The egg and larval stages of the generalist endoparasitoid Campoletis sonorensis Carlson (Hymenoptera: Ichneumonidae), which normally avoid the hemocytic reaction of many Lepidopteran host species, are encapsulated in 40% of Spodoptera frugiperda J. E. Smith (Lepidoptera: Noctuidae) larvae. The effect of parasitism on inhibiting the spreading ability of S. frugiperda plasmatocytes in vitro is more pronounced in susceptible larvae which fail to encapsulate the parasitoid than in resistant ones permitting parasitoid development. This suggests that induction of plasmatocyte pathology is relevant to the successful evasion of encapsulation by the parasitoid. Some granular cells disappear from the hemolymph of the parasitized resistant larvae, which implicates their involvement in the encapsulation reaction. Calyx fluid of C. sonorensis injected into host larvae produced effects on host hemocytes identical to natural parasitism. Several mechanism may cooperate to protect the parasitoid from encapsulation. The pathological reactions by the host plasmatocytes is one main manifestation of the immunosuppressive parasitoid effect. Results are discussed in regard to the known effects of C. sonorensis on Heliothis virescens Fabricius (Lepidoptera: Noctuidae) larval hemocytes which are totally unable to respond with a successful cellular defense reaction.
Résumé L'ichneumonide Campoletis sonorensis Carlson, endoparasitoïde larvaire, se développe dans de nombreuses espèces de Lépidoptères. Son statut de généraliste est dû, notamment, à son aptitude à déjouer les défenses immunitaires de ses hôtes, c.a.d. la formation d'une capsule d'hémocytes autour de l'oeuf ou de la larve parasite. Cependant, chez le noctuide Spodoptera frugiperda J.E. Smith, C. sonorensis est encapsulé dans 40% des larves qu'il parasite. Nous avons étudié la population hémocytaire de deux catégories de larves de S. frugiperda, celles qualifiées de résistantes à C. sonorensis, et qui arrêtent son développement, et celles susceptibles où le parasitoïde échappe à l'encapsulation.Cinq types d'hémocytes ont été identifiés: les prohémocytes (PR), les sphérulocytes (SP), les granulocytes (GR), les plasmatocytes (PL) et les oenocytoïdes (OE). Chez les hôtes susceptibles et résistants, le parasite provoque une baisse identique de la concentration totale des hémocytes dans l'hémolymphe (THC). Par contre, les PLs sont davantage affectés chez les hôtes susceptibles que chez ceux résistants au parasitoïde. Les résultats montrent que, chez les hôtes susceptibles, 1) le nombre des PLs dans l'hémolymphe est davantage diminué, et 2) leur aptitude d'adhérence in vitro est davantage inhibée. Il existe donc une corrélation positive entre le degré de pathologies qui affectent les PLs de l'hôte et l'incapacité de celui-ci à encapsuler le parasitoïde. Ceci tend à démontrer le rôle-clé des PLs dans la réaction immunitaire d'encapsulation chez S. frugiperda, comme chez de nombreux insectes. De plus, ce résultat renforce l'hypothèse selon laquelle C. sonorensis éviterait l'encapsulation en agissant sur les hémocytes de l'hôte, et plus particulièrement sur les PLs. Inversement aux PLs, les GRs sont moins abondants dans l'hémolymphe des hôtes qui encapsulent C. sonorensis. Les GRs pourraient donc participer à la formation de la capsule hémocytaire.Il est possible que plusieurs facteurs contribuent à protéger C. sonorensis de l'encapsulation. Néanmoins, les pathologies affectant les hémocytes des hôtes parasités sont probablement une manifestation majeure de l'effet immunosuppresseur du parasitoïde.Les effets de C. sonorensis sur les hémocytes des larves parasitées peuvent être reproduits chez des larves saines, en leur injectant de venin extrait des glandes du calyx des femelles parasitoïdes. Ces sécrétions provenant de la glande du calyx, et normalement injectées dans l'hôte lors de l'oviposition, sont probablment responsable, au moins en partie, de l'effet immunosuppresseur du parasitoïde.Ces résultats peuvent être comparés à ceux obtenus chez l'hôte Heliothis virescens Fabricius (lépidoptère, noctuide) qui n'encapsule jamais C. sonorensis. Bien que le parasitoïde provoque les mêmes effets pathologiques sur les hémocytes des hôtes de S. frugiperda et d'H. virescens, on constate que l'effet apparait quelques heures après l'oviposition chez H. virescens, contre 48 heures post-oviposition chez S. frugiperda. Ce délai pourrait contribuer à la résistance immunitaire de certaines larves de S. frugiperda, résistantes, à C. sonorensis.

The rhodium(II)-catalyzed intermolecular C-H insertion of methyl aryldiazoacetates with either N-Boc-piperidine or N-Boc-pyrrolidine followed by deprotection with trifluoroacetic acid is a very direct method for the synthesis of methylphenidate analogues. By using either dirhodium tetraacetate or dirhodium tetraprolinate derivatives as catalyst, either the racemic or enantioenriched methylphenidate analogues can be prepared. The binding affinities of the methylphenidate analogues to both the dopamine and the serotonin transporters are described. The most notable compounds are the erythro-(2-naphthyl) analogues which display high binding affinity and selectivity for the serotonin transporter.  相似文献

Plant essential oils (and/or active components) can be used as alternatives or adjuncts to current antiparasitic therapies. Garlic oil has broad-spectrum activity against Trypanosoma, Plasmodium, Giardia and Leishmania, and Cochlospermum planchonii and Croton cajucara oils specifically inhibit Plasmodium falciparum and Leishmania amazonensis, respectively. Some plant oils have immunomodulatory effects that could modify host-parasite immunobiology, and the lipid solubility of plant oils might offer alternative, transcutaneous delivery routes. The emergence of parasites resistant to current chemotherapies highlights the importance of plant essential oils as novel antiparasitic agents.  相似文献

Cryptosporidium parvum excystation and host cell invasion have been characterized in some detail ultrastructurally. However, until recently, the biochemical and molecular basis of host-parasite interactions and parasite- and host-specific molecules involved in excystation, motility and host cell invasion have been poorly understood. This article describes our understanding of Cryptosporidium excystation and the events leading to host cell invasion, and draws from information available about these processes in other apicomplexans. Many questions remain but, once the specific mechanisms are identified, they could prove to be novel targets for drug delivery.  相似文献

Therapeutic inhibition of poly(ADP-ribose) polymerase (PARP), as monotherapy or to supplement the potencies of other agents, is a promising strategy in cancer treatment. We previously reported that the first PARP inhibitor to enter clinical trial, rucaparib (AG014699), induced vasodilation in vivo in xenografts, potentiating response to temozolomide. We now report that rucaparib inhibits the activity of the muscle contraction mediator myosin light chain kinase (MLCK) 10-fold more potently than its commercially available inhibitor ML-9. Moreover, rucaparib produces additive relaxation above the maximal degree achievable with ML-9, suggesting that MLCK inhibition is not solely responsible for dilation. Inhibition of nitric oxide synthesis using L-NMMA also failed to impact rucaparib’s activity. Rucaparib contains the nicotinamide pharmacophore, suggesting it may inhibit other NAD+-dependent processes. NAD+ exerts P2 purinergic receptor-dependent inhibition of smooth muscle contraction. Indiscriminate blockade of the P2 purinergic receptors with suramin abrogated rucaparib-induced vasodilation in rat arterial tissue without affecting ML-9-evoked dilation, although the specific receptor subtypes responsible have not been unequivocally identified. Furthermore, dorsal window chamber and real time tumor vessel perfusion analyses in PARP-1-/- mice indicate a potential role for PARP in dilation of tumor-recruited vessels. Finally, rucaparib provoked relaxation in 70% of patient-derived tumor-associated vessels. These data provide tantalising evidence of the complexity of the mechanism underlying rucaparib-mediated vasodilation.  相似文献

Philobryids (Bivalvia: Arcoida) are one of the most speciose marine bivalve families in the Southern Ocean and are common throughout the Southern Hemisphere. Considering this diversity and their brooding reproductive mode (limiting long-distance dispersal), this family may have been present in the Southern Ocean since its inception. However Philobrya and Adacnarca appear only in the Quaternary fossil record of the Antarctic, suggesting a much more recent incursion. Molecular dating provides an independent means of measuring the time of origin and radiation of this poorly known group. Here we present the first combined molecular and morphological investigation of the Philobryidae in the Southern Ocean. Two nuclear loci (18S and 28S) were amplified from 35 Southern Ocean Adacnarca and Philobrya specimens, with a combined sequence length of 2,282 base pairs (bp). Adacnarca specimens (A. nitens and A. limopsoides) were resolved as a strongly supported monophyletic group. Genus Philobrya fell into two strongly supported groups (‘sublaevis’ and ‘magellanica/wandelensis’), paraphyletic with Adacnarca. The A. nitens species complex is identified as at least seven morpho-species through morphological and genetic analysis of taxon clustering. Phylogenetic analyses resolve Philobryidae as a strongly supported monophyletic clade and sister taxon to the Limopsidae, as anticipated by their classification into the superfamily Limopsoidea. Bayesian relaxed clock analyses of divergence times suggest that genus Adacnarca radiated in the Southern Ocean from the Early Paleogene, while P. sublaevis and P. wandelensis clades radiated in the late Miocene, following the formation of the Antarctic Circumpolar Current.  相似文献

In the developing endosperm of bread wheat (Triticum aestivum), seed storage proteins are produced on the rough endoplasmic reticulum (ER) and transported to protein bodies, specialized vacuoles for the storage of protein. The functionally important gluten proteins of wheat are transported by two distinct routes to the protein bodies where they are stored: vesicles that bud directly off the ER and transport through the Golgi. However, little is known about the processing of glutenin and gliadin proteins during these steps or the possible impact on their properties. In plants, the RabD GTPases mediate ER‐to‐Golgi vesicle transport. Available sequence information for Rab GTPases in Arabidopsis, rice, Brachypodium and bread wheat was compiled and compared to identify wheat RabD orthologs. Partial genetic sequences were assembled using the first draft of the Chinese Spring wheat genome. A suitable candidate gene from the RabD clade (TaRabD2a) was chosen for down‐regulation by RNA interference (RNAi), and an RNAi construct was used to transform wheat plants. All four available RabD genes were shown by qRT‐PCR to be down‐regulated in the transgenic developing endosperm. The transgenic grain was found to produce flour with significantly altered processing properties when measured by farinograph and extensograph. SE‐HPLC found that a smaller proportion of HMW‐GS and large proportion of LMW‐GS are incorporated into the glutenin macropolymer in the transgenic dough. Lower protein content but a similar protein profile on SDS‐PAGE was seen in the transgenic grain.  相似文献

The movement ability of species in fragmented landscapes must be considered if habitat restoration strategies are to allow maximum benefit in terms of increased or healthier wildlife populations. We studied movements of a range of bird species between woodland patches within a high‐altitude Polylepis/matrix landscape in the Cordillera Vilcanota, Peru. Movement rates between Polylepis patches differed across guilds, with arboreal omnivores, arboreal sally‐strikers and nectarivores displaying the highest movement rates, and understorey guilds and arboreal sally‐gleaners the lowest movement rates. Birds tend to avoid flights to more distant neighboring patches, especially when moving from patches which were themselves isolated. The decline in bird flight frequencies with increasing patch isolation followed broken‐stick models most closely, and while we suggest that there is evidence for a decline in between‐patch movements over distances of 30–210 m, there was great variability in movement rates across individual patches. This variability is presumably a result of complex interactions between patch size, quality and configuration, and flight movement patterns of individual bird species. Our study does, however, highlight the contribution small woodland patches make toward fragmented Polylepis ecosystem functioning, and we suggest that, where financial resources permit, small patch restoration would be an important compliment to the restoration of larger woodland patches. Most important is that replanting takes place within 200 m or so of existing larger patches. This will be especially beneficial in allowing more frequent use of woodland elements within the landscape and in improving the total area of woodland patches that are functionally connected.  相似文献

Plant root architecture is regulated by the initiation and modulation of cell division in regions containing pluripotent stem cells known as meristems. In roots, meristems are formed early in embryogenesis, in the case of the root apical meristem (RAM), and during organogenesis at the site of lateral root or, in legumes, nodule formation. Root meristems can also be generated in vitro from leaf explants cultures supplemented with auxin. microRNAs (miRNAs) have emerged as regulators of many key biological functions in plants including root development. To identify key miRNAs involved in root meristem formation in Medicago truncatula, we used deep sequencing to compare miRNA populations. Comparisons were made between: (1) the root tip (RT), containing the RAM and the elongation zone (EZ) tissue and (2) root forming callus (RFC) and non-root forming callus (NRFC). We identified 83 previously reported miRNAs, 24 new to M. truncatula, in 44 families. For the first time in M. truncatula, members of conserved miRNA families miR165, miR181 and miR397 were found. Bioinformatic analysis identified 38 potential novel miRNAs. Selected miRNAs and targets were validated using Taqman miRNA assays and 5′ RACE. Many miRNAs were differentially expressed between tissues, particularly RFC and NRFC. Target prediction revealed a number of miRNAs to target genes previously shown to be differentially expressed between RT and EZ or RFC and NRFC and important in root development. Additionally, we predict the miRNA/target relationships for miR397 and miR160 to be conserved in M. truncatula. Amongst the predictions, were AUXIN RESPONSE FACTOR 10, targeted by miR160 and a LACCASE-like gene, targeted by miR397, both are miRNA/target pairings conserved in other species.  相似文献
