Characterization of L-threonine deaminase activity of guinea pig liver induced by a high protein diet

In a foregoing paper, we demonstrated that under equilibrated diet conditions, guinea pig liver L-threonine deaminase activity should be allocated to two distinct enzymes: a specific L-threonine deaminase without activity toward L-serine and a L-serine deaminase having a secondary activity toward L-threonine. In the present work, we observed that a high protidic diet caused an elevation of total threonine deaminase activity. Thus purification of guinea pig liver L-threonine deaminase was attempted, using ultracentrifugation, salt precipitation, heat treatment, ion exchange chromatography on DEAE Sephacel, Sephadex G 200 molecular sieve, 2 amino-2 methyl-1 propanol linked CH 4B Sepharose chromatography. The weak variations of the ratios of specific activities respectively toward L-threonine and L-serine observed at each stage of the purification procedure indicated that both activities are very likely supported by a single enzyme preexisting in the liver of guinea pigs fed an equilibrated diet. No isoenzyme was evidenced by polyacrylamide gel electrophoresis or DEAE Sephacel chromatography. Moreover, our purification procedure demonstrated that not only inducible L-threonine deaminase guinea pig liver activity was due to L-serine deaminase, but also that an initially existing specific L-threonine deaminase activity paradoxically disappeared with a protein rich diet.     

Early-replication DNA patterns in a derivative chromosome in a syrian hamster with only 42 chromosomes

From crosses within a 2n = 43 line of Syrian hamsters (Mesocricetus auratus) lacking one derivative (der 11) of an 11;20 reciprocal translocation we have obtained homozygotes with only 42 chromosomes These animals are homozygous deficient (nullisomic) for the centromere and short arm of chromosome 11 and for the bulk of the long arm of chromosome 20. — During cytogenetic studies, we investigated the frequency patterns of early-replicating bands in the surviving derivative (der 20) at two cytologically defined sub-phases of S using short-term fibroblast cultures. These patterns were compared with those observed in the component, untranslocated arms in normal 2n = 44 cells at the same two sub-phases. — Very close agreement was found, indicating that neither the nullisomy, nor the new arm combination has interfered detectably with the pattern or programme of early band replication.     

The strategy of “Useful Sun” improves physical endurance and adaptation of cardiac morphology

The action of solar light transformed by special screens has been studied on CD-1 male mice. In the active control group, mice were irradiated through screens absorbing the UV-component. In the experimental group, screens transforming the UV-component into the orange-red light were used. In the active control, changes in the swimming activity, as compared to the same parameter before irradiation, were manifested much less than in animals of the experimental group. A morphological analysis showed changes in the structure of all cardiomyocyte organelles studied: the relative area of mitochondria in the experimental mice increased by more than 20% compared to intact animals (p < 0.05). A significant increase in the area of the sarcoplasmic reticulum, by 23.4% (p < 0.05), and in the volume of the myofibrillar apparatus, by 19.4% (p < 0.05), was detected. The results of our experiment show that the irradiation with using an additional orangered component improves the physical endurance 1.5 times and initiates morphogenetic processes in cardiac muscle cells.     

Therapeutic effect of the transfusion of UV-irradiated donor plasma in mechanical jaundice

The method of intravenous administration of UV-irradiated donor plasma to patients with jaundice of different genesis has been tested in the experiments on rats and used clinically in 20 patients. Biochemical and morphological studies have shown that such treatment causes stable improvement in the liver functional state.
