? Premise of the study: Studies on genetic structure of plant populations help us understand the history of local flora and vegetation. In this study, we focus on the temperate-boreal herb Cardamine scutata from northeast Asia, an area with scarce phylogeographic studies. We explore patterns of genetic variation within this species, with an aim to infer its (post-) glacial history with reference to colonization routes and migrations via land bridges. ? Methods: We analyzed 46 populations sampled in Japan, Kamchatka, and Korea using AFLP and cpDNA sequence data. ? Key results: Two intraspecific genetic groups were resolved, distributed in the northeastern and southwestern part of the study area, most likely reflecting lineages isolated from each other during (at least) the last glaciation. A zone of secondary contacts was found in central/northern Honshu, and a few cases of long-distance dispersal were observed. We detected efficient gene flow across the marine straits, supporting the role of land bridges created by sea level decline during the last glacial period. The cpDNA data indicated extensive recent expansion and diversification within both lineages. We inferred recent colonization of Kamchatka from Hokkaido, associated with genetic impoverishment. ? Conclusions: The pattern of north-south genetic differentiation found in C. scutata is rather common among several other plant species studied in Japan, despite their distinct biological features. We assume that different processes and factors may have brought about this similarity. Overall, this study contributes to better understanding of the biogeography of northeast Asia.  相似文献

Previously we cloned the human MNB/DYRK1A gene from the "Down syndrome critical region" on chromosome 21. This gene encodes a dual specificity protein kinase that catalyzes its autophosphorylation on serine/threonine and tyrosine residues. The recovery process after chilling-induced photoinhibition of photosystem I (PSI) was studied in leaves of a chilling-sensitive plant, cucumber (Cucumis sativus L. cv. Nanshin). Determination of chlorophyll content, photosystem (PS) activities in vivo and in vitro, and the amount of reaction-center subunits of PSI revealed that: (i) The content of chlorophyll decreased to 70% of the original level gradually from 1 to 3 days after exposure to a low temperature. (ii) The amount of functional PSI per unit leaf area was reduced to 30% of the initial level by the chilling treatment. The amount of functional PSI gradually increased during the next 6 days but only to 50% of the original level. (iii) When expressed on a chlorophyll basis, however, the amount of functional PSI recovered to 90% of the original level 6 days after the treatment. (iv) The residual amount of chlorophyll on the third day after the treatment closely correlated with the amount of functional PSI at that point. These results indicate that the decrease in chlorophyll content at a normal growth temperature after chilling treatment is a consequence of the degradation of irreversibly damaged PSI complexes. Immunoblot analysis confirmed that PsaAB protein, the reaction-center subunits of PSI, was degraded during the 3 days after chilling treatment. Some characteristics of the chilling injury frequently reported, i.e. irreversibility of the injury and development of visible symptoms at room temperature, can be explained as a consequence of the chilling-induced photoinhibition of PSI.  相似文献

We have screened for possible disease-causing mutations in the peripherin/retinal degeneration slow (RDS) gene in 13 Japanese families with autosomal dominant retinitis pigmentosa (ADRP). Using polymerase chain reaction-single strand conformation polymorphism analysis, a novel mutation at codon 214 was found in which the highly conserved cysteine was replaced with a serine in one family. The mutation at codon 214 was found in all three affected siblings of this family, but none of the 40 normal control individuals had this mutation. These results strongly suggest, that the mutation is pathogenic for RP in this family. The clinical phenotype for this family is a late-onset form of ADRP.  相似文献

A HindIII fragment harboring the intact spo0C gene of Bacillus subtilis was cloned with rho 11 temperate bacteriophage as a vector. Transformation experiments with the DNA from rho 11 dspo0C+ specialized transducing phage showed that the spo0C gene resides on a 5.3-megadalton fragment generated by HindIII digestion. The 5.3-megadalton fragment also contains the intact spo0A gene, but not spoIIIA, spoIIIB, or spoIVB.  相似文献

Studies in fish and amphibia have shown that graded Bmp signalling activity regulates dorsal-to-ventral (DV) patterning of the gastrula embryo. In the ectoderm, it is thought that high levels of Bmp activity promote epidermal development ventrally, whereas secreted Bmp antagonists emanating from the organiser induce neural tissue dorsally. However, in zebrafish embryos, the domain of cells destined to contribute to the spinal cord extends all the way to the ventral side of the gastrula, a long way from the organiser. We show that in vegetal (trunk and tail) regions of the zebrafish gastrula, neural specification is initiated at all DV positions of the ectoderm in a manner that is unaffected by levels of Bmp activity and independent of organiser-derived signals. Instead, we find that Fgf activity is required to induce vegetal prospective neural markers and can do so without suppressing Bmp activity. We further show that Bmp signalling does occur within the vegetal prospective neural domain and that Bmp activity promotes the adoption of caudal fate by this tissue.  相似文献
