Insect cell metabolism was studied in substrate-limited fed batch cultures of Spodoptera frugiperda (Sf-9) cells. Results from a glucose-limited culture, a glutamine-limited culture, a culture limited in both glucose and glutamine and a batch culture were compared. A stringent relation between glucose excess and alanine formation was found. In contrast, glucose limitation induced ammonium formation, while, at the same time, alanine formation was completely suppressed. Simultaneous glucose and glutamine limitation suppressed both alanine and ammonium formation. Although the metabolism was influenced by substrate limitation, the specific growth rate was similar in all cultures. Alanine formation must involve incorporation of free ammonium, if ammonium formation is mediated by glutaminase and glutamate dehydrogenase, as our data suggest. On the basis of the results, two possible pathways for the formation of alanine in the intermediary metabolism in insect cells are suggested. The cellular yield on glucose was increased 6.6 times during glucose limitation, independently of the cellular yield on glutamine, which was increased 50–100 times during glutamine limitation. The results indicate that alanine overflow metabolism is energetically wasteful and that glutamine is a dispensable amino acid for cultured Sf-9 cells. Preliminary data confirm that glutamine can be synthesised by the cells themselves in amounts sufficient to support growth.  相似文献

The role of facilitation in the structuring of plant communities has been often demonstrated in environments under high abiotic stress, especially in semi‐arid and arid ecosystems and high elevations. Few studies, however, analysed facilitation in systems that are highly productive and rich in species, which are thought to be theoretically unlikely to demonstrate strong effects of facilitation. Here, we investigate the importance of Eryngium horridum, a rosette species, on the maintenance of plant diversity in subtropical grasslands in southern Brazil. We evaluated facilitation in areas under two different types of management: abandonment and grazing. Plots were established in areas with and without individuals of E. horridum and all species were identified and had their cover estimated. The Relative Neighbour Effect index was calculated in order to verify the presence of competition or facilitation. Our results indicated facilitation in both abandoned and in grazed grasslands, but apparently through different mechanisms. In the first case, the plant's architecture opens the canopy and allows more light to reach small forbs in the grass matrix. In the second case, E. horridum appears to protect more palatable species from herbivores. Otherwise considered an obnoxious species, E. horridum plays an important ecological role in subtropical grasslands in southern Brazil by facilitating other species and consequently, increasing local richness. Areas with this rosette species are important sources of diaspores, which are able to colonize new open sites and thus, maintain biodiversity.  相似文献

The nitrogen removal potential of phosphate accumulating organisms under anoxic conditions has been evaluated using a laboratory scale sequencing batch reactor fed with synthetic wastewater and operated in a sequence of anaerobic, anoxic and aerobic periods. The phosphate uptake rate under anoxic conditions was lower than that under aerobic conditions. However, in the presence of an external substrate such as glucose and acetate, the fate of phosphate was dependent on the substrate type; phosphate release occurred in the presence of nitrate as long as acetate was present and glucose did not cause any phosphate release. The nitrate uptake rate was also much lower with glucose than acetate. The results implied that poly-hydroxyalkanoates could be oxidized by nitrate and phosphate uptake during the anoxic phase should be introduced into process modeling. © Rapid Science Ltd. 1998  相似文献

This study focused on the investigation of (i) the tetrachloroethene (PCE) toxicity threshold of a reductively dechlorinating mixed culture containing Dehalococcoides spp., (ii) the adsorption of PCE on different types of granular activated carbon (GAC), and (iii) the bioavailability and reductive dechlorination in the presence of GAC. The abundance of Dehalococcoides spp. detected by quantitative real-time polymerase chain reaction (qPCR) was found to increase by 2-4 orders of magnitude during degradation of PCE. No degradation occurred at dissolved concentrations beyond 420 μM (70 mg/L). Different adsorption isotherms were determined for thermally and chemically activated carbons. The addition of GAC to biological assays reduced the dissolved PCE concentration below the toxicity threshold. The combination of microbial reductive dechlorination with GAC adsorption proved to be a promising method for remediation of groundwater contaminated by high concentrations of chloroethenes.  相似文献

There is an increasing awareness that not only area and isolation, but also the characteristics of the landscape surrounding habitat patches influence population persistence and species diversity in fragmented landscapes. In this study, we examine the effects of grassland fragmentation and land use in the landscape matrix (on a 2 km scale) on species richness of plants, butterflies, bees and hoverflies. These organisms were studied in replicated remnant patches of different sizes and isolation, embedded in landscapes dominated either by forest, arable land or a mix of these. We found positive effects of patch area on species richness of the three insect taxa, but not of plants. Isolation had a negative effect only on hoverflies. Matrix type had contrasting effects on the studied taxa. Species richness of plants and butterflies was lowest in patches in landscapes dominated by arable land and highest in forest‐dominated landscapes. For hoverflies, the negative effect of small patch area was strongest in forest‐dominated landscapes, and there was a similar non‐significant trend for bees. Our study shows the importance of considering matrix characteristics when studying responses to habitat fragmentation. Differences in matrix response among organism groups probably impinge on differing mechanisms. A forest matrix is likely to provide additional resources for butterflies but either constitute a barrier to dispersal or deprive resources as compared to an arable matrix for hoverflies. Enhanced plant diversity in grassland patches embedded in forested landscapes can be explained by habitat generalists more easily invading these patches, or by an unpaid extinction debt in these landscapes.  相似文献

1. Riparian plant communities are primarily structured by the hydrological regime of the stream. Models of climate change predict increased temperatures and changed patterns of precipitation that will alter the flow of rivers and streams with consequences for riparian communities. In boreal regions of Europe, stream flows will exhibit earlier spring‐flood peaks of lower magnitude, lower summer flows and higher flows in autumn and winter. We quantified the effects of predicted hydrological change on riparian plant species richness, using four different scenarios for the free‐flowing Vindel River in northern Sweden. 2. We calculated the hydrological niche of vegetation belts by relating the occurrence of species and vegetation belts to data on flood duration for 10 years preceding the vegetation survey. We then used the flood duration predicted for 2071–2100 to estimate expected changes in the extent of each vegetation belt. Using species accumulation curves, we then predicted changes in plant species richness as a result of changes in extent. 3. The two most species‐rich vegetation belts, riparian forest and willow shrub, were predicted to decrease most in elevational extent, up to 39 and 32%, respectively. The graminoid belt below the shrub belt will mainly shift upwards in elevation while the amphibious vegetation belt at the bottom of the riparian zone increases in size. 4. In the Vindel River, the riparian forest and willow shrub zone will lose most species, with reductions of 5–12% and 1–13% per site, respectively, depending on the scenario. The predicted loss from the entire riparian zone is lower, 1–9%, since many species occur in more than one vegetation belt. More extensive species losses are expected in the southern boreal zone for which much larger spring‐flood reductions are projected. 5. With an expected reduction in area of the most species‐rich belts, it becomes increasingly important to manage and protect riparian zones to alleviate other threats, thus minimising the risk of species losses. Restoring river and stream reaches degraded by other impacts to gain riparian habitat is another option to avoid species losses.  相似文献

Hennhöfer, D., Götz, S. & Mitchell, S.F. 2012: Palaeobiology of a Biradiolites mooretownensis rudist lithosome: seasonality, reproductive cyclicity and population dynamics. Lethaia, Vol. 45, pp. 450–461. During the Cretaceous, rudist bivalves were among the most important benthic carbonate producers on tropical to sub‐tropical carbonate platforms. Yet questions concerning the biology of rudist bouquets remain unanswered to a great extent. In this study a monospecific bouquet of the small radiolitid rudist Biradiolites mooretownensis has been evaluated from a palaeobiological angle. Three‐dimensional, high‐resolution, quantitative analysis provides a detailed evaluation of growth and reproduction in an in situ rudist association. A total of 1237 consecutive tomograms with a vertical spacing of 0.1 mm were produced of which 1150 have been digitally measured for total area, number of specimens, packing density, spat density, recruitment, survival time, mortality and accommodation space. The results show constant coverage of about 60%, a stable packing density of 3.2 specimens per cm2 and constant reproduction throughout the bouquet. Time series analysis (spectral analysis) using PAST statistical software shows cyclic spat density every 14.9 mm of vertical growth. Combined with the results of the δ18O isotope analysis (showing cyclicities of 14 mm) one reproduction cycle appears to be annual. 46.4% of all counted specimens died before 3 mm of vertical growth. More than 93% of the initial spat does not exceed 15 mm shell height or 1 year respectively. Two mortality peaks in the juvenile’s life at 4 and 10–15 mm shell height either represent important obstacles in the ontogenetic development of the species or reflect external influences. □ Biradiolites, grinding tomography, palaeobiology, population dynamics, reproduction, rudists, sclerochronology, seasonality.  相似文献

A new species, Alyssum kaynakiae Y?lmaz, is described from southwest Anatolia, Turkey. The new species belongs to Alyssum sect. Gamosepalum (Hausskn.) T. R. Dudley. Alyssum kaynakiae is closely related to A. niveum T. R. Dudley, but differs by its biennial life form, longer ascendent stems, not imbricated linear cauline leaves and equally inflated fruits.  相似文献
