The deleterious effects of H2O2 on the electron transport chain of yeast mitochondria and on mitochondrial lipid peroxidation were evaluated. Exposure to H2O2 resulted in inhibition of the oxygen consumption in the uncoupled and phosphorylating states to 69% and 65%, respectively. The effect of H2O2 on the respiratory rate was associated with an inhibition of succinate-ubiquinone and succinate-DCIP oxidoreductase activities. Inhibitory effect of H2O2 on respiratory complexes was almost completely recovered by β-mercaptoethanol treatment. H2O2 treatment resulted in full resistance to QO site inhibitor myxothiazol and thus it is suggested that the quinol oxidase site (QO) of complex III is the target for H2O2. H2O2 did not modify basal levels of lipid peroxidation in yeast mitochondria. However, H2O2 addition to rat brain and liver mitochondria induced an increase in lipid peroxidation. These results are discussed in terms of the known physiological differences between mammalian and yeast mitochondria.  相似文献

Tubulin polymerization promoting protein/p25 (TPPP/p25) modulates the dynamics and stability of the microtubule system and plays crucial role in the myelination of oligodendrocytes. Here we showed by CD, fluorescence, and NMR spectroscopies that Zn(2+) is the first ligand that induces considerable rearrangement of the disordered TPPP/p25. Zinc finger motif (His(2)Cys(2)) (His(61)-Cys(83)) was identified within the flexible region of TPPP/p25 straddled by extended unstructured N- and C-terminal regions. The specific binding of the Zn(2+) to TPPP/p25 induced the formation of molten globule but not that of a well-defined tertiary structure. The Zn(2+)-induced partially folded structure accommodating the zinc binding motif is localized at the single Trp(76)-containing region as demonstrated by fluorescence resonance energy transfer and quenching experiments. We showed that the Zn(2+)-induced change in the TPPP/p25 structure modified its interaction with tubulin and GTP coupled with functional consequences: the TPPP/p25-promoted tubulin polymerization was increased while the TPPP/p25-catalyzed GTPase activity was decreased as detected by turbidimetry and by malachite green phosphate release/(31)P NMR assays, respectively. The finding that the Zn(2+) of the bivalent cations can uniquely influence physiological relavant interactions significantly contributes to our understanding of the role of Zn(2+)-related TPPP/p25 processes in the differentiation/myelination of oligodendrocytes possessing a high-affinity Zn(2+) uptake mechanism.  相似文献

Replicon architecture in bacteria is commonly comprised of one indispensable chromosome and several dispensable plasmids. This view has been enriched by the discovery of additional chromosomes, identified mainly by localization of rRNA and/or tRNA genes, and also by experimental demonstration of their requirement for cell growth. The genome of Rhizobium etli CFN42 is constituted by one chromosome and six large plasmids, ranging in size from 184 to 642 kb. Five of the six plasmids are dispensable for cell viability, but plasmid p42e is unusually stable. One possibility to explain this stability would be that genes on p42e carry out essential functions, thus making it a candidate for a secondary chromosome. To ascertain this, we made an in-depth functional analysis of p42e, employing bioinformatic tools, insertional mutagenesis, and programmed deletions. Nearly 11% of the genes in p42e participate in primary metabolism, involving biosynthetic functions (cobalamin, cardiolipin, cytochrome o, NAD, and thiamine), degradation (asparagine and melibiose), and septum formation (minCDE). Synteny analysis and incompatibility studies revealed highly stable replicons equivalent to p42e in content and gene order in other Rhizobium species. A systematic deletion analysis of p42e allowed the identification of two genes (RHE_PE00001 and RHE_PE00024), encoding, respectively, a hypothetical protein with a probable winged helix-turn-helix motif and a probable two-component sensor histidine kinase/response regulator hybrid protein, which are essential for growth in rich medium. These data support the proposal that p42e and its homologous replicons (pA, pRL11, pRLG202, and pR132502) merit the status of secondary chromosomes.  相似文献

Leishmania mexicana is able to interact with the fibrinolytic system through its component plasminogen, the zymogenic form of the protease plasmin. In this study a new plasminogen binding protein of this parasite was identified: LACK, the Leishmania homolog of receptors for activated C-kinase. Plasminogen binds recombinant LACK with a Kd value of 1.6 ± 0.4 μM, and binding is lysine-dependent since it is inhibited by the lysine analog ε-aminocaproic acid. Inhibition studies with specific peptides and plasminogen binding activity of a mutated recombinant LACK have highlighted the internal motif 260VYDLESKAV268, similar to those found in several enolases, as involved in plasminogen binding. Recombinant LACK and secreted proteins, in medium conditioned by parasites, enhance plasminogen activation to plasmin by the tissue plasminogen activator (t-PA). In addition to its localization in the cytosol, in the microsomal fraction and as secreted protein in conditioned medium, LACK was also localized on the external surface of the membrane. The results presented here suggest that LACK might bind and enhance plasminogen activation in vivo promoting the formation of plasmin. Plasminogen binding of LACK represents a new function for this protein and might contribute to the invasiveness of the parasite.  相似文献

Quantifying population genetic structure is fundamental to testing hypotheses regarding gene flow, population divergence and dynamics across large spatial scales. In species with highly mobile life‐history stages, where it is unclear whether such movements translate into effective dispersal among discrete philopatric breeding populations, this approach can be particularly effective. We used seven nuclear microsatellite loci and mitochondrial DNA (ND2) markers to quantify population genetic structure and variation across 20 populations (447 individuals) of one such species, the European Shag, spanning a large geographical range. Despite high breeding philopatry, rare cross‐sea movements and recognized subspecies, population genetic structure was weak across both microsatellites and mitochondrial markers. Furthermore, although isolation‐by‐distance was detected, microsatellite variation provided no evidence that open sea formed a complete barrier to effective dispersal. These data suggest that occasional long‐distance, cross‐sea movements translate into gene flow across a large spatial scale. Historical factors may also have shaped contemporary genetic structure: cluster analyses of microsatellite data identified three groups, comprising colonies at southern, mid‐ and northern latitudes, and similar structure was observed at mitochondrial loci. Only one private mitochondrial haplotype was found among subspecies, suggesting that this current taxonomic subdivision may not be mirrored by genetic isolation.  相似文献

Canopy trees are largely responsible for the environmental heterogeneity in the understory of tropical and subtropical species‐rich forests, which in turn may influence sapling community dynamics. We tested the effect of the specific identity of four cloud forest canopy trees on total solar radiation, canopy openness, soil moisture, litter depth, and soil temperature, as well as on the structure and dynamics of the sapling community growing beneath their canopies. We observed significant effects of the specific identity of canopy trees on most understory microenvironmental variables. Soil moisture was higher and canopy openness lower beneath Cornus disciflora. In turn, canopy openness and total solar radiation were higher beneath Oreopanax xalapensis, while the lowest soil moisture occurred beneath Quercus laurina. Moreover, Chiranthodendron pentadactylon was the only species having a positive effect on litter depth under its canopy. In spite of these between‐species environmental differences, only C. pentadactylon had significant, negative effects on sapling density and species richness, which may be associated to low seed germination and seedling establishment due to an increased litter depth in its vicinity. The relevance of the specific identity of canopy trees for natural regeneration processes and species richness maintenance depends on its potential to differentially affect sapling dynamics through species‐specific modifications of microenvironmental conditions.  相似文献

Abstract: This paper reports the first record of Desmanella (Mammalia, Soricomorpha) from the Granada Basin in southern Iberian Peninsula, which represents its south‐westernmost occurrence in all Eurasia. It is a controversial taxon whose systematic assignment has been discussed for a long time. This genus belongs to the family Talpidae, a group of insectivores that include extant moles, shrew moles and desmans. Desmanella was very abundant in the late Miocene and early Pliocene of Eurasia, including several basins of northern Iberian Peninsula, but it has not been found until now in southern Iberia. Previous studies have shown that the faunal record and the climatic conditions during the Neogene in the Granada Basin were different from other areas of the Iberian Peninsula. The particular distribution of Desmanella supports the hypothesis that specific climatic features (wetter than neighbouring areas) prevailed in the Granada Basin during the late Turolian (late Miocene).  相似文献

The ubiquitin-proteasome system is responsible for the degradation of numerous proteins in eukaryotes. Degradation is an essential process in many cellular pathways and involves the proteasome degrading a wide variety of unrelated substrates while retaining specificity in terms of its targets for destruction and avoiding unneeded proteolysis. How the proteasome achieves this task is the subject of intensive research. Many proteins are targeted for degradation by being covalently attached to a poly-ubiquitin chain. Several studies have indicated the importance of a disordered region for efficient degradation. Here, we analyze a data set of 482 in vivo ubiquitinated substrates and a subset in which ubiquitination is known to mediate degradation. We show that, in contrast to phosphorylation sites and other regulatory regions, ubiquitination sites do not tend to be located in disordered regions and that a large number of substrates are modified at structured regions. In degradation-mediated ubiquitination, there is a significant bias of ubiquitination sites to be in disordered regions; however, a significant number is still found in ordered regions. Moreover, in many cases, disordered regions are absent from ubiquitinated substrates or are located far away from the modified region. These surprising findings raise the question of how these proteins are successfully unfolded and ultimately degraded by the proteasome. They indicate that the folded domain must be perturbed by some additional factor, such as the p97 complex, or that ubiquitination may induce unfolding.  相似文献

Santos, A., Mayoral, E.J., da Silva, C.M., Cachão, M., Johnson, M.E. & Baarli, B.G. Miocene intertidal zonation on a volcanically active shoreline: Porto Santo in the Madeira Archipelago, Portugal. Lethaia, Vol. 44, pp. 26–32. Short‐term biological colonization of rockgrounds on the basaltic shorelines of oceanic islands has, as yet, been poorly explored. A Miocene sea cliff on Ilhéu de Cima off Porto Santo in the Madeira Archipelago of Portugal provides a case study showing intertidal zonation with two types of barnacles, serpulid worm tubes, two coral species, epifaunal bivalves and the trace fossils of endolithic bivalves. Large barnacles (Balanus sp.) and serpulids are limited to the upper 400 mm of a basalt cliff of 1.6 m in height. Small barnacles, possibly of the same species, extend to the base. The upper half includes the corals Isophyllastrea orbignyana and Tarbellastraea reussiana, to which many small, coral‐inhabiting, barnacles (Ceratoconcha costata) are fixed. Borings identified as Gastrochaenolites torpedo appear through the bottom two‐thirds of the cliff face. Rarely, Gastrochaenolites lapidicus is exposed in longitudinal section with borings up to 45 mm deep in solid basalt. Epifaunal bivalves, such as Spondylus sp., are limited to a middle zone. Associated with the sea cliff is an outer platform on which a multitude of T. reussiana colonies occur in growth position. The corals exhibit planar erosion over180 m2. The shelf was faulted and cut by a basalt dike prior to the brief recolonization of I. orbignyana, found attached to low fault scarps. Habitation of the sea cliff was facilitated by rising sea level, but abruptly terminated by burial under volcanoclastic ejecta. □Basalt bioerosion, coastal dynamics, Ilhéu de Cima (Porto Santo), Miocene intertidal zonation, volcanogenic perturbations.  相似文献

Sexual cannibalism usually involves females attacking and consuming males before, during or after copulation. Sex role reversed systems may provide insight into the debate about whether it arises as mistaken identity, a spillover in female aggressiveness, foraging decisions, and/or extreme mate choice. In such systems, males may be selective and voracious to compensate for their higher reproductive costs, and thus males may be the sexually cannibalistic sex. Allocosa brasiliensis shows a reversal in sex roles and male‐biased sexual size dimorphism (the opposite of the common pattern in spiders). The present study aimed to test whether males cannibalize or mate according to female reproductive status or body characteristics. Each of 20 adult males was consecutively exposed to one virgin and one mated female, alternating the order of exposure. Males preferred to mate with virgin females in good body condition and heavier‐mated females. Males attacked 15% of virgins and 40% of mated females and cannibalized 10% and 25% of the total trials, respectively. The astonishing male cannibalistic behaviour best agrees with extreme mate choice hypotheses because attacks were more frequent on mated females of low body condition.
