Objectives
Low levels of high-density lipoprotein (HDL) cholesterol are associated with an increased risk of acute myocardial infarction possibly through impaired endothelial atheroprotection and decreased nitric oxide (NO) bioavailability. Asymmetric dimethylarginine (ADMA) mediates endothelial function by inhibiting nitric oxide synthase activity. In patients with acute myocardial infarction, we investigated the relationship between serum levels of HDL and ADMA.Approach and Results
Blood samples from 612 consecutive patients hospitalized for acute MI <24 hours after symptom onset were taken on admission. Serum levels of ADMA, its stereoisomer, symmetric dimethylarginine (SDMA) and L-arginine were determined using high-performance liquid chromatography. Patients with low HDL (<40 mg/dL for men and <50 mg/dL for women) were compared with patients with higher HDL. Most patients (59%) had low HDL levels. Median ADMA levels were markedly higher in the low HDL group (0.69 vs. 0.50 µmole/L, p<0.001). In contrast, SDMA and L-arginine levels were similar for the two groups (p = 0.120 and p = 0.064). Notably, ADMA, but not SDMA or L-arginine, was inversely correlated with HDL (r = −0.311, p<0.001). In stratified analysis, this relationship was only found for low HDL levels (r = −0.265, p<0.001), but not when HDL levels were higher (r = −0.077, p = 0.225). By multivariate logistic regression analysis, ADMA level was strongly associated with low HDL levels (OR(95%CI):6.06(3.48–10.53), p<0.001), beyond traditional confounding factors.Conclusions
Our large population-based study showed for the first time a strong inverse relationship between HDL and ADMA in myocardial infarction patients, suggesting a functional interaction between HDL and endothelium, beyond metabolic conditions associated with low HDL levels. 相似文献92.
Three middle Eocene localities (Silica North, Silica South, Black Crow) recently discovered in Namibia have produced terrestrial faunas that rank among the few known from the period of insulation of Africa (Aptian-early Miocene). Collectively, the three localities have yielded anuran amphibians (one pipid frog, the earliest assemblage [three taxa] of ranoid frogs in Africa, one indeterminate family) and squamate reptiles (an amphisbaenian ‘lizard’, a snake that likely represents a colubroid, and two indeterminate ‘lizards’). These Eocene faunas suggest that ranoids, colubroids and African pipids are autochthonous to Africa. However, whereas pipids are vicariants inherited from West Gondwana, ranoids and colubroids (if really autochthonous) originated in Africa from unknown stems. Background and aims
Urea is the major nitrogen (N) form supplied as fertilizer in agriculture. However, urease, a nickel-dependent enzyme, allows plants to use external or internally generated urea as a nitrogen source. Since a urease inhibitor is frequently applied in conjunction with urea fertilizer, the N-metabolism of plants may be affected. The aim of this study was to determine physiological and molecular effects of nickel deficiency and a urease inhibitor on urea uptake and assimilation in oilseed rape.Methods
Plants were grown on hydroponic solution with urea as the sole N source under three treatments: plants treated with nickel (+Ni) as a control, without nickel (?Ni) and with nickel and phenylphosphorodiamidate (+Ni+PPD). Urea transport and assimilation were investigated.Results
The results show that Ni-deficiency or PPD supply led to reduced growth and reduced 15N-uptake from urea. This effect was more pronounced in PPD-treated plants, which accumulated high amounts of urea and ammonium. Thus, Ni-deficiency or addition of PPD, limit the availability of N and decreased shoot and root amino acid content. The up-regulation of BnDUR3 in roots indicated that this gene is a component of the stress response to nitrogen-deficiency. A general decline of glutamine synthetase (GS) activity and activation of glutamate dehydrogenase (GDH) and increases in its expression level were observed in control plants. At the same time, in (?N) or (+Ni+PPD) treated plants, no increases in GS or GDH activities and expression level were found.Conclusions
Overall results showed that plants require Ni as a nutrient (while most widely used nutrient solutions are devoid of Ni), whether they are grown with or without a urea supply, and that urease inhibitors may have deleterious effects at least in hydroponic grown oilseed rape. 相似文献94.

Aims
This work examines Zn accumulation in four Anthyllis vulneraria subspecies supplemented with mineral nitrogen or grown in the presence of their symbiotic bacteria.Methods
Anthyllis vulneraria subspecies were grown hydroponically in the presence of high levels of ZnSO4. The plants were either grown in symbiosis with one of two non-metallicolous or metallicolous Mesorhizobium inoculants or in the presence of KNO3.Results
When exposed to 1,000 μM Zn, shoot and root biomass of three out of our four Anthyllis subspecies cultivated with NO3 dropped significantly by about 24–28 %; carpatica, the fourth subspecies, was not affected. Subspecies carpatica Zn tolerance was confirmed when in symbiosis with the metallicolous strain. In the presence of 1,000 μM Zn, the different Anthyllis subspecies concentrated more Zn in their roots than in their shoots and only subsp. carpatica accumulated a significant amount of Zn in its shoots. The most remarkable feature was the drastic decrease in Zn concentration in both roots (up to 2.5–3 fold) and shoots (2.6-fold) of subsp. carpatica exposed to 1,000 μM Zn and nodulated whatever the Mesorhizobium strain used, compared to the N-grown plants.Conclusions
Our results bring new perspectives as regards phytostabilization, with the potential use of a rhizobium-inoculated leguminous subspecies displaying unusual Zn tolerance. 相似文献95.

The melanoma cell adhesion molecule (CD146) contains a circulating proteolytic variant (sCD146), which is involved in inflammation and angiogenesis. Its circulating level is modulated in different pathologies, but its intracellular transduction pathways are still largely unknown. Using peptide pulldown and mass spectrometry, we identified angiomotin as a sCD146-associated protein in endothelial progenitor cells (EPC). Interaction between angiomotin and sCD146 was confirmed by enzyme-linked immunosorbent assay (ELISA), homogeneous time-resolved fluorescence, and binding of sCD146 on both immobilized recombinant angiomotin and angiomotin-transfected cells. Silencing angiomotin in EPC inhibited sCD146 angiogenic effects, i.e. EPC migration, proliferation, and capacity to form capillary-like structures in Matrigel. In addition, sCD146 effects were inhibited by the angiomotin inhibitor angiostatin and competition with recombinant angiomotin. Finally, binding of sCD146 on angiomotin triggered the activation of several transduction pathways that were identified by antibody array. These results delineate a novel signaling pathway where sCD146 binds to angiomotin to stimulate a proangiogenic response. This result is important to find novel target cells of sCD146 and for the development of therapeutic strategies based on EPC in the treatment of ischemic diseases. 相似文献

The chicken major histocompatibility complex (MHC) is located on the microchromosome 16 and is described as the most variable region in the genome. The genes of the MHC play a central role in the immune system. Particularly, genes encoding proteins involved in the antigen presentation to T cells. Therefore, describing the genetic polymorphism of this region is crucial in understanding host–pathogen interactions. The tandem repeat LEI0258 is located within the core area of the B region of the chicken MHC (MHC-B region) and its genotypes correlate with serology. This marker was used to provide a picture of the worldwide diversity of the chicken MHC-B region and to categorize chicken MHC haplotypes. More than 1,600 animals from 80 different populations or lines of chickens from Africa, Asia, and Europe, including wild fowl species, were genotyped at the LEI0258 locus. Fifty novel alleles were described after sequencing. The resulting 79 alleles were classified into 12 clusters, based on the SNPs and indels found within the sequences flanking the repeats. Furthermore, hypotheses were formulated on the evolutionary dynamics of the region. This study constitutes the largest variability report for the chicken MHC and establishes a framework for future diversity or association studies. 相似文献

Three bumblebee species, foraging on Aconitum spp. have been commonly observed in Eyne (France, East Pyrénées): Bombus gerstaeckeri, B. hortorum and B. wurflenii. We estimated the population of these three species. For B. hortorum and B. wurflenii, the total workers populations foraging on Aconitum spp. ranged from 101 to 523 and 156 to 270, respectively. These two species also forage on other plants while B. gerstaeckeri visits only Aconitum spp. The population of B. gerstaeckeri observed was extremely small, founded by 33 queens only in 2001. With a total number of workers estimated from 40 to 102, the observed workers/queens ratio, 1 to 3 workers for each queen, is very unusual for a eusocial species. Also we observed queens foraging during the whole life of the colony. This kind of social organisation is similar to that of some high arctic species. Summary. Taxonomic update and geographic distribution of fleas of the genus Ctenophthalmus Kolenati 1856 in the Western Palearctic Region (Insecta: Siphonaptera: Ctenophthalmidae). Among fleas (Siphonaptera), the genus Ctenophthalmus is the one that comprises the largest number of taxa and is also characterized by a large geographical range. Here, we present a taxonomic revision of the Western Paleartic subgenera, groups, species and subspecies. We recognized a total of 143 taxa (57 species and 86 subspecies). These taxa are clustered into 23 groups of species, which fall into seven of the 16 subgenera of the genus Ctenophthalmus. According to Hopkins & Rothschild (1966), the subgenus Ctenophthalmus would only include the agyrtes group, itself divided into subgroups. We decided to raise these subgroups to group status to clarify taxonomic relationships within the subgenus Ctenophthalmus. Within this subgenus, the arvernus group is renamed baeticus, the fransmiti group is confirmed, and the egregius group is created. Dispersal is critically linked to the demographic and evolutionary trajectories of populations, but in most seabird species it may be difficult to estimate. Using molecular tools, we explored population structure and the spatial dispersal pattern of a highly pelagic but philopatric seabird, the Cory''s shearwater Calonectris diomedea. Microsatellite fragments were analysed from samples collected across almost the entire breeding range of the species. To help disentangle the taxonomic status of the two subspecies described, the Atlantic form C. d. borealis and the Mediterranean form C. d. diomedea, we analysed genetic divergence between subspecies and quantified both historical and recent migration rates between the Mediterranean and Atlantic basins. We also searched for evidence of isolation by distance (IBD) and addressed spatial patterns of gene flow. We found a low genetic structure in the Mediterranean basin. Conversely, strong genetic differentiation appeared in the Atlantic basin. Even if the species was mostly philopatric (97%), results suggest recent dispersal between basins, especially from the Atlantic to the Mediterranean (aprox. 10% of migrants/generation across the last two generations). Long-term gene flow analyses also suggested an historical exchange between basins (about 70 breeders/generation). Spatial analysis of genetic variation indicates that distance is not the main factor in shaping genetic structure in this species. Given our results we recommend gathering more data before concluded whether these taxa should be treated as two species or subspecies. 相似文献
