We isolated and characterized 60 novel microsatellite markers from the closely related oldfield mouse (Peromyscus polionotus) and deer mouse (Peromyscus maniculatus) for studies of conservation, ecological, quantitative and population genetics. We assessed all 60 markers in a wild population of Peromyscus polionotus rhoadsi (N = 20) from central Florida and found an average of nine alleles per marker and an observed heterozygosity (HO) of 0.66 (range = 0.00–1.00). These polymorphic markers contribute to the growing number of genomic resources for Peromyscus, an emerging model system for ecological and evolutionary research.  相似文献

A significant challenge to population genetic studies of the dengue vector, Aedes aegypti, has been the lack of polymorphic microsatellite loci. In an effort to develop useful markers, we evaluated the genetic variation at 17 microsatellite loci identified in the A. aegypti genome. Nine loci with at least five alleles were identified in field‐collected specimens from Thailand. An additional two loci carried five alleles if samples from an A. aegypti laboratory colony were included. Our results greatly increase the number of highly variable markers available for the study of the genetics and the population structure of this medically important species.  相似文献

PICROTOXIN has been shown to antagonize specifically the inhibitory action of λ-aminobutyric acid (GABA) as opposed to glycine (Gly) on cells of the cuneate nucleus, either spontaneously active or excited by peripheral stimulation1. Enzymatic Detachment of Endplate Acetylcholinesterase from Muscle   总被引:23,自引:0,他引:23  
AT the vertebrate neuromuscular junction acetylcholinesterase catalyses the hydrolysis of the transmitter, acetylcholine, which is released from presynaptic nerve terminals1,2. The enzyme is present in high concentration at the endplate, where it can be located by histochemical3 and autoradiographic4 methods. Electron microscopic studies of the endplate region show most of the histochemical reaction product to be in the synaptic cleft or associated with the nerve and muscle membranes5–9. We report here that enzymatic treatment of intact muscle causes the detachment of active endplate acetylcholinesterase from the muscle into the bathing fluid.  相似文献

Fitness‐related traits, such as immunity and reproduction, are typically condition dependent, and are predicted to trade off against each other because they share a pool of energy and resources. It is generally assumed that the resources and energy required for immune processes and reproduction is contained in the body fat; however, few studies quantify the effect of an immune insult on fat load. I tested the hypothesis that if mounting an immune response is physiologically costly in terms of consuming limited resources from body fat, and reproduction also relies on these resources, then an immune challenge should cause a reduction in fat load and an associated decline in reproduction. I tested this hypothesis by assaying the fat content of female Wellington tree weta (Hemideina crassidens) after repeated challenges with lipopolysaccharide (LPS), and counting the number of oviposited eggs in addition to assaying the egg protein content, a possible measure of egg quality. In line with my prediction, immune‐challenged females had significantly smaller fat loads and laid significantly fewer eggs than saline‐injected controls. Moreover, the protein content of the eggs laid by immune‐challenged females was significantly lower than that of controls. My experimental results show that an immune challenge with a non‐pathogenic stimulant increases fat consumption, which in turn limits the resources available for reproduction. 1. Ecosystems are strongly influenced by land use practices. However, identifying the mechanisms behind these influences is complicated by the many potential pathways (often indirect) between land use and ecosystems and by the long‐lasting effects of past land use. To support ecosystem restoration and conservation efforts, we need to better understand these indirect and lasting effects. 2. We constructed structural equation models (SEM) to evaluate the direct and indirect effects of contemporary (2002) land use (agriculture and development) and change in land use from 1952 to 2002 on present‐day streams (n = 190) in Maryland, U.S.A. Additional variables examined included site location, system size, altitude, per cent sand in soils, riparian condition, habitat quality, stream water NO3‐N and benthic macroinvertebrate and fish measures of stream condition. Our first SEM (2002 Land Use) included the proportions of contemporary agriculture and development in catchments in the model. The second SEM (Land Use Change) included five measures of land use change (proportion agricultural in both times, developed in both times, agricultural in 1952 and developed in 2002, forested in 1952 and developed in 2002 and agricultural in 1952 and forested in 2002). 3. The data set fit both SEMs well. The 2002 Land Use model explained 71% of variation in NO3‐N and 55%, 42% and 38% of variation in riffle quality, macroinvertebrate condition and fish condition, respectively. The Land Use Change model explained similar amounts of variation in NO3‐N (R2 = 0.72), riffle quality (R2 = 0.57) and macroinvertebrate condition (R2 = 0.44) but slightly more variation in fish condition (R2 = 0.43). 4. Both models identified pathways through which landscape variables affect stream responses, including negative direct effects of latitude on macroinvertebrate and fish conditions and positive direct and indirect effects of altitude on NO3‐N, riffle quality and macroinvertebrate and fish conditions. The 2002 Land Use model showed contemporary development and agriculture had positive total effects on NO3‐N (both through direct pathways); contemporary development had negative effects on macroinvertebrate condition. The Land Use Change model showed that contemporary developed land that was forested in 1952 had no effects on NO3‐N; current developed land that was developed or agricultural in 1952 showed positive effects on NO3‐N. Forests that were agricultural in 1952 had negative effects on NO3‐N, suggesting reduced NO3‐N export with reforestation. The Land Use Change model also showed negative total effects of all types of contemporary developed land (developed, agricultural or forested in 1952) on benthic condition. Developed land that was forested in 1952 had negative effects on fish condition. Forest sites that were agricultural in 1952 had negative effects on fish and macroinvertebrate conditions, suggesting a long‐term imprint of abandoned agriculture in stream communities. 5. Our analyses (i) identified multiple indirect effects of contemporary land use on streams, (ii) showed that current land uses with different land use histories can exhibit different effects on streams and (iii) demonstrated an imprint of land use lasting >50 years. Knowledge of these indirect and long‐term effects of land use will help to conserve and restore streams.  相似文献

We infer the first phylogenetic hypothesis for Latridiidae Erichson (Coleoptera: Cucujoidea). Portions of seven genes (18S ribosomal DNA, 28S ribosomal DNA, 12S ribosomal DNA, 16S ribosomal DNA, cytochrome c oxidase I and II and histone III) were analysed. Twenty‐seven latridiid species were included, representing both subfamilies and more than half of the currently recognized genera. Eight outgroup taxa from other families of Cucujoidea were included. Parsimony and partitioned Bayesian analyses were performed on the combined dataset. In both phylogenetic analyses, the enigmatic Akalyptoischion Andrews (Latridiinae) was recovered outside of Latridiidae. The subfamilies Corticariinae and Latridiinae (without Akalyptoischion) were each recovered as monophyletic in both analyses. A new family, Akalyptoischiidae fam.n. is erected based on the results of the phylogenetic study and further support from adult morphology, key features of which are illustrated.  相似文献

ABSTRACT The controversy over the use of null hypothesis statistical testing (NHST) has persisted for decades, yet NHST remains the most widely used statistical approach in wildlife sciences and ecology. A disconnect exists between those opposing NHST and many wildlife scientists and ecologists who conduct and publish research. This disconnect causes confusion and frustration on the part of students. We, as students, offer our perspective on how this issue may be addressed. Our objective is to encourage academic institutions and advisors of undergraduate and graduate students to introduce students to various statistical approaches so we can make well-informed decisions on the appropriate use of statistical tools in wildlife and ecological research projects. We propose an academic course that introduces students to various statistical approaches (e.g., Bayesian, frequentist, Fisherian, information theory) to build a foundation for critical thinking in applying statistics. We encourage academic advisors to become familiar with the statistical approaches available to wildlife scientists and ecologists and thus decrease bias towards one approach. Null hypothesis statistical testing is likely to persist as the most common statistical analysis tool in wildlife science until academic institutions and student advisors change their approach and emphasize a wider range of statistical methods.  相似文献
