Summary The nucleotide sequence has been determined of a 1400 by fragment from the chromosome of Yersinia enterocolitica containing the gene for -lactamase I. An ORF of 882 by was identified, which could code for a polypeptide of 294 amino acids, closely related to other -lactamases of molecular class A. Amino acids 1–30 could constitute a signal peptide. The mature protein would be 264 amino acids long with a calculated pI of 6.2. Alignment of the amino acid sequence of the class A -lactamases suggested the existence of two subgroups in the same class, and this is discussed in the context of the evolution of the enzymes.This sequence will appear in the EMBL Data Library under the accession number X57074 相似文献

Six mutants lacking the glycolytic enzyme fructose 1,6-bisphosphate aldolase have been isolated in the yeast Saccharomyces cerevisiae by inositol starvation. The mutants grown on gluconeogenic substrates, such as glycerol or alcohol, and show growth inhibition by glucose and related sugars. The mutations are recessive, segregate as one gene in crosses, and fall in a single complementation group. All of the mutants synthesize an antigen cross-reacting to the antibody raised against yeast aldolase. The aldolase activity in various mutant alleles measured as fructose 1,6-bisphosphate cleavage is between 1 to 2% and as condensation of triose phosphates to fructose 1,6-bisphosphate is 2 to 5% that of the wild-type. The mutants accumulate fructose 1,6-bisphosphate from glucose during glycolysis and dihydroxyacetone phosphate during gluconeogenesis. This suggests that the aldolase activity is absent in vivo. 相似文献

Aim To examine current biogeographical patterns of Aphodiinae dung beetles (Coleoptera: Scarabaeoidea: Scarabaeidae) in order to reveal relationships among regions and their potential impact on the diversification of this group.
Location Worldwide.
Methods Information about all Aphodiinae genera was obtained from the literature. An occurrence matrix was built for the six worldwide biogeographical regions, and their faunas were characterized through simple statistics. Regional variations and similarities were further explored using co-occurrence and nestedness analyses, sequential agglomerative, hierarchical and nested clustering (SAHN), and a parsimony analysis of endemicity (PAE). Mantel tests were also employed to assess the relationships between several characteristics of the regions and their faunas.
Results The Palaearctic and Palaeotropical regions showed the highest total numbers of Aphodiinae genera and the greatest generic endemism. Both these regions and the Oriental also showed higher numbers of genera than would be expected according to their size. Co-occurrence and nestedness analyses confirmed the non-randomness of the distribution of genera. Clustering and PAE showed that the Palaearctic and Oriental regions are the most similar, followed by the Palaeotropical region. Regional dissimilarity in genera composition was related to biological and historical traits, but not to ecoregions.
Main conclusions A structured geographical pattern for Aphodiinae was confirmed. Land continuity and proximity in the long term could have played a unifying role in regional faunas. We suggest that the different biogeographical regions have acted as either macroevolutionary sources (basically the Palaearctic and the Palaeotropical regions) or sink regions, according to their role as diversification centres. Morphology can be adaptive through its effect on performance of an organism. The effect of performance may, however, be modulated by behavior; an organism may choose a behavioral option that does not fully utilize its maximum performance. Behavior may therefore be decoupled from morphology and performance. To gain insight into the relationships between these levels of organization, we combined morphological data on defensive structures with measures of defensive performance, and their utilization in defensive behavior. Scorpion species show significant variation in the morphology and performance of their main defensive structures; their chelae (pincers) and the metasoma (“tail”) carrying the stinger. Our data show that size-corrected pinch force varies to almost two orders of magnitude among species, and is correlated with chela morphology. Chela and metasoma morphology are also correlated to the LD50 of the venom, corroborating the anecdotal rule that dangerously venomous scorpions can be recognized by their chelae and metasoma. Analyses of phylogenetic independent contrasts show that correlations between several aspects of chela and metasoma morphology, performance and behavior are present. These correlations suggest co-evolution of behavior with morphology and performance. Path analysis found a performance variable (pinch force) to partially mediate the relationship between morphology (chela aspect ratio) and behavior (defensive stinger usage). We also found a correlation between two aspects of morphology: pincer finger length correlates with the relative “thickness” (aspect ratio) of the metasoma. This suggests scorpions show a trade-off between their two main weapon complexes: the metasoma carrying the stinger, and the pedipalps carrying the chelae. 相似文献

Changes in the total phenolics, condensed tannins (CT), protein-precipitable phenolics content and protein precipitation capacity
were determined on mangrove leaves representing a range of maturation and senescence in the Jiulong River Estuary, Fujian,
China. The results showed that the total phenolics, extractable condensed tannins, total condensed tannins, protein-precipitable
phenolics content and protein precipitation capacity in young leaves were higher than those in mature and senescent leaves.
With leaf maturation and senescence, total pehnolics decreased. The rapid loss of phenolics observed following senescence
and abscission can be ascribed to leaching and degradation. Protein-bound CT and fibre-bound CT tended to increase with leaf
maturity, and decreased with senescence. Protein-bound CT and fibre-bound CT increased with leaf decomposition, with CT binding
more strongly to protein than to fibre. The increases in nitrogen contents and ash free caloric values, and declines in total
phenolics and total condensed tannins of mangrove leaf litter suggests that partially decomposed mangrove detritus is a palatable
heterotrophic substrate, and thus may be an important source of matter and energy for the estuarine food web.

Refractoriness to existing medications of up to 80 % of the patients with mesial temporal lobe epilepsy (MTLE) prompts for finding new antiepileptic drug targets. The adenosine A2A receptor emerges as an interesting pharmacological target since its excitatory nature partially counteracts the dominant antiepileptic role of endogenous adenosine acting via inhibitory A1 receptors. Gain of function of the excitatory A2A receptor has been implicated in a significant number of brain pathologies commonly characterized by neuronal excitotoxicity. Here, we investigated changes in the expression and cellular localization of the A2A receptor and of the adenosine-generating enzyme, ecto-5′-nucleotidase/CD73, in the hippocampus of control individuals and MTLE human patients. Western blot analysis indicates that the A2A receptor is more abundant in the hippocampus of MTLE patients compared to control individuals. Immunoreactivity against the A2A receptor predominates in astrocytes staining positively for the glial fibrillary acidic protein (GFAP). No co-localization was observed between the A2A receptor and neuronal cell markers, like synaptotagmin 1/2 (nerve terminals) and neurofilament 200 (axon fibers). Hippocampal astrogliosis observed in MTLE patients was accompanied by a proportionate increase in A2A receptor and ecto-5′-nucleotidase/CD73 immunoreactivities. Given our data, we hypothesize that selective blockade of excessive activation of astrocytic A2A receptors and/or inhibition of surplus adenosine formation by membrane-bound ecto-5′-nucleotidase/CD73 may reduce neuronal excitability, thus providing a novel therapeutic target for drug-refractory seizures in MTLE patients. 相似文献

Microarray-based comparative genomic hybridizations (CGH) interrogate genomic DNA to identify structural differences such as amplifications and deletions that are easily detected as large signal aberrations. Subtle signal deviations caused by single nucleotide polymorphisms (SNPs) can also be detected but is challenged by a high AT content (81%) in P. falciparum. We compared genome-wide CGH signal to sequence polymorphisms between parasite strains 3D7, HB3, and Dd2 using NimbleGen microarrays. From 23,191 SNPs (excluding var/rif/stevor genes), our CGH probe set detected SNPs with > 99.9% specificity but low (< 10%) sensitivity. Probe length, melting temperature, GC content, SNP location in the probe, mutation type, and hairpin structures affected SNP sensitivity. Previously unrecognized variable number tandem repeats (VNTRs) also were detected by this method. These findings will guide the redesign of a probe set to optimize an openly available CGH microarray platform for high-resolution genotyping suitable for population genomics studies. 相似文献

For many applications the continuous prediction afforded by species distribution modeling must be converted to a map of presence or absence, so a threshold probability indicative of species presence must be fixed. Because of the bias in probability outputs due to frequency of presences (prevalence), a fixed threshold value, such as 0.5, does not usually correspond to the threshold above which the species is more likely to be present. In this paper four threshold criteria are compared for a wide range of sample sizes and prevalences, modeling a virtual species in order to avoid the omnipresent error sources that the use of real species data implies. In general, sensitivity–specificity difference minimizer and sensitivity–specificity sum maximizer criteria produced the most accurate predictions. The widely-used 0.5 fixed threshold and Kappa-maximizer criteria are the worst ones in almost all situations.
