exponents of mortality in ponds/cages and tanks were very similar at about b=?0.43, and significantly more negative than in natural ecosystems. Mortalities at unit weight were significantly lower in tanks (0.91 year?1) than in ponds/cages (2.24 year?1), and both were significantly lower than in natural ecosystems. No systematic differences were found between the mortality-weight relationships determined for individual populations, species or families, and fish in the respective ecosystems. It is hypothesized that aquaculture mortality-weight relationships indicate the allometric scaling of non-predation mortality, which is therefore more strongly size dependent than predation mortality. If non predation mortality in natural ecosystems shows a similar scaling with body weight, then the allometric exponent of predation mortality must be less negative than that observed for total natural mortality. Implications of the established mortality-weight relationships for aquaculture and culture-based fisheries are discussed.  相似文献
