Chitotriosidase protein (ChT) is the most important biochemical marker described for Gaucher disease (GD). ChT activity is increased several hundred-fold in plasma of GD patients and shows a strong positive correlation with the severity of the disease. However, a recessively inherited enzyme deficiency, with an incidence of about 6% in the Caucasian population, means that not all patients with GD can be monitored by measuring ChT activity. Applying two-dimensional gel electrophoresis (2-DE) technology this study describes the localization and identification of five ChT isoforms in 2-DE images obtained from plasma of GD patients. All these isoforms were unequivocally identified using MALDI-TOF mass spectrometry (MS) and validated by western blot analysis. The features of each ChT isoform separated by 2-DE in plasma from GD patients homozygous for the wild-type ChT allele, carriers of one defective allele and patients homozygous for the mutant allele are presented. We also show the correlation between each ChT isoform and the plasma ChT enzymatic activity of the GD patients sampled in this study.  相似文献

Calving date in many mammals is matched to the time of greatest food availability. Out of season calving results in heavy penalties in terms of own and offspring survival or body condition. This study examined whether gestation length is affected by advancing fertilisation. Thirty-six red deer hinds (of the Iberian and Scottish subspecies) were subjected to a synchronisation treatment of oestrus, ovulation, and artificial insemination on three dates, with remaining non-pregnant females mated with an intact male in a last group. Gestation was longer the more the fertilisation was advanced; gestation lasted 241.5+/-1.3 days (d) in the first group, 237.4+/-1.2 d in the second, 235.1+/-1.3 d in the third, and 231.2+/-1.6 d in the last. Mean gestation lasted 234.2+/-0.7 d. Hinds gained less weight during gestation the more the fertilisation was advanced. The difference was due at least in part to net body weight of the hind after calving compared to that at mating, and calves did not differ in birth weight. As early born calves suffer greater mortality in the field, this enlargening of gestation might be a compensatory response of the hinds to match calving with food availability. Under natural conditions, similar small modifications of gestation length may help hinds to overcome short-term adverse conditions for calving. Because calf mortality is correlated with birth weight, hinds may have kept calf birth weight constant at the expense of greater body weight loss.  相似文献

A strategy for the preparation of enantiomerically pure (R)- and (S)-alpha-methyldiphenylalanine, constrained phenylalanine analogs, is described. A racemic precursor was prepared in high yield from easily available starting products and subjected to HPLC resolution on a noncommercial chiral stationary phase. More than 600 mg of each enantiomer was isolated in optically pure form by using a 150 x 20 mm ID column containing mixed 10-undecenoate/3,5-dimethylphenylcarbamate of cellulose covalently bonded to allylsilica gel and a mixture of n-hexane/2-propanol/acetone as the mobile phase.  相似文献

Endophytic fungi inhabit living plant tissues without causing disease symptoms. Although abundant, the extent of their contribution to fungal biodiversity remains unclear. Since endophytic fungi are poorly known, especially in the tropics, current estimates of fungal species are probably conservative. Here we tested strategies for sampling endophytic fungi in tropical plants. We compared the number of fungi isolated from 400 mm2 leaf pieces that were divided into increasingly small fragments. Leaf pieces were surface-sterilized, cut into fragments and plated on culture media. For a given area, cutting leaf pieces into smaller fragments significantly increased the number of fungal morphospecies recovered. There was a strong linear relationship between size of fragments and number of fungi isolated. By extrapolation, an estimated 16 ± 3 fungi could be recovered from a 2 × 2 cm leaf piece, using infinitely small fragments. This represents a large part of the fungal diversity estimated to exist in leaf endophytes in a population. We conclude that reducing the size and increasing the number of leaf fragments will increase the number of fungal species isolated. Objectives and results

Mesenchymal stromal cells (MSCs) are potential targets for cell and gene therapy-based approaches against a variety of different diseases. The MSCs from bone marrow are a promising target population as they are capable of differentiating along multiple lineages and have significant expansion capability. These characteristics make them strong candidates for delivering genes and restoring organ systems function. However, as other primary cells, MSCs are difficult to transfect. In order to standardize a simple protocol for transfection of MSCs, we conducted a series of experiments and achieved a protocol that does not require the use of viral particles or specific expensive equipment.

Conclusion

MSCs transfection at early passages using a ratio lipid/DNA of 3.0 µL/µg with Lipofectamine 3000® yields good transfection efficiencies for human MSCs (up to 26%) and is rapid, simple, and safe.

G protein-coupled receptor (GPCR) kinases (GRKs) regulate the sensitivity of GPCRs, including dopamine receptors. The GRK4 locus is linked to, and some of its polymorphisms are associated with, human essential hypertension. Transgenic mice overexpressing human (h) GRK4gamma A142V on a mixed genetic background (C57BL/6J and SJL/J) have impaired renal D(1)-dopamine receptor (D(1)R) function and increased blood pressure. We now report that hGRK4gamma A142V transgenic mice, in C57BL/6J background, are hypertensive and have higher blood pressures than hGRK4gamma wild-type transgenic and nontransgenic mice. The hypertensive phenotype is stable because blood pressures in transgenic founders and F6 offspring are similarly increased. To determine whether the hypertension is associated with increased production of reactive oxygen species (ROS), we measured renal NADPH oxidase (Nox2 and Nox4) and heme oxygenase (HO-1 and HO-2) protein expressions and urinary excretion of 8-isoprostane and compared the effect of Tempol on blood pressure in hGRK4gamma A142V transgenic mice and D(5)R knockout (D(5)(-/-)) mice in which hypertension is mediated by increased ROS. The expressions of Nox isoforms and HO-2 and the urinary excretion of 8-isoprostane were similar in hGRK4gamma A142V transgenic mice and their controls. HO-1 expression was increased in hGRK4gamma A142V relative to hGRK4gamma wild-type transgenic mice. In contrast with the hypotensive effect of Tempol in D(5)(-/-) mice, it had no effect in hGRK4gamma A142V transgenic mice. We conclude that the elevated blood pressure of hGRK4gamma A142V transgenic mice is due mainly to the effect of hGRK4gamma A142V transgene acting via D(1)R and increased ROS production is not a contributor.  相似文献

The cell division cycle 25A (CDC25A) phosphatase is a key regulator of cell cycle progression that acts on the phosphorylation status of Cyclin–Cyclin-dependent kinase complexes, with an emergent role in the DNA damage response and cell survival control. The regulation of CDC25A activity and its protein level is essential to control the cell cycle and maintain genomic integrity. Here we describe a novel ubiquitin/proteasome-mediated pathway negatively regulating CDC25A stability, dependent on its phosphorylation by the serine/threonine kinase DYRK2. DYRK2 phosphorylates CDC25A on at least 7 residues, resulting in its degradation independent of the known CDC25A E3 ubiquitin ligases. CDC25A in turn is able to control the phosphorylation of DYRK2 at several residues outside from its activation loop, thus affecting DYRK2 localization and activity. An inverse correlation between DYRK2 and CDC25A protein amounts was observed during cell cycle progression and in response to DNA damage, with CDC25A accumulation responding to the manipulation of DYRK2 levels or activity in either physiological scenario. Functional data show that the pro-survival activity of CDC25A and the pro-apoptotic activity of DYRK2 could be partly explained by the mutual regulation between both proteins. Moreover, DYRK2 modulation of CDC25A expression and/or activity contributes to the DYRK2 role in cell cycle regulation. Altogether, we provide evidence suggesting that DYRK2 and CDC25A mutually control their activity and stability by a feedback regulatory loop, with a relevant effect on the genotoxic stress pathway, apoptosis, and cell cycle regulation.Subject terms: Proteins, Cell biology, Proteomics  相似文献

In order to generalize the recently described method for the insolubilization of polysaccharide derivatives to benzoates of cellulose, five mixed 10-undecenoate/benzoates of this polysaccharide have been prepared and linked to allyl silica gel by means of a radical reaction. The chiral recognition ability of the resulting materials when used as high-performance liquid chromatographic chiral stationary phases was evaluated using heptane/2-propanol and heptane/chloroform mixtures as mobile phases.
