球形棕囊藻(Phaeocystis globosa)是近年来北部湾常见的有害藻华原因种,涠洲岛海域作为广西沿海藻华的高发区之一,目前仍缺乏关于该海域球形棕囊藻藻华过程的相关研究。为探究涠洲岛海域球形棕囊藻藻华过程中营养盐的状况及藻华的发生对浮游植物群落组成的影响,对2017年3月下旬涠洲岛周边发生的球形棕囊藻藻华区域进行取样调查。结果表明,藻华期间水体中溶解有机磷含量与细胞及囊体密度呈显著正相关。本次藻华主要受水体中溶解态无机氮含量影响,多个调查站点的无机氮/无机磷(DIN/DIP)<10,且硅酸盐/无机氮(SiO32–/DIN)>1,表明这些站点存在氮限制现象,而氮限制可能是本次藻华消亡的主要因素。调查区域共鉴定出浮游植物4门58种,球形棕囊藻细胞密度最高可达1.04×108 cells/L,占浮游植物总细胞密度的98.28%。球形棕囊藻细胞密度与物种多样性指数存在显著的负相关关系。在营养缺乏条件下,有利于球形棕囊藻成囊的链状硅藻种类具有相对较大的细胞密度。  相似文献

To understand the mechanism of mutual recognition between aminoacyl tRNA synthetases (EC 6.1.1) and tRNAs, it is important to obtain more information about the structure of these enzymes. X-ray crystallography offers one approach, but although several aminoacyl tRNA synthetases have been isolated in very pure form1 and crystallization of tryptophanyl2 and seryl3 tRNA synthetases has been reported, no crystals were available that were suitable for structural analysis. Only very recently have Rymo and Lagerkvist4 described the formation of crystals of yeast lysyl tRNA synthetase which are apparently suitable for further X-ray analysis. Here we deal with the phenomenon of polymerization of the tryptophanyl tRNA synthetase complex with tryptophan in the form of rod-like particles and the subsequent formation of para-crystalline aggregates.  相似文献

将缺失两端ITR的AAV -2基因组克隆于HSV -1扩增子载体获得了质粒pHSV AAV .以HSV- 1tsK株为辅助病毒感染转染了该质粒的Vero细胞 ,可获得一种具有包装重组AAV质粒能力的混合毒种 .连续传代可使其包装能力迅速升高 ,至 5~ 6代达到最高并趋于平稳 .此混合毒种中 ,含有包装了多拷贝AAV -2基因组的复制缺陷性假型HSV- 1病毒 ,它可表达AAV -2编码的复制与包装所需的各种反式蛋白 ;同时也含有HSV -1tsK病毒 ,它提供AAV -2复制与包装所需的全部辅助病毒功能 .实验结果也表明 ,HSV -1tsK病毒的复制为混合毒种辅助重组AAV载体的包装所必需 .这项研究首次报道了一种全新的重组AAV载体包装系统 ,与传统的双质粒共转染、腺病毒辅助包装方案相比 ,本系统简便易行 ,并明显提高了重组AAV的包装效率 .  相似文献

Temporal auditory mechanisms were measured in killer whales ( Orcinus orca ) by recording auditory evoked potentials (AEPs) to clicks. Clicks were presented at rates from 10/sec to 1,600/sec. At low rates, clicks evoked an AEP similar to the auditory brainstem response (ABR) of other odontocetes; however, peak latencies of the main waves were 3–3.7 msec longer than in bottlenose dolphins. Fourier analysis of the ABR showed a prominent peak at 300–400 Hz and a smaller one at 800–1,200 Hz. High-rate click presentation (more than 100/sec) evoked a rate-following response (RFR). The RFR amplitude depended little on rate up to 400/sec, decreased at higher rates and became undetectable at 1,120/sec. Fourier analysis showed that RFR fundamental amplitude dependence on frequency closely resembled the ABR spectrum. The fundamental could follow clicks to around 1,000/sec, although higher harmonics of lower rates could arise at frequencies as high as 1,200 Hz. Both RFR fundamental phase dependence on frequency and the response lag after a click train indicated an RFR group delay of around 7.5 msec. This corresponds to the latency of ABR waves PIII-NIV, which indicates the RFR originates as a rhythmic, overlapping ABR sequence. The data suggest the killer whale auditory system can follow high click rates, an ability that may have been selected for as a function of high-frequency hearing and the use of rapid clicks in echolocation.  相似文献

Previous reports have shown that neuronal and glial cells express functionally active thrombin receptors. The thrombin receptor (PAR-1), a member of a growing family of protease activated receptors (PARs), requires cleavage of the extracellular amino-terminus domain by thrombin to induce signal transduction. Studies from our laboratory have shown that PAR-1 activation following the addition of thrombin or a synthetic thrombin receptor activating peptide (TRAP) induces motoneuron cell death both in vitro and in vivo. In addition to increasing motoneuron cell death, PAR- 1 activation leads to decreases in the mean neurite length and side branching in highly enriched motoneuron cultures. It has been suggested that motoneuron survival depends on access to sufficient target-derived neurotrophic factors through axonal branching and synaptic contacts. However, whether the thrombininduced effects on motoneurons can be prevented by neurotrophic factors is still unknown. Using highly enriched avian motoneuron cultures, we show here that alone, soluble chick skeletal muscle extracts (CMX), brain-derived neurotrophic factor (BDNF), ciliary neurotrophic factor (CNTF), and glial cell line-derived neurotrophic factor (GDNF) significantly increased motoneuron survival compared to controls, whereas nerve growth factor (NGF) did not have a significant effect on motoneuron survival. Furthermore, cotreatment with muscle-derived agents (i.e., CMX, BDNF, GDNF) significantly prevented the death of motoneurons induced by alpha-thrombin. Yet, non-muscle-derived agents (CNTF and NGF) had little or no significant effect in reversing thrombin-induced motoneuron death. CMX and CNTF significantly increased the mean length of neurites, whereas NGF, BDNF, and GDNF failed to enhance neurite outgrowth compared to controls. Furthermore, CMX and CNTF significantly prevented thrombin-induced inhibition of neurite outgrowth, whereas BDNF and GDNF only partially reversed thrombin-induced inhibition of neurite outgrowth. These findings show differential effects of neurotrophic factors on thrombin-induced motoneuron degeneration and suggest specific overlaps between the trophic and stress pathways activated by some neurotrophic agents and thrombin, respectively.  相似文献

The Chinese crested ibis, Nipponia nippon, is a critically endangered bird. The current population of this species has developed from four wild individuals rediscovered in 1981. Given its conservation status, there is considerable interest in assessing the genetic diversity and individual relatedness in this species. For this purpose, a set of eight polymorphic di‐ or trinucleotide microsatellite loci was developed for the crested ibis. The expected heterozygosity at these loci ranges from 0.01 to 0.50, with less than four alleles being observed at individual loci, a reflection of the serious population bottleneck experienced by this species.  相似文献

Based on the sequence information of Arabidopsis PIN1, two cDNAs encoding PIN homologues from Brassica juncea, Bjpin2 and Bjpin3, were isolated through cDNA library screening. Bjpin2 and Bjpin3 encoded proteins containing 640 and 635 amino acid residues, respectively, which shared 97.5% identities with each other and were highly homologous to Arabidopsis PIN1, PIN2 and other putative PIN proteins. BJPIN2 and BjPIN3 had similar structures as AtPIN proteins. Northern blot analysis indicated that Bjpin2 was expressed in stem, leaf and floral tissues, while Bjpin3 was expressed predominantly in stem and hypocotyls. Two promoter fragments of pin genes, Bjpin-X and Bjpin-Z, were isolated by 'genome walking' technique using primers at 5'-end of pin cDNA. Promoter-gus fusion studies revealed the GUS activities driven by Bjpin-X were at internal side of xylem and petal; while those driven by Bjpin-Z were detected at leaf vein, epidermal cell and cortex of stem, vascular tissues and anther. The induction of heat shock proteins (Hsps) serves not only as a marker for cellular stress but also as a promoter of cell survival, which is especially important in the nervous system. We examined the regulation of the constitutive and stress-induced 70-kD Hsps (Hsc70 and Hsp70, respectively) after sciatic nerve (SN) axotomy in the neonatal mouse. Additionally, the prevention of axotomy-induced SN cell death by administration of several preparations of exogenous Hsc70 and Hsp70 was tested. Immunohistochemistry and Western blot analyses showed that endogenous levels of Hsc70 and Hsp70 did not increase significantly in lumbar motor neurons or dorsal root ganglion sensory neurons up to 24 hours after axotomy. When a variety of Hsc70 and Hsp70 preparations at doses ranging from 5 to 75 microg were applied to the SN stump after axotomy, the survival of both motor and sensory neurons was significantly improved. Thus, it appears that motor and sensory neurons in the neonatal mouse do not initiate a typical Hsp70 response after traumatic injury and that administration of exogenous Hsc/Hsp70 can remedy that deficit and reduce the subsequent loss of neurons by apoptosis.  相似文献
