Atheroma formation and restenosis following percutaneous vascular intervention involve the growth and migration of vascular smooth muscle cells (SMCs) into neointimal lesions, in part due to changes in the extracellular matrix. While some clinical studies have suggested that, in comparison to non-diabetics, β3 integrin inhibition in diabetic patients confers protection from restenosis, little is known regarding the role of β3 integrin inhibition on SMC responses in this context. To understand the molecular mechanisms underlying integrin-mediated regulation of SMC function in diabetes, we examined SMC responses in diabetic mice deficient in integrin β3 and observed that the integrin was required for enhanced proliferation, migration and extracellular regulated kinase (ERK) activation. Hyperglycemia-enhanced membrane recruitment and catalytic activity of PKCβ in an integrin β3-dependent manner. Hyperglycemia also promoted SMC filopodia formation and cell migration, both of which required αVβ3, PKCβ, and ERK activity. Furthermore, the integrin–kinase association was regulated by the αVβ3 integrin ligand thrombospondin and the integrin modulator Rap1 under conditions of hyperglycemia. These results suggest that there are differences in SMC responses to vascular injury depending on the presence or absence of hyperglycemia and that SMC response under hyperglycemic conditions is largely mediated through β3 integrin signaling.  相似文献

The autoinducer (4S)-4,5-dihydroxypentane-2,3-dione ((S)-DPD, AI-2) facilitates chemical communication, termed ‘quorum sensing’, amongst a wide range of bacteria, The synthesis of (S)-DPD is challenging in part due to its instability. Herein we report a novel synthesis of (S)-DPD via (2S)-2,3-O-isopropylidene glyceraldehyde, through Wittig, dihydroxylation and oxidation reactions, with a complimentary asymmetric synthesis to a key precursor. Its enantiomer (R)-DPD, was prepared from d-mannitol via (2R)-2,3-O-isopropylideneglyceraldehyde. The synthesized enantiomers of DPD have AI-2 bioluminescence-inducing properties in the Vibrio harveyi BB170 strain.  相似文献

Telomeres and telomerase appear to participate in the repair of broken DNA ends produced by oxidative damage. Arsenite is an environmental contaminant and a potent human carcinogen, which induces oxidative stress on cells via the generation of reactive oxygen species affecting cell viability and chromosome stability. It promotes telomere attrition and reduces cell survival by apoptosis. In this study, we used mouse embryonic fibroblasts (MEFs) from mice lacking telomerase RNA component (mTERC(-/-) mice) with long (early passage or EP) and short (late passage or LP) telomeres to investigate the extent of oxidative damage by comparing the differences in DNA damage, chromosome instability, and cell survival at 24 and 48 h of exposure to sodium arsenite (As3+; NaAsO2). There was significantly high level of DNA damage in mTERC(-/-) cells with short telomeres as determined by alkaline comet assay. Consistent with elevated DNA damage, increased micronuclei (MN) induction reflecting gross genomic instability was also observed. Fluorescence in situ hybridization (FISH) analysis revealed that increasing doses of arsenite augmented the chromosome aberrations, which contributes to genomic instability leading to possibly apoptotic cell death and cell cycle arrest. Microarray analysis has revealed that As3+ treatment altered the expression of 456 genes of which 20% of them have known functions in cell cycle and DNA damage signaling and response, cell growth, and/or maintenance. Results from our studies imply that short dysfunctional telomeres impair the repair of oxidative damage caused by arsenite. The results will have implications in risk estimation as well as cancer chemotherapy.  相似文献

Discharge of wastewater from electroplating and leather industries is a major concern for the environment due to the presence of toxic Cr6+ and other ions, such as sulfate, nitrate, phosphate, etc. This study evaluated the potential of Tradescantia pallida, a plant species known for its Cr bioaccumulation, for the simultaneous removal of Cr6+, SO42?, NO3?, and PO43?. The effect of different co-ions on Cr6+ removal by T. pallida was examined following the Plackett-Burman design of experiments carried out under batch hydroponics conditions. The results revealed a maximum removal of 84% Cr6+, 87% SO42?, 94% NO3? and 100% PO43? without any phytotoxic effect on the plant for an initial Cr6+ concentration in the range 5–20 mg L?1. SO42? and NO3? enhanced Cr uptake at a high initial Cr concentration (20 mg L?1), whereas PO43? did not affect Cr uptake both at high and low initial Cr concentrations. The Cr6+ removal kinetics in the presence of different ions was well described by the pseudo-second-order kinetic model which revealed that both biosorption and bioaccumulation of the metal played an important role in Cr6+ removal. Increase in the total carbohydrate and protein content of the plant following Cr6+ and co-ions exposure indicated a good tolerance of the plant toward Cr6+ toxicity. Furthermore, enhancement in the lipid peroxidation and catalase activity in T. pallida upon Cr6+ exposure revealed a maximum stress-induced condition in the plant. Overall, this study demonstrated a very good potential of the plant T. pallida for Cr6+ removal from wastewater even in the presence of co-ions.  相似文献

IgG antibody to Mycobacterium tuberculosis from the sera of patients with 'definite' pulmonary tuberculosis (PT) was isolated and coupled with Cyanogen bromide-Sepharose 4B. Using an immunoabsorbent affinity chromatography, 14 kDa antigen was recovered from the culture filtrates of M. tuberculosis. With this mycobacterial antigen, a dot immunobinding assay (Dot-Iba) was developed for the detection of specific antibody to M. tuberculosis in the sera of patients with PT and controls. The assay gave positive results in all the 12 sputum-smear positive [acid fast bacilli (AFB)] patients with PT and gave negative results in the 50 sera from control groups. The Dot-Iba as described in this study, is simple, rapid and specific for laboratory diagnosis of PT.  相似文献

Several anesthetics are known to cause respiratory and cardiovascular depression in humans and animals; but, these diverse effects have not been extensively investigated in laboratory rodents. The objective of this study is to choose a suitable anesthetic combination for use in surgical models eg. coronary artery ligation in rats. Male Wistar rats were anesthetized with three different drugs viz. diazepam-ketamine (DK) (2.5 mg/Kg, intraperitoneally (i.p); 50 mg/Kg, i.p), xylazine-ketamine (XK) (5 mg/Kg i.p; 50 mg/Kg i.p) and thiopentone (T) (40 mg/Kg i.p) and the respiratory and cardiovascular functions were assessed after coronary artery ligation. Heart rate (HR), mean arterial pressure (MAP), partial pressure of carbon dioxide (PaCO2), partial pressure of oxygen (PaO2), oxygen saturation percentage (O2 sat (%)), arterial blood pH and rectal body temperature were studied in detail. During the anesthetic regime, HR was lower till 60 min in XK and T ligated group (333 +/- 6; 304 +/- 8 beats/min) and it was near normalcy in the case of DK ligated group (394 +/- 6 beats/min). Significant respiratory depression was particularly reflected in the T ligated group with an increase in PaCO2 at 30 min (40.32 +/- 2.64 mmHg), which decreased to 38.2 +/- 2.23 mmHg at 60 min. Throughout the investigation, DK showed the least overall effects compared to XK and T on respiratory functions. Thus, DK could be considered to be a suitable anesthetic for use in a surgical model such as coronary artery ligation in albino rats.  相似文献

The EPR and magnetic Mössbauer spectra of a series of axial ligand complexes of tetrakis(2,6-dimethoxyphenyl)porphyrinatoiron(III), [(2,6-(OMe)2)4TPPFeL2]+, where L=N-methylimidazole, 2-methylimidazole, or 4-(dimethylamino)pyridine, of one axial ligand complex of tetraphenylporphyrin, the bis(4-cyanopyridine) complex [TPPFe(4-CNPy)2]+, and of one axial ligand complex of tetraphenylchlorin, [TPCFe(ImH)2]+, where ImH=imidazole, have been investigated and compared to those of low-spin Fe(III) porphyrinates and ferriheme proteins reported in the literature. On the basis of this and previous complementary spectroscopic investigations, three types of complexes have been identified: those having (dxy)2(dxz,dyz)3 electronic ground states with axial ligands aligned in perpendicular planes (Type I), those having (dxy)2(dxz,dyz)3 electronic ground states with axial ligands aligned in parallel planes (Type II), and those having the novel (dxz,dyz)4(dxy)1 electronic ground state (Type III). A subset of the latter type, with planar axial ligands aligned parallel to each other or strong macrocycle asymmetry that yield rhombic EPR spectra, cannot be created using the porphyrinate ligand. Type I centers are characterized by "large gmax" EPR spectra with g>3.2 and well-resolved, widely spread magnetic Mössbauer spectra having Azz/gNN>680 kG, with Axx negative in sign but much smaller in magnitude than Azz, while Type II centers have well-resolved rhombic EPR spectra with gzz=2.4–3.1 and also less-resolved magnetic Mössbauer spectra, and usually have Azz/gNN in the range of 440–660 kG (but in certain cases as small as 180 kG) and Axx again negative in sign but only somewhat smaller (but occasionally larger in magnitude) than Azz, and Type III centers have axial EPR spectra with g2.6 or smaller and g<1.0–1.95, but often not resolved, and less-resolved magnetic Mössbauer spectra having Azz/gNN in the range of 270–400 kG, and Axx again negative in sign but much smaller in magnitude than Azz. An exception to this rule is [TPPFe(4-CNPy)2]+, which has Axx/gNN=–565 kG, Ayy/gNN=629 kG, and Azz/gNN=4 kG. A subset of Type II complexes (Type II) have rhombicities (V/) much greater than 0.67 and Azz/gNN ranging from 320 to 170 kG, with Axx also negative but with the magnitude of Axx significantly larger than that of Azz. These classifications are also observed for a variety of ferriheme proteins, and they lead to linear correlations between Azz and either Axx, gzz, or V/ for Types I and II (but not for Azz versus V/ for Type II). Efficacy of Butea monosperma on dermal wound healing in rats   总被引:2,自引:0,他引:2  
Wound healing occurs as a fundamental response to tissue injury. Several natural products have been shown to accelerate the healing process. The present investigation was undertaken to determine the efficacy of topical administration of an alcoholic bark extract of Butea monosperma (B. monosperma) on cutaneous wound healing in rats. Full-thickness excision wounds were made on the back of rat and B. monosperma extract was administered topically. The granulation tissue formed on days 4, 8, 12 and 16 (post-wound) was used to estimate total collagen, hexosamine, protein, DNA and uronic acid. The extract increased cellular proliferation and collagen synthesis at the wound site, as evidenced by increase in DNA, total protein and total collagen content of granulation tissues. The extract treated wounds were found to heal much faster as indicated by improved rates of epithelialization and wound contraction, also confirmed by histopathological examinations. Also, the tensile strength of drug-treated wounds was increased significantly. In addition, we show that B. monosperma possesses antioxidant properties, by its ability to reduce lipid peroxidation. The results clearly substantiate the beneficial effects of the topical application of B. monosperma in the acceleration of wound healing.  相似文献

Development of roosting patterns under a limited resource was studied in the short-nosed fruit bat C. sphinx in captivity. Spatial fidelity during the resting period (day time) and the individual male bat's presence/absence in the roost (occupancy index) were estimated during the active period (night time). Results show the presence of three groups on the basis of spatial fidelity. The first group was associated with the tent consisting of a harem male and seven females. The second group stayed near to the harem. The third group consisting of two males showed little occupancy index and no spatial fidelity. Female turnover between the first and second groups, and harem male replacement were observed. These findings of male groupings and female loyalty on the basis of "resource", suggest that resource defence polygyny is the primary mating strategy in C. sphinx.  相似文献
