Escherichia coli heat-labile enterotoxin (LT) consists of an A subunit and five B subunits. These subunits oligomerize into an assembled holotoxin within the periplasm. Structural analysis of LT has revealed that the A subunit interacts with the B subunit through its carboxy terminus. This indicates that the carboxy-terminal portion of the protein is required for assembly of holotoxin in the periplasm. However, it is not known whether other regions of the A subunit contribute to the assembly. The A subunit constituting the holotoxin contains a disulfide bond between Cys-187 and Cys-199. It has been observed in many proteins that the intramolecular disulfide bond is deeply involved in the function and tertiary structure of the protein. We speculated that the disulfide bond of the A subunit contributes to the assembly in the periplasm, although the bond is not a structural element of the carboxy-terminal portion of the A subunit. We replaced these cysteine residues of the A subunit by oligonucleotide-directed site-specific mutagenesis and analyzed the LTs produced by cells containing the mutant LT genes. The amount of the mutant holotoxin produced was small compared with that of the wild-type strain, indicating that the disulfide bond of the A subunit contributes to the structure which functions as the site of nucleation in the assembly. A reconstitution experiment in vitro supported the notion. Subsequently, we found that the mutant A subunit constituting holotoxin is easily degraded by trypsin and that in cells incubated with mutant LTs, the lag until the intracellular cyclic AMP begins to accumulate is longer than in cells incubated with native LTs. These results might be useful for the analysis of the interaction of LT with target cells at the molecular level.  相似文献

Certain rpsL (which encodes the ribosomal protein S12) mutations that confer resistance to streptomycin markedly activate the production of antibiotics in Streptomyces spp. These rpsL mutations are known to be located in the two conserved regions within the S12 protein. To understand the roles of these two regions in the activation of silent genes, we used site-directed mutagenesis to generate eight novel mutations in addition to an already known (K88E) mutation that is capable of activating antibiotic production in Streptomyces lividans. Of these mutants, two (L90K and R94G) activated antibiotic production much more than the K88E mutant. Neither the L90K nor the R94G mutation conferred an increase in the level of resistance to streptomycin and paromomycin. Our results demonstrate the efficacy of the site-directed mutagenesis technique for strain improvement.  相似文献

Heavy metals, depending on their oxidation states, can be highly reactive and, as a consequence, toxic to most organisms. They are produced by an expanding variety of anthropogenic sources suggesting an increasingly important role for this form of pollution. The toxic effect of heavy metals appears to be related to production of reactive oxygen species (ROS) and the resulting unbalanced cellular redox status. Algae respond to heavy metals by induction of several antioxidants, including diverse enzymes such as superoxide dismutase, catalase, glutathione peroxidase and ascorbate peroxidase, and the synthesis of low molecular weight compounds such as carotenoids and glutathione. At high, or acute, levels of metal pollutants, damage to algal cells occurs because ROS levels exceed the capacity of the cell to cope. At lower, or chronic, levels algae accumulate heavy metals and can pass them on to organisms of other trophic levels such as mollusks, crustaceans, and fishes. We review here the evidence linking metal accumulation, cellular toxicity, and the generation of ROS in aquatic environments.  相似文献

Animal models are currently used to verify the biodistribution of different radiopharmaceuticals before its clinical application in Nuclear Medicine; however, there may be some limitations. The utilization of labelled anti-tumor monoclonal antibodies (MoAb) in experimental models often requires implant of human antigens (usually a cellular implant), which cannot be achieved in immunocompetent animals. Our purpose was to label an anti-CEA MoAb with technetium-99m (99Tc) and to validate a simplified animal model using a noncellular antigenic implant. MoAb was directly labelled with 99mTc, after reduction with 2-mercaptoethanol. Labeling efficiency was checked by ascending chromatography and immunoreactive fraction was measured in plastic wells sensitized with the antigen. Radiopharmaceutical biodistribution was evaluated by dissection and scintigraphy in 5 mice groups; following the subcutaneous administration of Al(OH)3, CEA adsorbed Al(OH)2 and a control group evaluation. Labeling efficiency was 94+/-3%, which showed to be stable for 24 hr, with immunoreactive fraction above 50%. Invasive biodistribution evaluation showed prolonged blood retention, hepatic and renal uptake. A significant increase in uptake was observed in scintigraphic studies of animals with CEA-adsorbed Al(OH)3 implants compared with the other groups (p<0.05). The non-cellular antigenic implant model simplifies the pre-clinical evaluation of labelled MoAb.  相似文献

Male crickets, Gryllus bimaculatus Degeer, turned lights on or off in a chamber by a seesaw device: (1) during a 12 h, and (2) during a 24 h day. The crickets in (1) and the last-instar nymph in (2) turned the lights on and off at irregular intervals and duration. The selection rate for darkness was greater than that for light by an average exceeding 80%. The locomotor activity of the nymph in (1) was arrhythmic. In (1) the adults stridulated and were active in continuous darkness during the 12 h, while, in contrast in (2) they turned on the light and stridulated without switching the light off. The locomotor and stridulatory activities of the adult crickets in (2) were free-running. These activities resulted in a free-running rhythm of selection for light or darkness in (2). Under the conditions of the present experiments, the circadian pacemaker functioned in the same way in light and dark cycles as in constant light conditions.  相似文献

Auxin activates pumping of protons from the symplast to theapoplast and causes hyperpolarization of the symplast membranein the elongation zone of Vigna stems prior to the accelerationof growth. This auxin-induced hyperpolarization has been studiedin most cases in hypocotyl segments excised from the elongationzone. In the present study, mature-zone segments were perfusedwith IAA by the xylem perfusion technique in an effort to determinewhether or not IAA has any effects in the mature zone. Althoughno hyperpolarization of the symplast membrane was observed uponthe perfusion with auxin alone, auxin-induced hyperpolarizationwas observed when mature-zone segments had been pretreated withGA3, in the absence of an increase in the growth rate. Theseresults suggest that cells in the mature zone have lost theability to activate the proton-pumping machinery in responseto auxin but that this ability can be restored by treatmentwith GA3. This effect of GA3 suggests the possibility that theconcentration of gibberellin in a tissue controls one of thecell's responses to auxin, namely, activation of the protonpump. To attempt to develop physicochemical and physiological modelling for methane transport from the rhizosphere to the atmosphere through rice plants, methane flux, methane concentration in the soil water, and the biomass of rice were measured in lysimeter rice paddies (2.5 × 4 m, depth 2.0 m) once per week throughout the entire growing season in 1992 at Tsukuba, Japan. The addition of exogenous organic matter (rice straw) or soil amendments with the presence or absence of vegetation were also examined for their influence on methane emissions. The total methane emission over the growing season varied from 3.2 g CH4 m-2 y-1 without the addition of rice straw to 49.7 g CH4 m-2 y-1 with rice straw and microbiological amendment. In the unvegetated plot with the addition of rice straw, there was much ebullition of gas bubbles, particularly in the summer. The annual methane emission due to the ebullition of gas bubbles,from the unvegetated plot with the addition of rice straw was estimated to be almost the same as that from the vegetated site with the addition of rice straw. In the early growth stage, the methane flux can be analyzed by the diffusion model (Flux=Methane concentration × Conductance of rice body) using parameters for methane concentration in the soil water as a difference in concentration between the atmosphere and the rhizosphere, and for the biomass of rice as a conductance of rice body. On the other hand, although the diffusion model was inapplicable to a large extent from the middle to late growth stage, methane flux could be estimated by air temperature and concentration in the soil water. Thus, methane transport from the rhizosphere to the atmosphere through rice plants consisted of two phases: one was an explainable small part by diffusion in rice body; the other was a large part strongly, governed by air temperature.
