Summary Protoplasts isolated from embryogenic suspension cultures of European larch (Larix decidua Mill.) were cultured in thin alginate layers using a nylon mesh to enable a monitoring of the development of single cells. The patterns of cell division and differentiation are characterized and compared with zygotic embryogenesis to which homologies can only be drawn to some extent when the protoplasts grow in an auxinfree environment. Already at 2.5 M both 2,4-dichlorophenoxyacetic acid or indole-3-acetic acid cause vacuolation and elongation of individual cells, thus disturbing the process of somatic embryogenesis which generally lacks the precise quantitative patterns occurring in vivo. Prior to the formation of an embryo, a proembryonal mass develops. Oligonucleated products of spontaneous protoplast fusions are able to cellularize even without preceding karyokinesis and perform a normal embryogenic program.Abbreviations BAP

Epithelial shedding occurs in health and, extensively, in inflammatory airway diseases. This study describes deepithelialisation, reepithelialisation and associated events in guinea-pig trachea after shedding-like epithelial denudation in vivo. Mechanical deepithelialisation of an 800-m wide tracheal zone was carried out using an orotracheal steel probe without bleeding or damage to the basement membrane. Reepithelialisation was studied by scanning- and transmission electron microscopy and light microscopy. Nerve fibres were examined by immunostaining. Cell proliferation was analysed by [3H]-thymidine autoradiography. Immediately after epithelial removal secretory and ciliated (and presumably basal) epithelial cells at the wound margin dedifferentiated, flattened and migrated rapidly (2–3 m/min) over the denuded basement membrane. Within 8–15 h a new, flattened epithelium covered the entire deepithelialised zone. At 30 h a tight epithelial barrier was established and after 5 days the epithelium was fully redifferentiated. After completed migration an increased mitotic activity occurred in the epithelium and in fibroblasts/smooth muscle beneath the restitution zone. Reinnervating intraepithelial calcitonin gene-related peptide-containing nerve fibres appeared within 30 h. We conclude that (1) reproducible shedding-like denudation, without bleeding or damage to the basement membrane, can be produced in vivo; (2) secretory and ciliated cells participate in reepithelialisation by dedifferentiation and migration; (3) the initial migration is very fast in vivo; (4) shedding-like denudation may cause strong secretory and exudative responses as well as proliferation of epithelium, and fibroblasts/smooth muscle. Morphological features of the gill and opercular epithelia of tilapia (Oreochromis mossambicus) have been compared in fish acclimated to either fresh water (FW) or hypersaline water (60 S) by scanning electron and fluorescence microscopy. In hyperosmoregulating, i.e., FW-acclimated, tilapia only those mitochondria-rich (MR) cells present on the filament epithelium of the gill were exposed to the external medium. After acclimation of fish to hypersaline water these cells become more numerous, hypertrophy extensively, and form apical crypts not only in the gill filament but also in the opercular epithelium. Regardless of salinity, MR cells were never found to be exposed to the external medium on the secondary lamellae. In addition, two types of pavement cells were identified having distinct morphologies, which were unaffected by salinity. The gill filaments and the inner operculum were generally found to be covered by pavement cells with microridges, whereas the secondary lamellae were covered exclusively by smooth pavement cells. 相似文献

Laccase, a ligninolytic enzyme, was secreted by each ofthe white rot fungi Trametes versicolor and Pycnoporus cinnabarinusduring growth in a nitrogen-rich medium under agitated conditions. Afteraddition of 2-hydroxydibenzofuran to cell-freesupernatants of the cultures, yellow precipitates wereformed. These precipitates were poorly soluble in waterand therefore readily separated from the supernatant. Theproducts formed were more hydrophobic than thesubstrate, as indicated by their longer retention times on areverse phase high-performance liquid chromatographycolumn. Mass spectrometric analysis of the purifiedproducts indicated the formation of oligomers. Analysis ofthe mixture of products by gas chromatography and massspectrometry after derivatization with diazomethanesuggested the formation of at least three dimeric and ninetrimeric products. Carbon-carbon and carbon-oxygenbonds were identified in the dimers and trimers,respectively. Propionibacterium shermanii CDB 10015 was able to grow at different volumetric oxygen transfer coefficients (KLa) of 10, 22, 53h–1. These results demonstrate that this bacterium, known as anaerobic, is able to grow well under aerobic conditions. The cell biomass increased from 7.9 in anaerobic conditions to 18.3g/l at KLa 53h–1, increasing also the cell yield from 0.3 to 0.7g/g. The organic acid production pattern also changed with aeration. The acetic: propionic acid ratio increased from 0.38 in anaerobiosis to 6.25 at KLa 53h–1. The vitamin B12 production decreased from 3.1mg/l in anaerobiosis to 0.5mg/l at KLa 53h–1. 相似文献

Objective: To analyse sick leave in women at risk of primary hyperparathyroidism before its diagnosis. Design: Case-control study nested within a screened cohort of postmenopausal women. Cases were women with hyperparathyroidism without prior knowledge of their disease and no traditional symptoms or complications. Controls were women from the screened population without hyperparathyroidism. Setting: Population based screening within a Swedish community. Subject: 48 case-control pairs of women aged 55-70 years. Main outcome measure: Sick leave during the 5 years before diagnosis. Results: Total duration of sickness benefits was longer in the cases than controls, and this discrepancy included sick leave on full time or half time and for periods of longer than a week. Cases had an increased risk of sick leave more than half of the investigated time compared with controls (odds ratio 12). Doctors’ certificates showed that the overrepresented sick leave in the cases related mainly to cardiovascular diseases. Conclusion: Asymptomatic mild primary hyperparathyroidism in postmenopausal women is accompanied by a previously unrecognised morbidity, which has consequences for clinical management of the disorder and its impact on the health economy.

The importance of management regime on floristic variation (mosses and vascular plants) in four Danish beech forests was investigated. Sixty-four blocks were sampled, representing beech stands of different age and management types. Nineteen potential explanatory variables were recorded and tested with Monte-Carlo tests and Canonical Correspondence Analysis. In addition results were evaluated by use of Detrended Correspondence Analysis. Explanatory variables were divided into three groups; soil, microclimatic and management parameters. The amount of variation explained by each group of variables was calculated by use of variation partitioning. The group consisting of management variables explained most variation, on local as well as regional scale. Management related variables explained more variation in vegetation than any other variables together. This indicates the importance of management as determining species composition in Danish beech forests. Management related variables explained most variation on local scale. On a regional scale, soil parameters explained the major part of the variation. The results suggest that thirty years without management are sufficient to change species composition significantly, as compared to managed forests. 相似文献

In a continued investigation of lecithin:cholesterol acyltransferase reaction with micellar, discoidal complexes of phosphatidylcholine (PC) . cholesterol . apolipoprotein A-I (apo-A-I), we prepared well defined complexes with variable free cholesterol contents and examined their reactivity with purified enzyme. The complexes, prepared by the sodium cholate dialysis method, were fractionated into "small" and "large" classes by gel filtration of the reaction mixtures through a Bio-Gel A-5m column. The small complexes had egg-PC/cholesterol/apo-A-I molar ratios from 68:14:1 to 80:1:1, discoidal shapes with diameters around 114 (+/- 13) A and widths of 42 A by electron microscopy, and Stokes radii from 47 to 49 A corresponding to molecular weights near 2 X 10(5). The corresponding properties of the large complexes, isolated from samples with higher cholesterol contents, were egg-PC/cholesterol/apo-A-I molar ratios from 84:26:1 to 96:17:1, diameters of 161 (+/- 20) A, widths of 43 A, Stokes radii around 80 A, and estimated molecular weights in the vicinity of 5 X 10(5). Both types of complexes, when adjusted to equal apo-A-I concentrations, gave essentially identical initial reaction velocities with purified lecithin:cholesterol acyltransferase over a wide range of cholesterol concentrations (from 2 X 10(-7) to 4 X 10(-4) M), PC/cholesterol molar ratios (from 3:1 to 12:1), and quite different lipid fluidity conditions as detected by diphenylhexatriene fluorescence polarization. When complexes were adjusted to a constant cholesterol concentration, the initial velocities of the lecithin:cholesterol acyltransferase reaction followed Michaelis-Menten kinetics relative to the apo-A-I concentrations. Arrhenius plots of initial reaction rates for various complexes with variable cholesterol content and fluidity, measured at constant apo-A-I concentrations, gave identical temperature dependences with an average activation energy of 18.0 kcal/mol. These results strongly suggest that the cholesterol esterification on high density lipoprotein particles does not depend on their unesterified-cholesterol contents, PC/unesterified-cholesterol molar ratios, nor on the fluidity of their lipid domains. 相似文献

Structure and composition of woody vegetation in Sahelian forest types (not including riparian and riverine types) were studied in Sénégal. The application of classification and ordination resulted in the recognition of seven distinctive woody community types, related to soil and precipitation gradients. The described vegetation types only contained four strictly Saharan species, the rest being of Sahelian and Soudanean affinity, so a floristic change from savanna to desert vegetation after two decades of drought could not be confirmed. Tree density was lower in the northern part than earlier, possibly an indication of degradation. Rejuvenation was greatest at the northernmost arenosolic stations, but low in the southern, ferrouginous areas. This presumably demonstrates the resilience of North-Sahelian vegetation, at the moment being in a dynamic rejuvenation phase, after a period of adverse conditions. 相似文献

Effect of dipalmitoylphosphatidylcholine vesicle curvature on the reaction with human apolipoprotein A-I 总被引:1,自引:0,他引:1
Large unilamellar vesicles of dipalmitoylphosphatidylcholine (DPPC) were prepared by sonication and were fractionated by gel filtration on Sepharose Cl-2B in the size range from 180- to 380-A Stokes radii. Negatively stained electron micrographs of these preparations indicated the presence of unilamellar, spheroidal structures of the expected size. Fluorescence polarization of diphenylhexatriene, dissolved in the vesicles, revealed progressively broader phase transitions, shifted to lower temperatures for vesicles of decreasing sizes. The fractionated unilamellar vesicles and multilamellar vesicles of DPPC were reacted with human apolipoprotein A-I at 41 degrees C for periods from 1 to 120 h. The reaction mixtures were then passed through a Bio-Gel A-5m column to separate unreacted lipid vesicles and protein from micellar complexes of DPPC with apolipoprotein A-I. Smaller vesicles were much more reactive than larger vesicles or multilamellar vesicles with the apolipoprotein. This difference in reactivity was explained by the increasing bilayer curvature of smaller vesicles which changes the packing of DPPC molecules in the bilayer and facilitates its penetration by the apolipoprotein. 相似文献
