Premelting at the surface of ice crystals is caused by factors such as temperature, radius of curvature, and solute composition. When polycrystalline ice samples are warmed from well below the equilibrium melting point, surface melting may begin at temperatures as low as -15 degrees C. However, it has been reported (. Biophys. J. 65:1853-1865) that when polycrystalline ice was warmed in a differential scanning calorimetry (DSC) pan, melting began at about -50 degrees C, this extreme behavior being attributed to short-range forces. We show that there is no driving force for such premelting, and that for pure water samples in DSC pans curvature effects will cause premelting typically at just a few degrees below the equilibrium melting point. We also show that the rate of warming affects the slope of the DSC baseline and that this might be incorrectly interpreted as an endotherm. The work has consequences for DSC operators who use water as a standard in systems where subfreezing runs are important.  相似文献

A steroid which stabilizes lysosomes in vitro and a pyrogenic steroid which labilizes lysosomes in vitro were compared with respect to their ability to modify lysosomal uptake and lysosomal enzyme levels in vivo. Cortisone acetate increased the uptake of acridine orange by rat liver lysosomes when the dye was administered by intrathoracic injection. The steroid increased and accelerated the uptake of acridine orange so that, in liver lysosomes from treated rats, the maximum uptake was double that of controls and was reached at 2h, whereas in controls the maximum uptake was at 4h after the injection of the dye. This large elevation of uptake is specific to the lysosomal fraction and is not seen in other subcellular fractions of rat liver. The specific activities of a lysosomal enzyme β-N-acetylglucosaminidase were increased in lysosomal fractions from cortisone acetate-treated rats. Etiocholanolone, a steroid which labilizes lysosome in vitro, similarly accelerated and increased acridine orange uptake by lysosomes but had little effect on lysosomal β-N-acetylglucosaminidase levels. Thus the ability of steroids to stabilize or labilize lysosomes in vitro does not correlate with their effect on lysosomal uptake of injected substances in vivo, or with their ability to induce increased specific activities of lysosomal enzymes.  相似文献

Field workers were monitored for dermal and respiratory exposure to chlorpyrifos (with and without crop oil), carbaryl, and permethrin at reentry intervals of 2, 4, 8, 24, and 48 h after application. Insecticides were applied to R3 stage corn through an overhead center-pivot irrigation system. Dermal exposure was measured by analyzing 18 gauze pads attached to the clothing of workers to represent human body regions. Hand exposure was determined using cotton gloves. Respiratory exposure was determined using portable air samplers equipped with polyurethane foam plugs to trap ambient insecticide residues. Gas liquid chromatography was used to quantify residues of chlorpyrifos and permethrin in gauze pads, gloves, and foam plugs. Carbaryl residues in pads, gloves, and foam plugs were analyzed using high-performance liquid chromatography. Highest dermal and respiratory exposures were found at the 2-h reentry interval. Exposures decreased as reentry interval increased. Dermal exposure was primarily confined to the hands. Residues detected by air samplers ranged from 0 to 0.03 micrograms/liter. Based on the estimated percentages of acute toxic dose (all less than 0.00038%), the risk of acute toxicity to workers at the intervals studied was low.  相似文献

A novel series of 22-membered macrocyclic complexes of the type [MLCl2] (M?Co2+, Ni2+ and Cu2+) have been synthesized with newly derived biologically active ligands (L1–LIV). These ligands were synthesized by the condensation of ortho-phthalaldehyde and bis-(4-amino-5-mercapto-1, 2, 4-triazole-3-yl)alkanes. The mode of bonding and overall geometry of the complexes have been inferred through IR, EPR, electronic spectral studies, conductivity, magnetic, thermal, and electrochemical studies. All these complexes have been screened for their antibacterial (Escherichia coli, Staphylococus aureus, Salmonella typhi, Pseudomonas aeruginosa) and antifungal activities (Aspergillus niger, Aspergillus flavus, and Cladosporium) by the minimal inhibitory concentration (MIC) method. Aims: To evaluate the effect of different physicochemical parameters such as agitation, aeration and pH on the growth and nitrile hydratase production by Rhodococcus erythropolis MTCC 1526 in a stirred tank reactor. Methods and Results: Rhodococcus erythropolis MTCC 1526 was grown in 7‐l reactor at different agitation, aeration and controlled pH. The optimum conditions for batch cultivation in the reactor were an agitation rate of 200 rev min?1, aeration 0·5 v/v/m at controlled pH 8. In this condition, the increase in nitrile hydratase activity was almost threefold compared to that in the shake flask. Conclusion: Agitation and aeration rate affected the dissolved‐oxygen concentration in the reactor which in turn affected the growth and enzyme production. Significance and Impact of the Study: Cultivation of R. erythropolis MTCC 1526 in the reactor was found to have significant effect on the growth and nitrile hydratase production when compared to the shake flask.  相似文献

A series of novel 6-methoxy-2-(piperazin-1-yl)-4H-chromen-4-one and 5,7-dimethoxy-2-(piperazin-1-ylmethyl)-4H-chromen-4-one derivatives of biological interest were prepared and screened for their pro-inflammatory cytokines (TNF-α and IL-6) and antimicrobial activity (antibacterial and antifungal). Among all the compound screened (5aj and 10kt), the compounds 5c, 5g, 5h, 10l, 10m, 10n and 10r found to have promising anti-inflammatory activity (up to 65–87% TNF-α and 70–93% IL-6 inhibitory activity) at concentration of 10 μM with reference to standard dexamethasone (71% TNF-a and 84% IL-6 inhibitory activities at 1 μM) while the compounds 5b, 5i, 5j, 10s and 10t found to be potent antimicrobial agent showing even 2 to 2.5-fold more potency than that of standard ciprofloxacin and miconazole at the same MIC value of 10 μg/mL.  相似文献

A protocol for efficient direct gene transfer by using particle gun bombardment was developed for mothbeanVigna aconitifolia L. Jacq. Marechal. Hypocotyl explants from 2 cultivars of mothbean were transformed with 3 plasmids: pBI121, pHS101, and pHS102. Stable transformants were regenerated on MS medium supplemented with benzyladenine, α-naphthaleneacetic acid, and kanamycin. The helium pressure, plasmid type, and cultivar that were used determined the stable transformation frequency. Complete plants were regenerated and transferred to soil. The integration of the stable transgenes and reporter genes in plant genomes was shown by means of PCR amplification of these genes from plant genomic DNA and Southern blot hybridization with gene-specific probes.
