Synthesis,docking studies,in vitro cytotoxicity evaluation and DNA damage mechanism of new tyrosine-based tripeptides

Peptides are one of the leading groups of compounds that have been the subject of a great deal of biological research and still continue to attract researchers' attention. In this study, a series of tripeptides based on tyrosine amino acids were synthesized by the triazine method. The cytotoxicity properties of all compounds against human cancer cell lines (MCF-7), ovarian (A2780), prostate (PC-3), and colon cancer cell lines (Caco-2) were determined by the 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide assay method, and % cell viability and logIC50 values of the compounds were calculated. Significant decreases in cell viability were observed in all cells (p < 0.05). The comet assay method was used to understand that the compounds that showed a significant decrease in cell viability had this effect through DNA damage. Most of the compounds exhibited cytotoxicity by DNA damage mechanism. Besides, their interactions between investigated molecule groups with PDB ID: 3VHE, 3C0R, 2ZCL, and 2HQ6 target proteins corresponding to cancer cell lines, respectively, were investigated by docking studies. Finally, molecules with high biological activity against biological receptors were determined by ADME analysis.     

Development of doxorubicin loading platform based albumin-sporopollenin as drug carrier

Albumin is thought as an drug carrier for doxorubicin (DOX). The binding of doxorubicin to albumin was studied on the surface of sporopolleninin (SP) to produce a new drug system based natural materials. Human serum albumin (HSA) was immobilized on SPIONs in 20 mM Tris buffer, 7.4 of pH. Data showed that binding amount of HSA has been found to be as 285.53 µg to the 25 mg of Sporopolleninin which also bounded 319.76 µM of DOX. Binding of protein and drug to Sp were clarified by SEM, EDX and FT-IR analysis.     

Synthesis and characterization of cyclic peptides that are β‐helical in trifluoroethanol

We show that three designed cyclic d ,l ‐peptides are β‐helical in TFE—a solvent in which the archetypal β‐helical peptide, gA, is unstructured. This result represents an advance in the field of β‐helical peptide foldamers and a step toward achieving β‐helical structure under a broad range of solvent conditions. We synthesized two of the three peptides examined using an improved variant of our original CBC strategy. Here, we began with a commercially available PEG–PS composite resin prefunctionalized with the alkanesulfonamide ‘SCL’ linker and preloaded with glycine. Our new conditions avoided C‐terminal epimerization during the CBC step and simplified purification. In addition, we present results to define the scope and limitations of our CBC strategy. These methods and observations will prove useful in designing additional cyclic β‐helical peptides for applications ranging from transmembrane ion channels to ligands for macromolecular targets. Identification of factor XI deficiency in Holstein cattle in Turkey

Factor XI (FXI) is a plasma protein that participates in the formation of blood clots. Factor XI deficiency is autosomal recessive hereditary disorder that may be associated with excess bleeding in Holstein cattle.     

A 2-year aeropalynological survey of allergenic pollen in the atmosphere of Kastamonu, Turkey

Knowledge of airborne pollen concentrations and the weather conditions influencing them is important for air quality forecasters, allergists and allergy sufferers. For this reason, a 7-day recording volumetric spore trap of the Hirst design was used for pollen monitoring between January 2006 and December 2007 in Kastamonu, Turkey. A total of 293,427 pollen grains belonging to 51 taxa were recorded during the study period. In the 2?years of study, the period March–August was identified as the main pollination season for Kastamonu. The highest monthly pollen counts were observed in May in both years. Six taxa made up 86.5% of the total amount of pollen recorded in the atmosphere of Kastamonu. These were as follows: Pinaceae (42.9%), Cupressaceae (20.6%), Poaceae (9.7%), Quercus (5.5%) Betula (5.3%) and Carpinus (2.6%). Four of these are considered to be highly allergenic (Betula, Carpinus, Cupressaceae and Poaceae). There were also a greater percentage of highly allergenic taxa found within the city, including Betula pendula that is not part of the local flora. This shows that through urban planting, the public and municipalities can unconsciously create a high risk for allergy sufferers. Daily average pollen counts from the six most frequently recorded pollen types were entered into Spearman’s correlation analysis with meteorological data. Mean daily temperature, relative humidity, daily rainfall and wind speed were found to significantly (p?<?0.05) affect atmospheric pollen concentrations, but the relationships between pollen concentrations and meteorological variables can vary and so there is a need for more local studies of this nature.     

Novel polymorphisms in ovine prion protein gene

The aim of this study was to identify the PRNP polymorphisms outside the standard codons 136, 154 and 171 in 1110 sheep with no clinical sign of scrapie from all 18 Turkish native sheep breeds and compare our results with published data on ovine PRNP polymorphism from other regions of the world. Among the 22 amino acid polymorphisms and three silent mutations, 10 were novel for ovine PRNP: p.Gly94Gly, p.Leu128Ile, p.Met132Leu, p.Ser135Arg, p.Met137Val, p.Asn146Lys, p.Arg159Arg, p.Tyr160Asn, p.Gln163His and p.Thr193Ser. These data reveal that sheep breeds close to the historic center of small ruminant domestication have remained highly diverse in the prion gene locus, with distinctive genetic similarities to both Asian and European sheep breeds.
