Spatial patterns of entomopathogenic and other free‐living nematode populations were analyzed at the landscape scale. Free‐living bacterial feeder populations can be classified into trophic groups based on their functional and life history characteristics. Differences in life history traits were hypothesized to result in different spatial structures of populations. Spatial autocorrelations (Moran's I index) and exponential variogram models were calculated and estimated for each trophic group, including entomopathogenic nematodes, and comparisons were made among them. Spatial autocorrelation results showed that the spatial structure of entomopathogenic nematode populations is similar to that of relatively r‐selected free‐living bacterial feeders, which share several life history traits with entomopathogenic nematodes.  相似文献

Silkworm pupae have much potential and many applications as a natural medicine to promote human health. However, their chemical components have not been fully characterized or understood. HPLC analysis was conducted to determine the content ratio (%) of individual amino acids in total protein of the pupae. It showed that glutamic acid (18.3%), histidine (14.6%) and alanine (10.2%) are the most common amino acids in silkworm pupae. Fatty acid composition of silkworm pupae oil was revealed by high‐pressure liquid chromatography and gas chromatography – mass spectroscopy analyses. They contain a high ratio of essential fatty acids, [α‐linolenic acid (ω‐3 fatty acid]+ linoleic acid) (49.0%), and also contain non‐essential fatty acids, oleic acid (19.9%), palmitoleic acid (2.5%), palmitic acid (19.7%), stearic acid (8.6%), and eicosapentaenoic acid (EPA) (0.3%). In addition, they also contain antioxidants, quercetin diglucoside and nutritionally important riboflavin (vitamin B2). This study suggests that silkworm pupae are a nutritionally valuable food product and are applicable as cosmetic components with essential amino acids, essential fatty acids, antioxidants and vitamins. The animal experiment showed that alcohol dehydrogenase (ADH) activity was significantly higher in the liver of mice orally administered with 0.5 mg/mL of silkworm extract and alcohol than with commercial Dawn808? and alcohol, indicating that silkworm pupae extracts have alcohol detoxification activity.  相似文献

DNA was extracted from surface-sterilized salt of different geological ages (23, 121, 419 million years of age, MYA) to investigate haloarchaeal diversity. Only Haloarcula and Halorubrum DNA was found in 23 MYA salt. Older crystals contained unclassified groups and Halobacterium . The older crystals yielded a unique 55-bp insert within the 16S rRNA V2 region. The secondary structure of the V2 region completely differed from that in haloarchaea of modern environments. The DNA demonstrates that unknown haloarchaea and the Halobacterium were key components in ancient hypersaline environments. Halorubrum and Haloarcula appear to be a dominant group in relatively modern hypersaline habitats.  相似文献

1. A series of experiments examined growth of Daphnia magna on three algal diets ( Rhodomonas minuta , Scenedesmus acutus and Synechococcus sp.) at varying physiological states [nitrogen and phosphorus (P) limitations] to test whether variation in algal fatty acid and/or elemental composition can predict Daphnia growth.
2. These algae differed widely in their essential fatty acid (EFA) composition while phosphorus (P) or nitrogen limitation had only a small influence on their ω3-polyunsaturated fatty acid (PUFA) content.
3. Individually, algal ω3-PUFA content explained 69% of the variation in the normalised growth of D. magna, while algal phosphorus content explained 11% of the variation. Quantitative models for D. magna growth used both algal ω3-PUFA content and algal C : P ratio as food quality indices. Together, algal ω3-PUFA content and C : P ratio explained 70% of the variation in the normalised growth rate of D. magna .
4. Our results indicate that EFA influenced algal food quality much more strongly than P. The EFA and mineral P impacts appear to be independent.  相似文献

Polyclonal antibodies made against 86 kDa (86 k), 80 kDa (80 k) and 54 kDa (54 k) vitellins of Oxya japonica japonica are used for Western blotting. Anti‐80k vitellin antibody is cross‐reacted with a 95 kDa (95 k) vitellin. While 95 k vitellin is present both in the female hemolymph and in the oocyte, 80 k vitellin is detected only in the oocyte and the laid egg. In the growing oocytes, as 95 k vitellin is faded out gradually, 80 k vitellin is accumulated increasingly, indicating postendocytic processing of 95 k vitellin brings 80 k vitellin. Further conforming the hypothesis, partial digestion of 95 k vitellin with pepsin and α‐chymotrypsin makes several protein bands of molecular weight around 80 kDa. Thus, the 95k vitellin may have a cleavage site (s) to produce 80 k vitellin which forms fairly stable tertiary structure. In the reduced condition (20 mM glutathion), both 95 k and 80 k vitellins were digested throughly by endogenous proteinase at pH 4. Both 86 k and 54 k vitellins, respectively, show no apparent molecular weight changes in the growing oocyte and in the hemolymph.  相似文献

The species is found on host plants, Zelkova serrata Makino and Ulmus davidiana var. japonica Nakai. and is a well‐known forest pest. ABSTRACT From Bombyx mori larvae, RT-PCR and cDNA library screening isolated masquerade-like serine proteinase homologue cDNA gene, proposed to be related to insect immunity and its characteristics were examined. The isolated gene is composed of 1.3 kb of nucleotide and 420 amino acid residues were encoded. According to the results of database search, the isolated gene showed high sequence homology with Holotrichia and Tenebrio's 45 kDa protein, Drosophila CG5390 gene. Moreover, it is composed of regulatory domain and catalytic domain, which is characteristic of serine proteinase that can be found in the insect immune reaction and embryonic development processes. Enzyme activation site by proteolytic cleavage and the sequence of three amino acids participate in the catalytic triad of enzyme and 14 cystein residues used in disulfide bridges are well conserved with the compared genes. The mRNA expression was increased following E. coli injection and constitutive expression was also observed before injection by Northern blot analysis.  相似文献

A morphologically explicit numerical model for analysing wateruptake by individual roots was developed based on a conductornetwork, with specific conductors representing axial or radialconductivities for discrete root segments. Hydraulic conductivity(Lp; m s–1 MPa–1) was measured for roots of Agavedeserti Engelm. and Opuntia ficus-indica (L.) Miller by applyinga partial vacuum to the proximal ends of excised roots in solution.Lp was also measured for 40- to 80-mm segments along a root,followed by measurements of axial conductivity and calculationof radial conductivity. Predicted values of Lp for entire rootsbased on two to ten segments per root averaged 1.04±0.07(mean±s.e. mean for n = 3) of the measured Lp for A.deserti and 1.06±0.10 for O. ficus-indica. The modelalso closely predicted the drop in water potential along theroot xylem (xylem); when a tension of 50 kPa was applied tothe proximal ends of 0.2 m-long roots of A. deserti and O. ficus-indica,the measured xylem to midroot averaged 30 kPa compared witha predicted decrease of 36 kPa. Such steep gradients in xylemsuggest that the driving force for water movement from the soilto young distal roots may be relatively small. The model, whichagreed with an analytical solution for a simple hypotheticalsituation, can quantify situations without analytical solutions,such as when root and soil properties vary arbitrarily alonga root. Attached 2-month-old roots of the succulent plant, Opuntia ficus-indica,shrank 0.4% radially during periods of maximal transpirationunder wet conditions. In contrast, reversible decreases in diameterof nearly 20% occurred for these roots as their ambient waterpotential () in the vapour phase decreased from –0.01to –10 MPa over 8 d, the changes being slightly more rapidat 40 °C than at 10 °C. Such substantial diameter changesbecame progressively less with root age, from a 43% decreasein diameter at 3 weeks to a 6% decrease at 12 months Root shrinkagewas slight when was decreased from –0.01 to –0.3MPa, the latter being similar to the root water potential.As was further decreased from –0.3 to –10 MPa,water movement out of cortical cells caused considerable rootshrinkage. The root hydraulic conductivity (Lp) decreased only30 to 60% for a change in from –0.01 to –10 MPacompared with a decrease of over 106-fold for the soil hydraulicconductivity over this range. The overall conductivity of thesoil, the root-soil air gap, and the root was predicted to bedominated by Lp for soil above –0.3 MPa. As simulatedsoil decreased below –0.3 MPa, the root-soil air gap initiallybecame the primary limiter of water loss from the roots. Below–5 MPa for 1-month-old roots and below –2 MPa for12-month-old roots, the soil became the main limiter of waterloss.
