Mitochondria transfer into mouse ova by microinjection   总被引:10,自引:0,他引:10

A method for mitochondria isolation and interspecific transfer of mitochondria was developed in mice. Mitochondria were isolated from Mus spretus liver samples for microinjection into fertilized ova obtained from superovulated M. musculus domesticus females. Electron microscopic observations of mitochondria preparations used for microinjection demonstrated intact mitochondrial vesicles with little microsomal contamination. Species-specific nested PCR primers complementary to sequence differences in the mitochondrial DNA D-loop region revealed high rates of successful transfer of foreign mitochondria after isolation and injection into zygotes cultured through the blastocyst stage of embryonic development. Of 217 zygotes, 67 survived mitochondria injection and 23 out of 37 zygotes developed were at the blastocyst-stage of embryonic development after 4.5 days of in vitro culture. All 23 of these blastocysts contained detectable levels of foreign mitochondria. These results represent an initial step in developing a model system to study mitochondrial dynamics and development of therapeutic strategies for human metabolic diseases affected by aberrations in mitochondrial function or mutation  相似文献

Cabbage root fly ( Delia brassicae ) egg-laying around radish plants ranging in age from newly emerged seedlings to plants with flowering shoots was studied in the field and laboratory. The attractiveness of radish changed with age, one peak in egg-laying occurring just before plants were of marketable size and further peaks as they produced flowering shoots. The pattern of egg-laying was similar for the six radish cultivars tested, both in the field and laboratory, irrespective of whether flies did or did not have a choice of plants of different ages. At any one age, there was no correlation between egg-laying and plant size. Selection within radish cultivars for the extremes of the range of egg-laying preference appeared to alter the cycle of attractiveness of different radish stocks to cabbage root fly, the response being influenced by environmental conditions.  相似文献

Abstract Sites of acid-phosphatase activity were found in the differentiating root protophloem of Nymphoides peltata by lead-salt and by azo-dye methods. Different substrates revealed different subcellular locations of the enzyme. The substrates β-glycerophosphate (β-GP) and naphthol ASBI phosphate revealed enzyme activity at similar sites within the sieve element. These sites included plasmodesmata, dictyosomes and small vacuoles in the cytoplasm. The substrate p-nitrophenylphosphate (p-NPP), however, revealed additional sites of acid-phosphatase activity which were not detectable by either naphthol ASBI phosphate or β-GP. For example, the inner region of the wall in mature sieve elements showed conspicuous acid-phosphatase activity only when p-NPP was used as substrate. The significance of the different locations of acid phosphatase within the sieve element is discussed. The convoluted ER, characteristic of immature sieve elements of N. peltata, failed to show acid-phosphatase activity whichever substate was used. By contrast, the stacked ER found in the parietal layer of mature sieve elements showed prominent acid-phosphatase activity regardless of the substrate used. The demonstration of acid-phosphatase activity in the stacked ER, and by both lead-salt and azo-dye methods, suggests that this organelle is a true site of acid-phosphatase activity. The onset of acid-phosphatase activity in the ER in later stages of sieve-element differentiation is compatible with the view that stacked ER plays a role in the final autolysis of the sieve-element protoplast.  相似文献

SYNOPSIS. The carbohydrate of variant-specific surface antigen glycoproteins from bloodstream forms of 13 cloned variants of Trypanosoma brucei was analyzed by gas-liquid chromatography. The glycoproteins contained from 6 to 17% carbohydrate by weight, and all contained the same 4 sugars: mannose, galactose, glucose, and glucosamine (probably as N-acetyl-glucosamine). The glycoprotein from variant 048, strain 427 contained (±20%) 11 mannose, 4 galactose, 4 glucose, and 5 glucosamine residues/mole of glycoprotein (molecular weight 65,000). Glucose was an integral component of the glycoproteins, not dissociable by sodium dodecyl sulphate, 8 M urea, or 1 M acetic acid. Some of the glucose was dissociated by trichloroacetic acid. Most of the glycoproteins formed precipitin bands with concanavalin A in Ouchterlony double diffusion, but none formed such bands with wheat germ agglutinin or Ricinus communis lectin (molecular weight 120,000).  相似文献

A new species of dragonet, Callionymus russelli, is described from Queensland waters. Callionymus russelli may be distinguished from the closely related species, C. punctatus, by the colour patterns of the first dorsal fins, by the smaller size, by the snout length being the same or slightly less than the eye length, by the preopercular spine being slightly longer than the eye, and by the caudal fin length in the female being only slightly longer than the head, and rarely twice the length of the head in the male. From the other Queensland species of Callionymus, its small size will eliminate it from being confused with the majority of species. It differs from C. calliste and C. macdonaldi, the only two of similar size, by having a somewhat pointed snout, different dorsal fin colour patterns, and the distinct body markings of numerous light or dark brown vertical streaks along the sides of the body in males. A revised key to the Australian dragonet species within the genus Callionymus is given.  相似文献
