Urotensin II (UII), a cyclic dodecapeptide, is a potent mammalian vasoconstrictive substance recently shown to induce proliferation of vascular smooth muscle cells (VSMCs). However, little is known about mechanisms involved in UII-induced mitogenic response such as cell proliferation. To investigate the intracellular signaling pathways involved in this process, we examined the effects of UII on activation of extracellular signal-regulated kinase (ERK) and focal adhesion kinase (FAK) in VSMCs. UII stimulated in time- and dose-dependent manners the phosphorylation level of ERK. In contrast, UII failed to alter the phosphorylation level of FAK. Although angiotensin II-induced ERK phosphorylation was noted even in suspended cells, UII failed to induce an increase in ERK phosphorylation in such cells. On the other hand, UII induced an increase in the phosphorylation level of ERK, but not FAK, in cells adherent to fibronectin. Furthermore, UII-induced proliferation of VSMCs was inhibited by ERK kinase inhibitor PD98059. Our results suggested that activation of integrin-mediated signaling pathways play a critical role in UII-induced phosphorylation of ERK, leading to proliferation of VSMCs, which does not involved increased phosphorylation of FAK. 相似文献

Many free-swimming unicellular organisms show negative gravitaxis, i.e. tend to swim upward, although their specific densities are higher than the medium density. To obtain clues to the mechanism of this behavior, we examined how a mutation in motility or behavior affects the gravitaxis in Chlamydomonas. A phototaxis mutant, ptx3, deficient in membrane excitability showed weakened gravitaxis, whereas another phototaxis mutant, ptx1, deficient in regulation of flagellar dominance displayed normal gravitaxis. Two mutants that swim backwards only, mbo1 and mbo2, did not show any clear gravitaxis. We also isolated two novel mutants deficient in gravitaxis, gtx1 and gtx2. These mutants displayed normal motility and physical characteristics of cell body as assessed by the behavior of anesthetized cells. However, these cells were found to have defects in physiological responses involving membrane excitation. These observations are consistent with the idea that the gravitaxis in Chlamydomonas involves a physiological signal transduction system, which is at least partially independent of the system used for phototaxis. 相似文献

The TOR protein is a phosphoinositide kinase-related kinase widely conserved among eukaryotes. Fission yeast tor1 encodes an ortholog of TOR, which is required for sexual development and growth under stressed conditions. We isolated gad8, which encodes a Ser/Thr kinase of the AGC family, as a high-copy suppressor of the sterility of a tor1 mutant. Disruption of gad8 caused phenotypes similar to those of tor1 disruption. Gad8p was less phosphorylated and its kinase activity was undetectable in tor1Delta cells. Three amino acid residues corresponding to conserved phosphorylation sites in the AGC family kinases, namely Thr387 in the activation loop, Ser527 in the turn motif and Ser546 in the hydrophobic motif, were important for the kinase activity of Gad8p. Tor1p was responsible for the phosphorylation of Ser527 and Ser546, whereas Ksg1p, a PDK1-like kinase, appeared to phosphorylate Thr387 directly. Altogether, Tor1p, Ksg1p and Gad8p appear to constitute a signaling module for sexual development and growth under stressed conditions in fission yeast, which resembles the mTOR-PDK1-S6K1 system in mammals and may represent a basic signaling module ubiquitous in eukaryotes. 相似文献

Cichlid fishes of the east African Great Lakes represent a paradigm of adaptive radiation. We conducted a phylogenetic analysis of cichlids including pan-African and west African species by using insertion patterns of short interspersed elements (SINEs) at orthologous loci. The monophyly of the east African cichlids was consistently supported by seven independent insertions of SINE sequences that are uniquely shared by these species. In addition, data from four other loci indicated that the genera Tilapia (pan-African) and Steatocranus (west African) are the closest relatives to east African cichlids. However, relationships among Tilapia, Steatocranus, and the east African clade were ambiguous because of incongruencies among topologies suggested by insertion patterns of SINEs at six other loci. One plausible explanation for this phenomenon is incomplete lineage sorting of alleles containing or missing a SINE insertion at these loci during ancestral speciation. Such incomplete sorting may have taken place earlier than 14 MYA, followed by random and stochastic fixation of the alleles in subsequent lineages. These observations prompted us to consider the possibility that cichlid speciation occurred at an accelerated rate during this period when the African Great Lakes did not exist. The SINE method could be useful for detecting ancient exclusive speciation events that tend to remain hidden during conventional sequence analyses because of accumulated point mutations. 相似文献

The klotho gene encodes a novel type I membrane protein of beta-glycosidase family and is expressed principally in distal tubule cells of the kidney and choroid plexus in the brain. These mutants displayed abnormal calcium and phosphorus homeostasis together with increased serum 1,25-(OH)2D. In kl-/- mice at the age of 3 wk, elevated levels of serum calcium (10.9 +/- 0.31 mg/dl vs. 10.0 +/- 0.048 mg/dl in wild-type mice), phosphorus (14.7 +/- 1.1 mg/dl vs. 9.7 +/- 1.5 mg/dl in wild type) and most notably, 1,25-(OH)2D (403 +/- 99.7 mg/dl vs. 88.0 +/- 34.0 mg/dl in wild type) were observed.Reduction of serum 1,25-(OH)2D concentrations by dietary restriction resulted in alleviation of most of the phenotypes, suggesting that they are downstream events resulting from elevated 1,25-(OH)2D. We searched for the signals that lead to up-regulation of vitamin D activating enzymes. We examined the response of 1alpha-hydroxylase gene expression to calcium regulating hormones, such as PTH, calcitonin, and 1,25-(OH)2D3. These pathways were intact in klotho null mutant mice, suggesting the existence of alternate regulatory circuits. We also found that the administration of 1,25-(OH)2D3 induced the expression of klotho in the kidney. These observations suggest that klotho may participate in a negative regulatory circuit of the vitamin D endocrine system, through the regulation of 1alpha-hydroxylase gene expression. 相似文献

Scope In this study, a dynamic model was built in which LCA and PBM were integrated to quantitatively assess the total environmental
impacts induced by the product population in a society over time. Specifically, a determination was carried out concerning
how Japan’s air conditioner population is used (lifetime distribution, number of units, etc.) and an assessment was made concerning
the Global Warming Potential (GWP) associated with the air conditioner population.
Methods The proposed dynamic model was applied to air conditioners for analyzing the total GWP caused by the air conditioner population
in Japan from 1990 to 2010. To create a trend forecast model for future environmental load, scenarios for air conditioner
production up to 2010 were formulated and the total GWP from the air conditioner population was predicted. Conducted also
were sensitivity analyses whose parameters were air conditioner performance, lifetime and the rate of refrigerant recovery
when retired units are processed.
Results and Discussion Applying the PBM to the air conditioner population in 2000, it was found that 81.5 million units consumed 5.94 x 10p10 kWh in that year, which was a 6.1 % increase in the total annual power consumption in 1990. In both a stationary scenario
and a steady growth (1.5% annual increase), it was found that the total GWP would be 27.7% higher than in 1990 under the stationary
scenario and 37.8% higher under the steady growth scenario. The improvements in air conditioner performance will have a small
effect on reducing the total GWP from that population. Furthermore, in connection with the average lifetime, it was found
that the GWP, due to refrigerant releases when units are disposed of, would be relatively large in 2000 and the following
years.
Conclusions Thus, shorter product lifetimes will spur a replacement of air conditioners with new units, a situation that will only lead
to the reduction of GWP if the recovery rate of refrigerant is to be achieved to more than 50% under the stationary scenario.
Recommendations and Outlook To meet COP3 targets for Japan in 2010 (i.e. to reach the same level as in 1990 for household appliances), our study shows
that it will be vital to raise the refrigerant recovery rate. If the number of air conditioners in use remains unchanged,
recovery would have to be 45.7%, but under the steady growth scenario it would have to be at least 60.4%. Therefore, it will
be difficult to meet COP3 targets unless the refrigerant recovery rate is strongly increased. This method is applicable to
assess not only the GWP of air conditioners, but also other environmental impacts caused by a variety of product populations,
which will be quite effective for setting targets of products’ performance, policymaking, etc. 相似文献

4-Hydroxy-2-nonenal (HNE) and malondialdehyde (MDA) are well-known toxic products of lipid peroxidation. Phosphatidylcholine aldehydes are also known as oxidation products of phosphatidylcholine. The mechanism of the formation of these compounds in vivo has been a long-standing question. We observed that the rapid reaction of hemoproteins (methemoglobin, metmyoglobin, and cytochrome c) with 1-palmitoyl-2-(13-hydroperoxy-cis-9, trans-11-octadecadienoyl) phosphatidylcholine (PLPC-OOH), having a hydroperoxylinoleoyl residue, generated HNE, MDA, and the phosphatidylcholine aldehyde 1-palmitoyl-2-(9-oxononanoyl) phosphatidylcholine. The efficiencies (mol% yield) of the formation of HNE and MDA from decomposed PLPC-OOH by methemoglobin, metmyoglobin, and cytochrome c after incubation for 10 min were 1.6, 1.0, and 1.0% for HNE and 1.2, 0.6, and 0.9% for MDA, respectively. When 1-palmitoyl-2-linoleoyl phosphatidylcholine was incubated with lipoxidase and methemoglobin, the formation of HNE and the phosphatidylcholine aldehyde 1-palmitoyl-2-(9-oxononanoyl) phosphatidylcholine was observed. When 1-palmitoyl-2-arachidonyl phosphatidylcholine was used instead of 1-palmitoyl-2-linoleoyl phosphatidylcholine, the phosphatidylcholine aldehyde 1-palmitoyl-2-oxovaleroyl phosphatidylcholine was obtained. These data suggest that HNE and phosphatidylcholine aldehydes might be rapidly formed from phosphatidylcholine by lipoxygenase and hemoproteins. Furthermore, hemichrome, converted from methemoglobin by deoxycholic acid and ursodeoxycholic acid, showed marked decomposition of HNE.
