Female competition and reproductive success in northern elephant seals

The probability of weaning a healthy pup increases with age in female northern elephant seals, Mirounga angustirostris. On Año Nuevo Island, California, weaning success among ‘prime’ females, those 6 years of age or older, was more than double that of ‘young’ females, those 3 to 5 years old. Prime females were better mothers than young females because of superior size, higher social dominance, and greater maternal experience; they were more likely to mate with high-ranking males and gave birth at an optimal time and place, circumstances that maximized the probability that their pups would survive, develop, and reproduce. The competitive advantage of prime-age mothers over younger ones was greatest when female and pup density was high. Young females improved their chances of reproducing successfully by emigrating from crowded harems and establishing new colonies.     

Glucocorticoid binding to solubilized components of human placental trophoblast membranes

We have recently described glucocorticoid uptake by human placental membrane vesicles which is specific, saturable and has a low K (7 nM). This paper describes solubilization of these vesicles with Triton x-100 and successful demonstration of glucocorticoid binding to the putative transport site. This was accomplished by analysis of corticosterone binding to the 140,000 × g supernatant of solubilized vesicles using G-75 Sephadex chromatography. The amount of bound corticosterone present in the void volume was proportional to the concentration of solubilized vesicles. The specificity of binding was tested by coincubation of tritiated corticosterone with 100-fold excesses of various steroids. The relative ability of various steroids to inhibit binding was corticosterone=pregesterone- >aldosterone. Triamcinolone acetonide, and estradiol were ineffective competitors. We conclude from these studies that human placental membranes contain glucocorticoid-specific binding sites which can be solubilized with Triton x-100. It is possible that these sites are involved in membrane mediated transport of glucocorticoids by this tissue.     

Presynaptic dense bars at neuromuscular synapses of the lobster,Homarus americanus

Summary The threedimensional ultrastructure of presynaptic dense bars was examined by serial section electron microscopy in the excitatory neuromuscular synapses of the accessory flexor muscle in the limbs of larval, juvenile, and adult lobsters. The cross-sectional profile of the dense bar resembles an asymmetric hourglass, the part contacting the presynaptic membrane being larger than that projecting into the terminal. The bar has a height of 55–65 nm and varies in length from 75–600 nm. In its dimensions it resembles the dense projections in the synapses of the CNS of insects and vertebrates. The usual location of these dense bars is at well defined synapses, though a few are found at extrasynaptic sites either in the axon or terminal. In the latter case the bars are close to synapse-bearing regions, particularly in the larval terminals, suggesting that the extrasynaptic bars denote early events in synapse formation. In all cases the bars are intimately associated with electron lucent, synaptic vesicles located on either side, in the indentation of its hourglass-shaped cross sectional profile. The vesicles occur along the length of the bar and contact the presynaptic membrane. Consequently the dense bar may serve to align the vesicles at the presynaptic membrane prior to exocytosis. A similar role has been suggested for the presynaptic dense bodies at the neuromuscular junction of the frog, where synaptic vesicles form a row on either side of this structure.Supported by Muscular Dystrophy Association of Canada and NSERCC. Rat testis immunoreactive LH-RH differs structurally from hypothalamic LH-RH

Luteinizing hormone releasing hormone immunoreactivity (LH-RH-IR) has been identified in acetic acid extracts of adult rat testes and partially purified by immunoaffinity chromatography. On Sephadex G-100 this material separated into four major peaks of >100K, ～32K, ～5K and ≤4K daltons. The ≤4K peak of LH-RH-IR eluted later than synthetic hypothalamic LH-RH decapeptide on Sephadex G-25. Antibody binding studies on the various LH-RH-IR species with antisera specific for different regions of synthetic LH-RH decapeptide indicate that all the testicular LH-RH-IR molecules have C-terminal immunological homology with the hypothalmic decapeptide but differ towards the N-terminus of the decapeptide sequence.     

Radiosensitization of hypoxic mammalian cells in vitro by some 5-substituted-4-nitroimidazoles

The efficiencies of various 5-substituted-4-nitroimidazoles as radiation sensitizers have been determined in hypoxic Chinese hamster cells irradiated in vitro. Compared with published data on the sensitizing properties of substituted 2-nitro- and 5-nitroimidazoles, some of the 4-nitro derivatives show unusually high sensitizing efficiencies defined as the concentrations required to give an enhancement ratio of 1.6. The equilibrium one-electron reduction potentials of the compounds (E17) were measured by a pulse radiolysis technique and the results show that although sensitizing efficiencies are unexpectedly high, based on considerations of electron affinity, they still increase with increasing values of E17. Enhancement ratios were determined in two V79 cell lines for combinations of one of these compounds (a 4-nitroimidazole containing the group SO2.O.phenyl in the 5-position, NSC 38087) with various concentrations of misonidazole. The sensitization observed suggests that the two compounds may be operating by different mechanisms.     

Microfilament bundles in antheridial nuclei of Achlya ambisexualis E 87

This is the first report of intranuclear microfilaments within gametangial nuclei of oömycetous fungi. Longitudinal sections of four to six microfilaments were frequently observed in meiotic antheridial nuclei of Achlya ambisexualis. Each microfilament measured approximately 7–10 nm in diameter. Spindle tubules (25 nm in diameter) were also observed within some of the nuclei possessing microfilaments.     

Nonautotrophic Thiobacillus in Acid Mine Water

Nonautotrophic thiobacilli were isolated from the acidic water of a coal mine. Based on their mixotrophic physiology, the isolates are regarded as strains of Thiobacillus perometabolis.
