The ecology of macroinvertebrate communities in arid regions is still poorly understood. Here we examined how the community structure varied at spatial and temporal scales in streams and tributaries of the Huasco River in semi-arid region of Northern Chile. We expected that macroinvertebrate distribution may be responding to natural processes of mineralization described for Chilean semiarid basins. The relationships among biotic and abiotic variables were assessed through multivariate techniques (principal component analysis, non-metric multidimensional scaling, canonical correspondence analysis), and a two-way analysis of similarity was used to evaluate differences between basins and years (2007, 2008, and 2009). Significant differences in community structure and physical–chemical variables between basins (Del Carmen and Del Tránsito) were found, but not between years. Altitude, Mn, Al, Ca, Na, HCO3, and dissolved oxygen were the variables that best accounted for the communities distribution. In particular, high metals concentration in El Transito basin should determine low density and diversity of macroinvertebrates. Chironomidae, Ephydridae, and Glossiphoniidae were associated to waters with high metals content and acidic pH, whereas Baetidae, Hydroptilidae, and Blephariceridae were associated to sites with more favorable physical–chemical conditions. These results contribute to understand the ecological patterns of macroinvertebrates in arid regions and should lead to conservation and monitoring plans for this remote place.  相似文献

Niche variation owing to individual differences in ecology has been hypothesized to be an early stage of sympatric speciation. Yet to date, no study has tracked niche width over more than a few generations. In this study, we show the presence of isotopic niche variation over millennial timescales and investigate the evolutionary outcomes. Isotopic ratios were measured from tissue samples of sympatric killer whale Orcinus orca lineages from the North Sea, spanning over 10 000 years. Isotopic ratios spanned a range similar to the difference in isotopic values of two known prey items, herring Clupea harengus and harbour seal Phoca vitulina. Two proxies of the stage of speciation, lineage sorting of mitogenomes and genotypic clustering, were both weak to intermediate indicating that speciation has made little progress. Thus, our study confirms that even with the necessary ecological conditions, i.e. among-individual variation in ecology, it is difficult for sympatric speciation to progress in the face of gene flow. In contrast to some theoretical models, our empirical results suggest that sympatric speciation driven by among-individual differences in ecological niche is a slow process and may not reach completion. We argue that sympatric speciation is constrained in this system owing to the plastic nature of the behavioural traits under selection when hunting either mammals or fish.  相似文献

Methylation of histone arginine residues is an epigenetic mark related to gene expression that is implicated in a variety of biological processes and can be reversed by small-molecule modulators of protein arginine methyltransferases (PRMTs). A series of symmetrical ureas, designed as analogues of the known PRMT1 inhibitor AMI-1 have been synthesized using Pd-catalyzed Ar–N amide bond formation processes or carbonylation reactions as key steps. Their inhibitory profile has been characterized. The enzymatic assays showed a weak effect on PRMT1 and PRMT5 activity for most of the compounds. The acyclic urea that exhibited the strongest effect on the inhibition of the PRMT1 activity also showed the greatest effect on the expression of some androgen receptor target genes (TMPRSS2 and FKBP5), which may be related with its enzymatic activity. Surprisingly, AMI-1 behaved as an activator of PRMT5 activity, a result not reported so far.  相似文献

In this study we investigated the hypothesis that a high-salt diet to hyperinsulinemic rats might impair antioxidant defense owing to its involvement in the activation of sodium reabsorption to lead to higher oxidative stress. Rats were fed a standard (CON), a high-salt (HS), or a high-fructose (HF) diet for 10 weeks after which, 50% of the animals belonging to the HF group were switched to a regimen of high-fructose and high-salt diet (HFS) for 10 more weeks, while the other groups were fed with their respective diets. Animals were then euthanized and their blood and liver were examined. Fasting plasma glucose was found to be significantly higher (approximately 50%) in fructose-fed rats than in the control and HS rats, whereas fat liver also differed in these animals, producing steatosis. Feeding fructose-fed rats with the high-salt diet triggered hyperinsulinemia and lowered insulin sensitivity, which led to increased levels of serum sodium compared to the HS group. This resulted in membrane perturbation, which in the presence of steatosis potentially enhanced hepatic lipid peroxidation, thereby decreasing the level of antioxidant defenses, as shown by GSH/GSSG ratio (HFS rats, 7.098±2.1 versus CON rats, 13.2±6.1) and superoxide dismutase (HFS rats, 2.1±0.05 versus CON rats, 2.3±0.1%), and catalase (HFS rats, 526.6±88.6 versus CON rats, 745.8±228.7 U/mg ptn) activities. Our results indicate that consumption of a salt-rich diet by insulin-resistant rats may lead to regulation of sodium reabsorption, worsening hepatic lipid peroxidation associated with impaired antioxidant defenses.  相似文献

Genotype effects on lamb carcass traits were investigated in a 4-year study aimed at assessing potential benefits from introducing meat breeds into the wool-oriented extensive sheep systems of northeastern Patagonia, Argentina. Five ram [Corriedale: CO; Border Leicester: BL; Île de France: IF; Texel: TX; and synthetic CRIII (25% Merino, 37.5% IF, 37.5% TX)] and 5 dam (CO; synthetic CRIII; BLCO: BL × CO; IFCO: IF × CO; and TXCO: TX × CO) genotypes were represented in the study. Data were collected from 436 male lambs of 9 genotypes (CO × CO, BL × CO, IF × CO, TX × CO, CRIII × CO, CRIII × BLCO, CRIII × IFCO, CRIII × TXCO, and CRIII × CRIII). Hot carcass weights and dressing yields were determined after slaughtering. Carcasses were given conformation and subcutaneous fat scores using the EUROP system [scale varying from E (best) to P (poorest) for conformation, and from 1 (lean) to 5 (overfat) for subcutaneous fat]. Linear measurements of carcass length and width were recorded and carcass compactness indices were calculated from those. Purebred CO acted as a standard for comparisons. On a constant liveweight basis, genotypes CRIII × IFCO and CRIII × CRIII presented higher (P < 0.05) carcass weight and dressing yield than CO × CO and BL × CO. Crossbred and synthetic genotypes showed higher (P < 0.05) carcass width than CO × CO. With the exception of BL × CO the remaining genotypes showed higher (P < 0.05) carcass width/length ratio than CO × CO. The probability that carcasses of crossbred and synthetic lambs presented better conformation than CO × CO was higher than 84%. Carcasses of CRIII × IFCO lambs were given the best conformation scores. The probability that BL × CO carcasses presented higher subcutaneous fat than the remaining genotypes exceeded 79%. Our results indicate significant improvements in carcass conformation arising from crossing. Sheep farmers in extensive systems could take advantage of the higher fatness of BL crossbred lambs to produce light carcasses with adequate fat cover, a crucial industry requirement. Terminal crossbreeding with Île de France, Texel, and CRIII rams could be implemented to improve carcass conformation thus matching market demand for heavy carcasses with limited fat content. Second cross schemes did not improve carcass commercial traits over the best terminal cross or the synthetic CRIII breed.  相似文献

High‐throughput platforms for microalgae screening are not yet commercially available. In this study, the feasibility of 96‐well microplates was analyzed for microalgae research. Equivalence among wells, as culture microreactors, was investigated in controlled high CO2 conditions. Specific growth rates of two microalgae species, Scenedesmus sp. UTEX1589 and an environmental isolate, were significantly higher in border wells than in internal positions. Furthermore, growth rate gradients analyzed as contours throughout the platform were observed for Scenedesmus sp. However, the output variable exhibited high precision associated with a low coefficient of variation (CV), between 6.8 and 7.8%. In a demonstrative experiment to determine the effect of culture media dilution on six microalgae species, treatments were randomized in the central subset of a microplate. Results were consistent and statistically sound (CV 9.4–12.9%), and showed that microalgae species could grow with no detrimental effect in 50% (v/v) dilution of the culture medium. Provided border wells exclusion and a randomized design, 96‐well microplates are a practical and statistical robust platform for microalgae research. © 2013 American Institute of Chemical Engineers Biotechnol. Large amount of drilling waste associated with the expansion of the Orinoco Oil Belt (OOB), the biggest proven reserve of extra-heavy crude oil (EHCO) worldwide, is usually impregnated with EHCO and highly salinized water-based drilling fluids. Oxidative exoenzymes (OE) of the lignin-degrading enzyme system (LDS) of fungi catalyse the oxidation of a wide range of toxic pollutants. However, very little evidences on fungal degradation or biotransformation of EHCO have been reported, which contain high amounts of asphaltenes and its biodegradation rate is very limited. The aims of this work were to study the ability of Pestalotiopsis palmarum BM-04 to synthesize OE, its potential to biotransform EHCO and to survive in extreme environmental conditions. Enzymatic studies of the LDS showed the ability of this fungus to overproduce high amounts of laccase (LACp) in presence of wheat bran or lignin peroxidase (LIPp) with EHCO as sole carbon and energy source (1300 U mgP−1 in both cases). FT-IR spectroscopy with Attenuated Total Reflectance (ATR) analysis showed the enzymatic oxidation of carbon and sulfur atoms in both maltenes and asphaltenes fractions of biotreated EHCO catalysed by cell-free laccase-enriched OE using wheat bran as inducer. UV-visible spectrophotometry analysis revealed the oxidation of the petroporphyrins in the asphaltenes fraction of biotreated EHCO. Tolerance assays showed the ability of this fungus to grow up to 50 000 p.p.m. of EHCO and 2000 mM of NaCl. These results suggest that P. palmarum BM-04 is a hopeful alternative to be used in remediation processes in extreme environmental conditions of salinity and EHCO contamination, such as the drilling waste from the OOB.  相似文献

The C2 domain of PKCα possesses two different binding sites, one for Ca2+ and phosphatidylserine and a second one that binds PIP2 with very high affinity. The enzymatic activity of PKCα was studied by activating it with large unilamellar lipid vesicles, varying the concentration of Ca2+ and the contents of dioleylglycerol (DOG), phosphatidylinositol 4,5-bisphosphate (PIP2) and phosphadidylserine (POPS) in these model membranes. The results showed that PIP2 increased the Vmax of PKCα and, when the PIP2 concentration was 5 mol% of the total lipid in the membrane, the addition of 2 mol% of DOG did not increase the activity. In addition PIP2 decreases K0.5 of Ca2+ more than 3-fold, that of DOG almost 5-fold and that of POPS by a half. The K0.5 values of PIP2 amounted to only 0.11 µM in the presence of DOG and 0.39 in its absence, which is within the expected physiological range for the inner monolayer of a mammalian plasma membrane. As a consequence, PKCα may be expected to operate near its maximum capacity even in the absence of a cell signal producing diacylglycerol. Nevertheless, we have shown that the presence of DOG may also help, since the K0.5 for PIP2 notably decreases in its presence. Taken together, these results underline the great importance of PIP2 in the activation of PKCα and demonstrate that in its presence, the most important cell signal for triggering the activity of this enzyme is the increase in the concentration of cytoplasmic Ca2+.  相似文献

Trypanosoma cruzi is the etiological agent of Chagas disease, an illness that affects about 10 million people, mostly in South America, for which there is no effective treatment or vaccine. In this context, transgenic parasites expressing reporter genes are interesting tools for investigating parasite biology and host-parasite interactions, with a view to developing new strategies for disease prevention and treatment. We describe here the construction of a stably transfected fluorescent T. cruzi clone in which the GFP gene is integrated into the chromosome carrying the ribosomal cistron in T. cruzi Dm28c. This fluorescent T. cruzi produces detectable amounts of GFP only at replicative stages (epimastigote and amastigote), consistent with the larger amounts of GFP mRNA detected in these forms than in the non replicative trypomastigote stages. The fluorescence signal was also strongly correlated with the total number of parasites in T. cruzi cultures, providing a simple and rapid means of determining the growth inhibitory dose of anti-T.cruzi drugs in epimastigotes, by fluorometric microplate screening, and in amastigotes, by the flow cytometric quantification of T. cruzi-infected Vero cells. This fluorescent T. cruzi clone is, thus, an interesting tool for unbiased detection of the proliferating stages of the parasite, with multiple applications in the genetic analysis of T. cruzi, including analyses of host-parasite interactions, gene expression regulation and drug development.  相似文献
