Somatostatin receptors in malignant tissues   总被引:3,自引:0,他引:3  
High affinity somatostatin receptors (SS-R) have been identified in membrane homogenates or tissue sections from several hundred human tumors. SS-R were found in most tumors originating from SS target tissues, i.e. GH- and TSH-producing pituitary tumors, endocrine gastroenteropancreatic (GEP) tumors (including metastases) and brain tumors, including gliomas and neuroblastomas. SS-R were also expressed in several tumors originating from various other tissues, i.e. breast and small cell lung carcinomas, some colorectal cancers, and medullary tyroid carcinomas. In general, most of the SS-R+ tumors are well-differentiated and/or have neuroendocrine features. They often have low or absent epidermal growth factor receptor (EGF-R) expression. In some tumors (i.e. breast tumors) SS-R are not homogeneously distributed, making SS-R autoradiography a particularly useful tool for assessing SS-R status. SS-R are functional in pituitary and GEP tumors where they mediate hormone secretion inhibition. In these and in the other SS-R+ tumors, SS-R may also mediate antiproliferative effects of SS, as evidenced, in animals where growth of SS-R+ tumor xenografts is inhibited by SS analogs. For diagnosis, SS-R+ tumors and metastases can be localized in vivo by scanning techniques after 123I-labelled SS analog injection.  相似文献

We have applied a new equilibration procedure for the atomic level simulation of a hydrated lipid bilayer to hydrated bilayers of dioleyl-phosphatidylcholine (DOPC) and palmitoyl-oleyl phosphatidylcholine (POPC). The procedure consists of alternating molecular dynamics trajectory calculations in a constant surface tension and temperature ensemble with configurational bias Monte Carlo moves to different regions of the configuration space of the bilayer in a constant volume and temperature ensemble. The procedure is applied to bilayers of 128 molecules of POPC with 4628 water molecules, and 128 molecules of DOPC with 4825 water molecules. Progress toward equilibration is almost three times as fast in central processing unit (CPU) time compared with a purely molecular dynamics (MD) simulation. Equilibration is complete, as judged by the lack of energy drift in 200-ps runs of continuous MD. After the equilibrium state was reached, as determined by agreement between the simulation volume per lipid molecule with experiment, continuous MD was run in an ensemble in which the lateral area was restrained to fluctuate about a mean value and a pressure of 1 atm applied normal to the bilayer surface. Three separate continuous MD runs, 200 ps in duration each, separated by 10,000 CBMC steps, were carried out for each system. Properties of the systems were calculated and averaged over the three separate runs. Results of the simulations are presented and compared with experimental data and with other recent simulations of POPC and DOPC. Analysis of the hydration environment in the headgroups supports a mechanism by which unsaturation contributes to reduced transition temperatures. In this view, the relatively horizontal orientation of the unsaturated bond increases the area per lipid, resulting in increased water penetration between the headgroups. As a result the headgroup-headgroup interactions are attenuated and shielded, and this contributes to the lowered transition temperature.  相似文献

Background  

The Dmbx1 gene is important for the development of the midbrain and hindbrain, and mouse gene targeting experiments reveal that this gene is required for mediating postnatal and adult feeding behaviours. A single Dmbx1 gene exists in terrestrial vertebrate genomes, while teleost genomes have at least two paralogs. We compared the loss of function of the zebrafish dmbx1a and dmbx1b genes in order to gain insight into the molecular mechanism by which dmbx1 regulates neurogenesis, and to begin to understand why these duplicate genes have been retained in the zebrafish genome.  相似文献

Somatostatin analogs are used in the control of hormonal hypersecretion and tumor growth of patients with acromegaly, islet cell carcinomas and carcinoids. Recently we showed that somatostatin receptor positive tumors can be visualized in vivo after the administration of radioactive isotope-labelled somatostatin analogs. Receptor imaging was positive in 18/21 islet cell tumors, 30/31 carcinoids, 26/28 paragangliomas, 9/14 medullary thyroid carcinomas, 5/7 small cell lung cancers, 6/7 neuroblastomas, 38/49 primary breast cancers, and 0/18 pancreatic adenocarcinomas. Also 11/11 meningiomas, 4/4 astrocytomas and 0/3 glioblastomas could be visualized. Somatostatin receptor imaging is an easy, harmless and painless diagnostic method. It is an in vivo method for the recognition of neuroendocrine cancers. It localizes multiple and/or metastatic tumors, predicts the successful control of hormonal hypersecretion by octreotide and seems of prognostic value in certain types of cancer. This scintigraphic method might help in patient selection for clinical trials with somatostatin analogs in the treatment of neuroendocrine cancers.  相似文献

Estrogens prevent or diminish the sensitivity to dopamine of prolactin (PRL) secretion by cultured rat pituitary cells. Cultured tumor cells prepared from a transplantable rat PRL-secreting tumor were insensitive to dopamine and bromocriptine, while the anti-estrogen tamoxifen restored this sensitivity. Cultured normal human pituitary cells were shown to be more sensitive to dopamine, if they were preincubated with estradiol, while cultured human prolactinoma cells became insensitive to bromocriptine after they were exposed to estrogens. This sensitivity was restored, however, by tamoxifen. These results point to an important species difference between primates and rodents with regard to the normal regulation of PRL secretion.  相似文献

The estradiol positive feedback mechanism appears to become mature between days 10 and 20 after birth. Rising serum prolactin levels between day 20 after birth and puberty are correlated with high hypothalamic norepinephrine turnover. High prolactin levels stimulate hypothalamic dopamine (DA) turnover, which may actively inhibit hypothalamic luteinizing hormone-releasing hormone (LHRH) release. Hypothalamic DNA receptor sensitivity is high in 10- to 20-day-old rats and gradually decreases between day 20 after birth and puberty. The reason for this desensitization may be the high hypothalamic DA turnover. This may result in a less strong inhibition of LHRH release allowing the positive feedback action of estradiol to elicit the first preovulatory luteinizing hormone (LH) surge initiating puberty.  相似文献

Summary Plant breeding research on agricultural crops in the Netherlands is described. A generally high level of breeding can be expected, when there is close cooperation between free research in a university department, directed research in an institute (based on a program planned to produce practical results), and private plant breeders. The increasing activities and efficiency of private breeders call for more basic research and more help on a higher level. Therefore long term programs of the Foundation for Agricultural Plant Breeding (S.V.P.) are necessary to point out to all interested groups in which direction research may be expected to go in the coming period. For long term programs private breeders know in which parts of their work the S.V.P. is supposed to help them.The board and heads of the institute will have formed a better overall picture of the situation so that priority of the different problems can be estimated and flexibility in research work can be maintained. It is necessary that the research institute and private breeders continually be in contact to avoid misunderstanding, duplication and unwanted gaps. To realize this the research workers from the institute should regularly visit private breeders and vice versa.Furthermore working groups in each of the crops with which S.V.P. is working should meet at least once a year to exchange ideas and discuss the actual situation. The S.V.P. itself should not breed varieties but should leave this entirely to private breeders. In the Netherlands this system has proved to be very workable and satisfactory.The main tasks of this research work are: (1) Research in collections. — (2) Detection of important characters and their genetical analyses. — (3) Introduction of new characters. — (4) Building up parental lines for the benefit of private breeders. — (5) Research on breeding methods. — (6) Research on testing methods. — (7) Studies on the influence of environmental conditions.Consequently the type of specialized research workers has to be changed from crop experts to problem solvers. The plant material resulting from their research serves as a building stone in the program of the private breeders.The increasing number of specialized problems makes it impossible to do all the necessary fundamental research at a plant breeding institute. Cooperation with other institutes, specialized in the discipline involved, is very important. To maintain a high level of research activities it is suggested that strong cooperation between institutes in different countries should be aimed at.
