Structural investigations on the lipopolysaccharide isolated from Vibrio cholera,Inaba 569 B

On hydrolysis, the purified lipopolysaccharide (LPS) isolated from Vibrio cholera, Inaba 569 B, yielded glucose, mannose, a heptose behaving like d-glycero-l-manno-heptose and one behaving like d-glycero-l-gluco-heptose, 2-amino-2-deoxy-glucose, and glucuronic acid in the molar ratios of ～9:4:5:1:2:5. Studies on the LPS, the polysaccharide (PS), and carboxyl-reduced LPS showed that the PS has a branched structure, with (1→2)-linked mannopyranosyl and a heptopyranosyl, and (1→4)-linked glucopyranosyluronic and 2-amino-2-deoxyglucopyranosyl residues in the interior part of the molecule, and glucopyranosyl and heptopyranosyl residues as nonreducing end-groups.     

Dormancy studies in black gram (Phaseolus mnngo L.)

Mechanical scarification of seeds ofPhaseolus mungo L. resulted in 100 per cent gormination in comparison to 90 per cent dormant control. Seeds treated with concentrated sulphuric acid for 8 min led to 85 per cent germination. Heat treatment at 70 °C for 24 h exhibited 75 per cent gormination, while boiled water treatment for 7 min caused 65 per cent germination.     

Purification of and mechanism studies on citrate synthase. Use of biospecific adsorption-elution techniques.

Citrate synthases from animal tissues were found to bind to Sepharose-"ATP." A pure preparation of citrate synthase was obtained from a crude fraction of rat heart by the specific elution of the enzyme from the Sepharose-"ATP" with the dead end complex-forming substrates, oxalacetate and CoA. The proposed mechanisms of citrate synthase, obtained from steady state kinetics, were examined in light of the elution pattern of the enzyme obtained using combinations of substrates and substrate analogs.     

Activation of pyruvate dehydrogenase in adipose tissue by insulin. Evidence for an effect of insulin on pyruvate dehydrogenase phosphate phosphatase.

1. The mechanism by which insulin activates pyruvate dehydrogenase in rat epididymal adipose tissue was further investigated. 2. When crude extracts, prepared from tissue segments previously exposed to insulin (2m-i.u/ml) for 2min, were supplemented with Mg-2+, Ca-2+, glucose and hexokinase and incubated at 30 degrees C, they displayed an enhanced rate of increase in pyruvate dehydrogenase activity compared with control extracts. 3. When similar extracts were instead supplemented with fluoride, ADP, creatine phosphate and creatine kinase, the rate of decrease in pyruvate dehydrogenase activity observed during incubation at 30 degrees C was unaffected by insulin treatment. 4. It is suggested that insulin increases the fraction of pyruvate dehydrogenase present in the tissue in the active dephospho form by increasing the activity of pyruvate dehydrogenase phosphate phosphatase.     

Newer applications of the histological stain prepared from Pterocarpus santalinus

A histological stain prepared from the heartwood of Pterocarpus santalinus Linn. has been found to be an excellent nuclear stain for various cells of animal and plant origin. As an elastic tissue stain, the results are comparable to standard elastic tissue stains. The striations of voluntary muscle fibers are well shown. The Nissl granules and fibers of cranial nerves in the pons are visualized. When counterstained with light green, it differentially stains muscle and fibrous tissue. The stain can be used as counterstain with certain histochemical procedures with satisfactory results. The preparation and use of this versatile stain are described.
