BmMNPV, a Nucleopolyhedrovirus isolated from infected Bombyx mori (L.) larvae in Paraná State--Brazil, was used to inoculate healthy 5th-instar B. mori larvae and examine the infection on central nervous system (CNS) cells. Samples of nervous tissue were removed from the infected insects, at different sampling times, and processed for cytopathology studies by light and transmission electron microscopy using routine techniques. The experiment included both inoculated and non-inoculated larvae (control). BmMNPV infection was detected on the 5th day after inoculation in CNS cells. Initially, infection was characterized by nuclear hypertrophy and the presence of virogenic stroma, in which the progeny virions were produced. Virions are enveloped and occluded into protein crystal, the polyhedra. Lyses of infected CNS cells were undetected; however, free mature polyhedra were seen in spaces inside the CNS. These polyhedra possibly came from trachea that penetrate the CNS and its cells, which are susceptible to BmMNPV and lyses after infection. We conclude that the tracheal system is responsible for disseminating BmMNPV infection in B. mori CNS and that the tracheal branches allow non-occluded virions to pass through the blood-brain barrier. 相似文献

This work reports on the characterization of a metalloendopeptidase kininase present in Boophilus microplus salivary glands. Using the guinea pig ileum assay, salivary gland whole extracts (SGE) were found to have a potent kininase activity. Ion-exchange chromatography separated two kininase activities from SGE. The major enzymatic component, eluted at lower ionic strength, was named BooKase (Boophilus Kininase). Analysis of the hydrolysis products by capillary electrophoresis identified Phe5-Ser6 as the only hydrolyzable peptide bond in bradykinin after BooKase treatment. This is the same specificity as the mammalian thimet oligoendopeptidase (EC 3.4.24.15). Like this enzyme, BooKase is also a metallo-peptidase (requires Mn2+) and is activated by-SH protecting reagents. In addition, BooKase was partially inhibited by cFP-AAF-pAB, a specific inhibitor of thimet oligopeptidase. Contrary to other kininases, BooKase had no activity upon angiontensin I. Nucleoside hydrolases cleave the N-glycosidic bond of ribonucleosides. Because of their vital role in the protozoan purine salvage pathway, nucleoside hydrolases from parasitic protozoa in particular have been studied extensively by X-ray crystallography, kinetic methods and site-directed mutagenesis. An elaborate network of conserved interactions between the metalloenzyme and the ribose enables steric and electrostatic stabilisation of the oxocarbenium-ion-like transition state. Activation of the leaving group by protonation before the formation of the transition state is a recurring catalytic strategy of enzymes that cleave N-glycosidic bonds. Ornamental colouration is often due to carotenoid pigments and varies inter- and intra-specifically. This paper reports on
variation in beak colour of the gentoo penguin, Pygoscelis papua, corresponding to different geographical locations along a latitudinal gradient in the Antarctic Peninsula (from King George
Island (62o15′S–58o37′W) to Rongé Island (64o40′S–62o40′W). The gentoo penguin has a conspicuous red spot on both sides of
the beak that indicates the presence of the carotenoid pigment, astaxanthin. Beak colouration was measured with a portable
spectrophotometer for 20 individuals in three locations, along the Western coast of the Antarctic Peninsula. In the study
area, marked variation can be found in terms of factors such us parasite load, human impact, variations in UV radiation and
the abundance of krill; all possibly affecting carotenoid availability for signalling purposes. Colour traits were expected
to be more intense, that is more vivid, saturated and pure, in places where there is diminished pressure from factors such
as contamination, parasites or diseases, all of which may reduce the availability of carotenoids for other functions, such
as antioxidant or immune stimulation involving physiological trade-offs. Likewise, colour traits might be predicted to be
more intense where carotenoid sources, krill in the case of gentoo penguins, are more available. However, contrary to this
initial expectation, our results indicate that northerly penguins’ populations, which are in the most polluted and parasitized
areas, have more saturated beaks. An alternative hypothesis suggests that environmental constraints relating to the variation
in abundance of krill may explain the geographical variation in colour expression found among gentoo penguins. 相似文献

Objective: This in vivo study evaluated the effect of a constant electromagnetic field (CEF) on bone healing around dental implants in dogs. Materials and methods: Eight dental implants were placed immediately after extraction of the first pre‐molar and molar teeth on the mandible of two male dogs and divided into experimental (CEF) and control groups. A CEF at magnetic intensity of 0.8 mT with a pulse width of 25 μs and frequency of 1.5 MHz was applied on the implants for 20 min per day for 2 weeks. Result and conclusion: After qualitative histological analysis, a small quantity of newly formed bone was observed in the gap between the implant surface and alveolar bone in both groups. 相似文献

Background: As the population ages, there is a greater demand for differentiated care from health professionals. However, the treatment needs of the elderly population are not sufficiently addressed, particularly in developing countries. Materials and methods: An official list of all dentists registered in the city of Belo Horizonte, Brazil, was obtained from the Regional Dentistry Council. A questionnaire was delivered to 276 randomly selected dentists; the survey contained questions designed to characterise the sample and identify dentists’ conduct towards elderly patients. A Likert scale was used to identify knowledge regarding care for the elderly and attitudes towards ageing. Results: Women demonstrated more positive attitudes towards the elderly. Graduates who worked in the public sector demonstrated a greater knowledge of geriatric dentistry (p < 0.05). Attending specific continuing education courses had no influence over the dentists’ willingness to treat the elderly population. Significant associations were not found between attitudes and knowledge regarding the elderly. Conclusion: Dentists who worked in the public sector demonstrated a greater knowledge regarding the elderly. This knowledge did not influence attitudes towards ageing. Women demonstrated a more positive attitude regarding ageing and dental care. 相似文献

Peanut genomics is very challenging due to its inherent problem of genetic architecture. Blockage of gene flow from diploid wild relatives to the tetraploid; cultivated peanut, recent polyploidization combined with self pollination, and the narrow genetic base of the primary genepool have resulted in low genetic diversity that has remained a major bottleneck for genetic improvement of peanut. Harnessing the rich source of wild relatives has been negligible due to differences in ploidy level as well as genetic drag and undesirable alleles for low yield. Lack of appropriate genomic resources has severely hampered molecular breeding activities, and this crop remains among the less-studied crops. The last five years, however, have witnessed accelerated development of genomic resources such as development of molecular markers, genetic and physical maps, generation of expressed sequenced tags (ESTs), development of mutant resources, and functional genomics platforms that facilitate the identification of QTLs and discovery of genes associated with tolerance/resistance to abiotic and biotic stresses and agronomic traits. Molecular breeding has been initiated for several traits for development of superior genotypes. The genome or at least gene space sequence is expected to be available in near future and this will further accelerate use of biotechnological approaches for peanut improvement. 相似文献

In the context of recombinant DNA technology, the development of feasible and high-yielding plasmid DNA production processes has regained attention as more evidence for its efficacy as vectors for gene therapy and DNA vaccination arise. When producing plasmid DNA in Escherichia coli, a number of biological restraints, triggered by plasmid maintenance and replication as well as culture conditions are responsible for limiting final biomass and product yields. This termed "metabolic burden" can also cause detrimental effects on plasmid stability and quality, since the cell machinery is no longer capable of maintaining an active metabolism towards plasmid synthesis and the stress responses elicited by plasmid maintenance can also cause increased plasmid instability. The optimization of plasmid DNA production bioprocesses is still hindered by the lack of information on the host metabolic responses as well as information on plasmid instability. Therefore, systematic and on-line approaches are required not only to characterise this "metabolic burden" and plasmid stability but also for the design of appropriate metabolic engineering and culture strategies. The monitoring tools described to date rapidly evolve from laborious, off-line and at-line monitoring to online monitoring, at a time-scale that enables researchers to solve these bioprocessing problems as they occur. This review highlights major E. coli biological alterations caused by plasmid maintenance and replication, possible causes for plasmid instability and discusses the ability of currently employed bioprocess monitoring techniques to provide information in order to circumvent metabolic burden and plasmid instability, pointing out the possible evolution of these methods towards online bioprocess monitoring. 相似文献

Disrupted circadian rhythms are associated with obesity and metabolic alterations, but little is known about the participation of peripheral circadian clock machinery in these processes. The aim of the present study was to analyze RNA expression of clock genes in subcutaneous (SAT) and visceral (VAT) adipose tissues of male and female subjects in AM (morning) and PM (afternoon) periods, and its interactions with body mass index (BMI). Ninety-one subjects (41 ± 11 yrs of age) presenting a wide range of BMI (21.4 to 48.6 kg/m(2)) were included. SAT and VAT biopsies were obtained from patients undergoing abdominal surgeries. Clock genes expressions were evaluated by qRT-PCR. The only clock gene that showed higher expression (p .0001) in SAT in comparison to VAT was PER1 of female (372%) and male (326%) subjects. Different patterns of expression between the AM and PM periods were observed, in particular REV-ERBα, which was reduced (p .05) at the PM period in SAT and VAT of both women and men (women: ~53% lower; men: ~78% lower), whereas CLOCK expression was not altered. Relationships between clock genes were different in SAT vs. VAT. BMI was negatively correlated with SATPER1 (r =?-.549; p =?.001) and SATPER2 (r =?-.613; p =?.0001) and positively with VATCLOCK (r =?.541; p =?.001) and VATBMAL1 (r =?.468; p =?.007) only in women. These data suggest that the circadian clock machinery of adipose tissue depots differs between female and male subjects, with a sex-specific effect observed for some genes.
