Metabolism and excretion of exogenous [3H]-LTC4 in primates 总被引:1,自引:0,他引:1
Four novel omega- and beta-oxidation (from the omega end) products of peptide leukotrienes, 20-hydroxy and 20-carboxy-LTE4, 18-carboxy-19, 20-dinor-LTE4 and 16-carboxy-17,18,19,20-tetranor-14,15-dihydro-LTE4 were prepared by total synthesis and used as standards for identification of biliary and urinary metabolites in the cynomolgus monkey. After intravenous administration 14, 15-[3H] leukotriene C4 (10 microCi kg-1) was partially metabolized in and rapidly cleared from the vascular circulation. This resulted, within 24 hours, in significant urinary excretion (14.8 +/- 2.1%, n = 4), consisting largely of material more polar than LTE4 (61% of urinary excretion) as shown by reverse phase HPLC. The polar fraction demonstrated two predominant metabolites which coeluted in several HPLC solvent systems with synthetic 16-carboxytetranordihydro-LTE4 (major component) and 18-carboxydinor-LTE4 (minor component). Characterization of the major polar metabolite as 16-carboxytetranordihydro-LTE4 was substantiated by conversion to its N-acetylated derivative. The absence of the 14, 15 double bond was confirmed by product analysis of oxidative ozonolysis. In a single animal, the bile duct was cannulated, with significant biliary excretion of radioactivity demonstrated over 4 hours (58.6% recovery). The predominant polar biliary metabolites were also identified as the 18-carboxydinor and 16-carboxytetranordihydro derivatives of LTE4 mentioned above. These data suggest that beta-oxidation products generated from the omega-carboxyl end of the 20-carboxy-LTE4 are important products of [3H] LTC4 metabolism in the monkey. Summary The interior of Bowman's capsules of rat kidneys has been examined by scanning electron microscopy, and a distinctive population of cells around the exposed vascular poles of glomerular tufts were identified. The cells were situated in the annular groove at the root of the glomerulus, between the parietal epithelial cells and the podocytes. These peripolar cells were dendritic cells with long processes embracing the glomerular arterioles. Up to three peripolar cells were present at each vascular pole and they were mainly distributed in the glomeruli of the outer third of the renal cortex. This first detailed study of the surface morphology of the glomerular peripolar cell supports the suggestion that changes in the diameter of the polar region of the glomerular tuft may cause variations in stretching of the cuff of peripolar cells, and hence modulation of their secretory activity. 相似文献

Diet-induced thermogenesis (DIT) in young rats overeating a "cafeteria" (CAF) diet of palatable human foods is characterized by a chronic, propranolol-inhibitable elevation in resting metabolic rate (VO2) and is associated with various changes in brown adipose tissue (BAT) that have been taken as evidence for BAT as the effector of DIT. But direct evidence for participation of BAT in DIT has been lacking. By employing a nonocclusive cannula to sample the venous effluent of interscapular BAT (IBAT) for analysis of its O2 content and measuring tissue blood flow with microspheres, we accomplished direct determination (Fick principle) of the O2 consumption of BAT in conscious CAF rats. In comparison with normophagic controls fed chow, the CAF rats exhibited a 43% increase in metabolizable energy intake, reduced food efficiency, a 22% elevation in resting VO2 at 28 degrees C (thermoneutrality) or 24 degrees C (housing temperature), and characteristic changes in the properties of their BAT (e.g., increased mass, protein content and mitochondrial GDP binding). They also exhibited the greater metabolic response to exogenous noradrenaline characteristic of CAF rats and the near elimination by propranolol of their elevation in VO2. By the criterion of their elevated VO2, the CAF rats were exhibiting DIT at the time of the measurements of BAT blood flow and blood O2 levels. However, BAT O2 consumption was found to be no greater in the CAF rats than in the controls at either 28 or 24 degrees C. The olfactory response from male pheromone sensitive sensilla was investigated in the endemic New Zealand brownheaded leafrollers Ctenopseustis obliquana (Walker) and C. sp. ropeana (Lepidoptera, Tortricidae). The responses from 281 sensilla from the parental strains and from both the reciprocal crosses, including F1, F2 and maternal and paternal backcrosses were recorded, and statistically analysed using a multivariate analysis.In males of both the parental strains, a large amplitude cell responded to the main pheromone component of the conspecific female, in C. obliquana (Z)-8-tetradecenyl acetate (Z8-14:OAc) and in C. sp ropeana (Z)-5-tetradecenyl acetate (Z5-14:OAc). Both male types also possessed a small amplitude cell, which in C. obliquana responded weakly to Z5-14:OAc and tetradecyl acetate (14:OAc), and in C. sp ropeana responded to Z8-14:OAc. The responses from the different types of hybrid males were more variable than the responses from parental males. A main pattern could, however be seen, corresponding with the expected pattern in a sex-linked inheritance on the Z-chromosome of a C. sp ropeana type dominant genetic factor. The more pronounced variation in the hybrids could not be explained by this model, and might be due to the involvement of additional genes.
Résumé Les réactions olfactives des sensilles mâles sensibles aux phéromones ont été examinées par enregistrement de l'extrémité de la sensille chez les tordeuses C. obliquana Walker et C. sp. ropeana. Les enregistrements ont porté sur 281 sensilles des lignées parentales et des croisements réciproques de F1, F2 et de croisements en retour maternel et paternel. Les résultats des enregistrements d'une sensille ont été soumis à une analyse en composantes principales.Chez les mâles de chaque lignée parentale un seul type physiologique de sensille a été découvert; une cellule répond par un pic grand au principal constituant de la phéromone femelle conspécifique. (Z)-8-acétate tétradécényl (Z8-14:OAc) pour C. obliquana, et (Z)-5-acétate tétradécényl (Z5-14:OAc) pour C. sp. ropeana. Une seconde type de cellule dans les sensilles des deux espèces de mâles présente un pic petit pour Z5-14:OAc et pour l'acétate tétradécyl (14:OAc) chez C. obliquana, et pour C. sp. ropeana au Z8-14:OAc. Les réponses des sensilles des différents types de mâles hybrides sont plus hétérogènes que celles des sensilles de leurs pères. Un schéma général pourrait cependant être décelé, correspondant au schéma prévu avec une hérédité d'un facteur dominant liée au sexe sur le chromosome Z de C. sp. ropeana. Although cardiac dysfunction due to ischemia-reperfusion injury is considered to involve oxygen free radicals, the exact manner by which this oxidative stress affects the myocardium is not clear. As the occurrence of intracellular Ca2+ overload has been shown to play a critical role in the genesis of cellular damage due to ischemia-reperfusion, this study was undertaken to examine whether oxygen free radicals are involved in altering the sarcolemmal Ca2+-transport activities due to reperfusion injury. When isolated rat hearts were made globally ischemic for 30 min and then reperfused for 5 min, the Ca2+ -pump and Na+-Ca2+ exchange activities were depressed in the purified sarcolemmal fraction; these alterations were prevented when a free radical scavenger enzymes (superoxide dismutase plus catalase) were added to the reperfusion medium. Both the Ca2+- pump and Na+- Ca2+ exchange activities in control heart sarcolemmal preparations were depressed by activated oxygen-generating systems containing xanthine plus xanthine oxidase and H2O2; these changes were prevented by the inclusion of superoxide dismutase and catalase in the incubation medium. These results support the view that oxidative stress during ischemia-reperfusion may contribute towards the occurrence of intracellular Ca2+ overload and subsequent cell damage by depressing the sarcolemmal mechanisms governing the efflux of Ca2+ from the cardiac cell. 相似文献

Summary Immunocytochemistry was used to determine if photoperiod and/or sex have any effect on the pattern of the luteinizing hormone-releasing hormone (LHRH) system in the brain of the Japanese quail. Immunopositive perikarya were found within three major areas of the brain: the rostral paraolfactory lobe, the preoptic, and the septal region. A quantitative analysis of LHRH cell numbers was performed on male and female quail after two photoperiodic treatments: sexually mature birds exposed to 24 weeks of 20 h light: 4 h darkness (20L4D), and birds with a regressed reproductive system (induced by transfer from a photoregime of 20L4D to 25 short days of 8L16D). Two-way analysis of variance showed that short-day males display significantly (p < 0.05) more immunopositive perikarya (607 + 134) than long-day males (291 + 114), short-day females (293 + 103) or long-day females (330 + 92). The density of LHRH-immunoreactive nerve fibres and the intensity of the immunostaining in the median eminence were always greater in long-day sexually mature quail (male and female) than in animals exposed to 25 days of 8L16D. These results demonstrate that the LHRH system of the quail is influenced by photoperiod and mirrors sexual differentiation. 相似文献

Summary The distribution of FMRFamide-like immunoreactive neurons in the nervous system of the slug Limax maximus was studied using immunohistochemical methods. Approximately one thousand FMRFamide-like immunoreactive cell bodies were found in the central nervous system. Ranging between 15 m and 200 m in diameter, they were found in all 11 ganglia of the central nervous system. FMRFamide-like immunoreactive cell bodies were also found at peripheral locations on buccal nerve roots. FMRFamide-like immunoreactive nerve fibres were present in peripheral nerve roots and were distributed extensively throughout the neuropil and cell body regions of the central ganglia. They were also present in the connective tissue of the perineurium, forming an extensive network of varicose fibres. The large number, extensive distribution and great range in size of FMRFamide-like immunoreactive cell bodies and the wide distribution of immunoreactive fibres suggest that FMRFamide-like peptides might serve several different functions in the nervous system of the slug. 相似文献
