The respective effects of meristem temperature, vapour pressuredeficit (VPD) and photosynthetic photon flux density (PPFD)on leaf elongation rate (LER) of maize, in the absence of waterdeficit in the soil have been quantified. This analysis wascarried out in a series of field experiments in northern andsouthern France over several seasons and years, and in growthchamber experiments. LER was measured with 10 min steps, togetherwith meristem temperature, VPD and PPFD at leaf level in threetypes of experiments: in growth chamber experiments with stepsin PPFD or VPD at constant meristem temperature, in growth chamberexperiments with several combinations of constant, but contrasting,PPFDs, VPDs and meristem temperatures, and in the field withfluctuating conditions, (i) When evaporative demand was low(night or day with low air VPD), LER was only linked to meristemtemperature, regardless of other climatic conditions, (ii) Lighthad no effect per se on LER in the range from 0 to 1500 molm–2 s–1 for time-scales longer than 2 h, providedthat its indirect effects on meristem temperature and on evaporativedemand were corrected (in the growth chamber) or taken intoaccount (in the field), and provided that cumulated PPFD overa weekly time-scale was compatible with field conditions, (iii)Evaporative demand sensed by growing leaves, as estimated bymeristem-to-air vapour pressure difference, markedly affectedLER in the range from 1–4 kPa, at all time-scales understudy, with a unique relationship in the growth chamber (constantconditions) and in the field (fluctuating conditions). Thiseffect was only observed when PPFD was high enough for stomatato open. The negative effect of evaporative demand on LER wasprobably not due to long distance root-to-shoot signalling,since soil was wet, calculated root water potential remainedclose to 0 MPa and concentration of ABA in the xylem sap wasvery low. Therefore, it is proposed to model maize LER witha two-step process, involving the calculation of the maximumLER at a given meristem temperature and then the calculationof the reduction in LER due to evaporative demand. Joint analysisof the whole set of data by using the two equations yieldeda r2 of 0.75. This two-step process would be more accurate thanthe provision of LER from temperature only in cases where airVPD frequently exceeds 2 kPa. Abstract: The role of excitatory amino acid (EAA) receptors located in the ventral tegmental area (VTA) in tonic and phasic regulation of dopamine release in the ventral striatum was investigated. Microdialysis in conscious rats was used to assess dopamine release primarily from the nucleus accumbens shell region of the ventral striatum while applying EAA antagonists or agonists to the VTA. Infusion of the AMPA/kainate receptor antagonist 6-cyano-7-nitroquinoxaline-2,3-dione (25 and 100 µ M ) into the VTA did not affect dopamine release in the ventral striatum. In contrast, intra-VTA infusion of the NMDA receptor antagonist 2-amino-5-phosphopentanoic acid (100 and 500 µ M ) dose-dependently decreased the striatal release of dopamine. Intra-VTA application of the ionotropic EAA receptor agonists NMDA and AMPA dose-dependently (10 and 100 µ M ) increased dopamine efflux in the ventral striatum. However, infusion of 50 or 500 µ M trans -(±)-1-amino-1,3-cyclopentanedicarboxylic acid (ACPD), a metabotropic EAA receptor agonist, did not significantly affect these levels. These data suggest that NMDA receptors in the VTA exert a tonic excitatory influence on dopamine release in the ventral striatum. Furthermore, dopamine neurotransmission in this region may be enhanced by activation of NMDA and AMPA receptors, but not ACPD-sensitive metabotropic receptors, located in the VTA. These data further suggest that EAA regulation of dopamine release primarily occurs in the VTA as opposed to presynaptically at the terminal level.  相似文献

Synopsis Lake Malawi/Niassa is the second largest rift valley lake in Africa, with an area of 28 800 km2, and an average and maximum depth of 292 m and>700 m, respectively. The lake is well known for the great diversity of fish occurring in the inshore zone. However, the offshore fish community is poorly documented. To rectify this, regular sampling was undertaken over two years, using trawl and gillnets at six offshore locations. This paper reports on the species composition, spatial distribution and breeding biology of the dominant cichlids species from the offshore pelagic zone. Cichlids formed approximately 88% of the offshore fish biomass. Most abundant were two species of zooplanktivores in the genus Diplotaxodon that made up 71% of the offshore fish biomass. An undescribed species, given the cheironym D. bigeye, was mainly found at a depth of 220 m during the day, but moved into near surface waters at night when the moon was full. This species was absent from the shallow regions of the lake. The most abundant offshore species was D. limnothrissa, which was distributed evenly throughout the lake to depths of 220 m. A less common offshore zooplanktivore was Copadichromis quadrimaculatus that formed 5% of the biomass and was confined to the upper 100 m of the water column. The main piscivores were in the genus Rhamphochromis and formed approximately 10% of the offshore fish biomass. The two dominant taxa were R. longiceps and the large Rhamphochromis group, and both were more common in the southern half of the lake. The former occurred mainly in the upper 100 m of the water column and the latter mainly at depths of 100–150 m. The length at maturity and fecundity for the dominant offshore species were estimated and seasonal breeding cycles determined from gonad activity and gonado-somatic indices.  相似文献

In vitro plantlet regeneration systems for the seed geranium (Pelargonium x hortorum Bailey) using cotyledon, hypocotyl and root explants were optimized by studying the influence of seedling age, growth regulators and excision orientation on organogenesis. Indole-3-acetic acid combined with zeatin yielded the highest rate of shoot production on cotyledon explants (0.2–2 shoots per explant). More shoots were produced on explants cut from the most basal region of cotyledons from 2 to 4-day-old seedlings than from older seedlings or more distal cut sites. Hypocotyl explants produced the highest number of shoots, up to 40 shoots per explant, on indole-3-acetic acid (2.8–5.6 mM) + zeatin (4.6 mM) or thidiazuron (4.5 mM). Maximum shoot formation (0.3–1.4 shoots per explant) on root explants occurred when they were cultured on medium containing zeatin. Regenerated shoots rooted best on a basal medium containing no growth regulators. Nocturnal flying insects were collected monthly for 13 months using ultra violet light-traps set at various vertical levels in a weakly-seasonal, tropical lowland dipterocarp forest in Sarawak, Malaysia. Abundance, faunal composition, size distribution and guild structure of these samples were analyzed with respect to temperal and vertical distributions. The nocturnal flying insect community in the canopy level was highly dominated by fig wasps (84%) in individual number, and by scarabaeid beetles (28%) in weight. A principal component analysis on monthly catches detected non-random, seasonal trends of insect abundance. The first two principal trends were an alternation of wetter (September to January) and less wet seasons (February to August) and an alternation between the least wet (January to March) and the other seasons. Many insect groups were less abundant in the least wet season than the other seasons, whilst inverse patterns were found in Scarabaeidae and Tenebrionidae. Significantly positive and negative correlations between monthly catch and rainfall were detected only in ovule-feeders and in phloem-feeders, respectively. Delayed, significant negative correlations between monthly catch and 1–3 month preceding rainfall were more frequently detected in phytophages, phloem-feeders, seed-feeders, wood-borers and scavengers. The peak in abundance along vertical levels were found at the canopy level (35 m) for phloem-, ovule-, seed-, root-, fungal-feeders and nectar collectors, at an upper subcanopy level (25 m) for scavengers and aquatic predators, and at a middle subcanopy level (17 m) for ants. The fragile X syndrome is the result of amplification of a CGG trinucleotide repeat in the FMR1 gene and anticipation in this disease is caused by an intergenerational expansion of this repeat. Although regression of a CGG repeat in the premutation range is not uncommon, regression from a full premutation (>200 repeats) or premutation range (50–200 repeats) to a repeat of normal size (<50 repeats) has not yet been documented. We present here a family in which the number of repeats apparently regressed from approximately 110 in the mother to 44 in her daughter. Although the CGG repeat of the daughter is in the normal range, she is a carrier of the fragile X mutation based upon the segregation pattern of Xq27 markers flanking FMR1. It is unclear, however, whether this allele of 44 repeats will be stably transmitted, as the daughter has as yet no progeny. Nevertheless, the size range between normal alleles and premutation alleles overlap, a factor that complicates genetic counseling.  相似文献

A departure from single-system dynamics, that may arise in characterizing self-organized dynamics of complex systems, is dealt with by using the Karhunen-Loève expansion of the trajectory matrix to decompose an experimental signal in a sum of spectral features. For an electroencephalographic -signal, a separation of waves and extraction of additive sub-signals are achieved, each sub-signal covering a well-bounded and physiologically meaningful frequency range. From the subsignals, an attractor that vanishes on phase-randomizing the data is characterized, under conditions where none was found for the recorded signal.  相似文献

Free and immobilized cultures of Spirulina maxima for swine waste treatment   总被引:6,自引:0,他引:6  
We have analyzed the behavior of spirulina maxima at increasing concentration of ammonium nitrogen present in swine waste when it is either growing in suspension or immobilized in polymeric supports. We compared the response of spirulina maxima growth to different concentrations of aeration stabilized swine waste (total phosphorus, ammonium nitrogen) as a way to determine the treatment efficiency of both systems. At a dilution of 50 % of swine waste, the suspended system reached the best results for biomass concentration and nutrient removal. In the immobilized system, at dilutions of 25 and 50 % of swine waste, more than 90 % ammonium nitrogen removal was obtained, and the optimal cell concentration for immobilization was 2 g/l (wet basis).  相似文献

Addition of ATP (>0.1 mM) to cultures of human breast cancer T47D cells resulted in an inhibition of cell proliferation. The inhibition was found to be specific for ATP, and dependent on its concentration. Growth inhibition continued for at least three days, although ATP and its hydrolysis products were metabolized within one day. Conditioned medium from ATP-treated cultures (CM+) was found to inhibit the growth of cells that were not exposed to ATP. This is an indication that extracellular factors, besides ATP, are involved in the inhibition process. The inhibition was maintained after dialysis of the CM+, using an 8 kDa cut-off membrane. Conditioned medium from untreated cultures (CM-), however, only slightly affected cell growth. The data suggest that the CM+ -induced cell growth inhibition is mediated by an ATP-activated growth inhibiting factor. Flow microfluorometry and thymidine incorporation experiments have shown that the growth arrest is mainly due to the elongation of the S-phase of the cell cycle. © 1993 Wiley-Liss, Inc.  相似文献

For many years, the high prevalence of the fragile X syndrome was thought to be caused by a high mutation frequency. The recent isolation of the FMR1 gene and identification of the most prevalent mutation enable a more precise study of the fragile X mutation. As the vast majority of fragile X patients show amplification of an unstable trinucleotide repeat, DNA studies can now trace back the origin of the fragile X mutation. To date, de novo mutations leading to amplification of the CGG repeat have not yet been detected. Recently, linkage disequilibrium was found in the Australian and US populations between the fragile X mutation and adjacent polymorphic markers, suggesting a founder effect of the fragile X mutation. We present here a molecular study of Belgian and Dutch fragile X families. No de novo mutations could be found in 54 of these families. Moreover, we found significant (P < 0.0001) linkage disequilibrium in 68 unrelated fragile X patients between the fragile X mutation and an adjacent polymorphic microsatellite at DXS548. This suggests that a founder effect of the fragile X mutation also exists in the Belgian and Dutch populations. Both the absence of new mutations and the presence of linkage disequilibrium suggest that a few ancestral mutations are responsible for most of the patients with fragile X syndrome.  相似文献
