Spermatogenesis is a highly complicated metamorphosis process of male germ cells. Recent studies have provided evidence that the ubiquitin–proteasome system plays an important role in sperm head shaping, but the underlying mechanism is less understood. In this study, we localized membrane-associated RING-CH (MARCH)7, an E3 ubiquitin ligase, in rat testis. Northern blot analysis showed that March7 mRNA is expressed ubiquitously but highly in the testis and ovary. In situ hybridization of rat testis demonstrated that March7 mRNA is expressed weakly in spermatogonia and its level is gradually increased as they develop. Immunohistochemical analysis detected MARCH7 protein expression in spermiogenic cells from late round spermatids to elongated spermatids and in epididymal spermatozoa. Moreover, MARCH7 was found to be localized to the caudal end of the developing acrosome of late round and elongating spermatids, colocalizing with β-actin, a component of the acroplaxome. In addition, MARCH7 was also detected in the developing flagella and its expression levels were prominent in elongated spermatids. We also showed that MARCH7 catalyzes lysine 48 (K48)-linked ubiquitination. Immunolocalization studies revealed that K48-linked ubiquitin chains were detected in the heads of elongating spermatids and in the acrosome/acroplaxome, neck, midpiece and cytoplasmic lobes of elongated spermatids. These results suggest that MARCH7 is involved in spermiogenesis by regulating the structural and functional integrity of the head and tail of developing spermatids.  相似文献

Cytosolic lipid droplets (LDs) are multifunctional organelles that exist in all types of eukaryotic cells and control lipid homeostasis. In mammalian cells LDs contain a class of proteins in their surface layers that share a homologous sequence called the PAT domain, including perilipin, adipose differentiation-related protein (ADRP), a tail-interacting protein of 47 kDa (TIP47), and S3-12, which are distributed tissue- or cell type-selectively. Expression in some cases is regulated by peroxisome proliferator-activated receptors (PPARs). In this study we identified a new PAT family member named MLDP (myocardial LD protein) in a murine cDNA data base and showed the mRNA and protein to be highly enriched in the heart and also expressed at lower levels in the liver and adrenals. Upon subcellular fractionation, a substantial amount of MLDP was detected in the top fraction enriched with LDs. Furthermore, overexpressed MLDP tagged with green fluorescent protein accumulated at the surfaces of LDs and co-localized with perilipin and ADRP. Deletion analysis demonstrated the N-terminal region containing a PAT-1 domain and the following 33-mer domain to be required for targeting of MLDP to LDs. MLDP was found to be up-regulated at both mRNA and protein levels in the heart and liver by a selective ligand for PPARalpha, Wy14,643, but not in PPARalpha knock-out mice. MLDP expression was also increased upon fasting in parallel with ADRP. These results indicate that MLDP is a bona fide new PAT family member localized in LDs. Its expression depends on the physiological conditions and the action of PPARalpha.  相似文献

House dust mites (HDM) are the most common source of indoor allergens and are associated with allergic diseases worldwide. To benefit allergic patients, safer and non-invasive mucosal routes of oral administration are considered to be the best alternative to conventional allergen-specific immunotherapy. In this study, transgenic rice was developed expressing derivatives of the major HDM allergen Der f 2 with reduced Der f 2-specific IgE reactivity by disrupting intramolecular disulphide bonds in Der f 2. These derivatives were produced specifically as secretory proteins in the endosperm tissue of seeds under the control of the endosperm-specific glutelin GluB-1 promoter. Notably, modified Der f 2 derivatives aggregated in the endoplasmic reticulum (ER) lumen and were deposited in a unique protein body (PB)-like structure tentatively called the Der f 2 body. Der f 2 bodies were characterized by their intracellular localization and physico-chemical properties, and were distinct from ER-derived PBs (PB-Is) and protein storage vacuoles (PB-IIs). Unlike ER-derived organelles such as PB-Is, Der f 2 bodies were rapidly digested in simulated gastric fluid in a manner similar to that of PB-IIs. Oral administration in mice of transgenic rice seeds containing Der f 2 derivatives encapsulated in Der f 2 bodies suppressed Der f 2-specific IgE and IgG production compared with that in mice fed non-transgenic rice seeds, and the effect was dependent on the type of Der f 2 derivative expressed. These results suggest that engineered hypoallergenic Der f 2 derivatives expressed in the rice seed endosperm could serve as a basis for the development of viable strategies for the oral delivery of vaccines against HDM allergy.  相似文献

Growth of Bacillus subtilis AG169 that produced large amounts of xanthosine and guanosine was inhibited by psicofuranine. When AG169 was mutated to resistance against psicofuranine, a mutant, GP–1, which yielded more guanosine was obtained. Psicofuranine did not inhibit growth of GP–1 any more. The guanosine 5′-monophosphate (GMP) synthetase activities were then assayed. In GP–1, the specific activity decreased about half, the complete loss of repression by GMP was found, and the inhibition by GMP was slightly loosed, when compared with those of AG169.

Furthermore, as growth of GP–1 was strongly inhibited by decoyinine, decoyinine resistant mutants were derived from GP–1. Of these mutants, two strains, MG–1 and MG–4, were resistant to decoyinine completely and showed the exclusive accumulation of guanosine in high yields, i.e. 16.0 and 15.5 g of guanosine per liter with weight yields of 20.0 and 19.4% of consumed sugar, respectively. GMP synthetase activity of MG–1 increased remarkably in comparison with that of GP–1 or AG169, and the inhibitions by GMP, psicofuranine and decoyinine were completely released in MG–1. Namely, the psicofuranine and decoyinine resistances seemed to cause mainly variations of GMP synthetase, and as results, the conversion of xanthosine 5′-monophosphate (XMP) to GMP proceeded more smoothly, and a larger amount of guanosine was accumulated.  相似文献

Mammary gland chromatin from lactating rats was fractionated according to the Mg2+-solubility method after mild digestion with DNase II. The chromatin properties were compared between the Mg2+-soluble (S2) and the Mg2+-insoluble (P2) fractions. The weight ratio of RNA to DNA was much greater in the S2 fraction than in the P2 fraction (S2, 0.92; P2, 0.03). The DNA repeat length of the nucleosome was very similar between the two fractions (S2, 193 ± 7; P2, 1% ± 8 nucleotide pairs). A significant difference was seen in electrophoretic pattern of H1 histone between the two fractions in the acid-urea gel system. Nonhistone proteins with molecular weights higher than about 40,000 dalton were found to be enriched in the S2 fraction.  相似文献

As the basic research for the study of mechanism of ventilation in submerged fermentations, the formula of carbon dioxide in the liquid phase was investigated. Carbon dioxide in liquid phase was mainly classified into two groups. One was a gaseous form and the other was a dissociated form. Carbon dioxide was considered to be a weak acids when it dissociated into ionic form, and the dissociation of carbon dioxide was expressed in Henderson-Hasselbalch’s equation. The results in this study showed that those rules concerning the dissolution and dissociation of carbon dioxide held for a model liquid system.  相似文献

The thiol-dependent gelation of conalbumin was highly dependent on both pH and the buffers used. The hardness of the gels was the maximum at pH 8.0 ~8.8. At pH 8.0, it was several time greater in sodium phosphate buffer than in Tris-HCl buffer. Comparison of the effects of inorganic and organic salts revealed that the difference in gel hardness was caused by the difference in anion species. The hardness of the thiol-dependent gel formed under different anion conditions was inversely correlated with the rate of gel formation. Scanning electron microscopic study showed that the gel formed in sodium phosphate appeared as a sponge-like, uniform network structure, while that in Tris-HCl was coarse.  相似文献

This study investigated the effects of working night shifts on social and family life by examining changes in workers' daily life before and after a change in their shifts. Subjects were 40 women aged 27-59 years, working at a dish factory. During the health examination of night workers in autumn of 2000, the subjects were directly interviewed about changes in their lives induced by the shift change. Question parameters consisted of 8 items including 30 sub-items related to social and family life, such as sleep, rest, meals, sports, family time, hobbies, neighborhood association and social activities. The subjects selected one of four response categories: "becoming worse", "no change", "becoming better" and "difficult to determine." With regard to the percentage of "becoming worse", meal-related items ranked high in all of the shift types. "Family time" and "hobbies" showed high percentages in the subjects transferring from day shifts to night shifts, and in those transferring from early-morning shifts to night shifts. "Rest", "sports" and "hobbies" showed high percentages in the subjects transferring from night shifts to midnight shifts. Decreased sleeping hours were confirmed in all of the shift types, while the subjects tended to sleep more soundly. As the workers transfer to shifts at earlier hours, they were obliged to make sacrifices in various aspects of their social and family life. Therefore, much assistance in this regard should be given to them.  相似文献

Three types of straight doughs different in combination of yeast and shortenings (RLS20, FTS20, and FTS80) were prepared, and the structure of the frozen doughs was examined under a microscope after staining protein or lipid droplets. Even after 2 months of frozen storage, distinct changes were not found in the gluten network of FTS80, although significant damages in the dough structures of FTS20 and RLS20 appeared after only one month of frozen storage. These results suggest that the gluten networks loosen and decrease in the water retention ability, and it may be concluded that the lipid is removed from the gluten protein due to the decrease in water in the continuous protein phase. The resulting product from the damage to the gluten matrix gave rise to fusion of lipid droplets and an increase in their size. Because of the difference in fatty acid composition, the lipids of shortening S80 are presumed to interact more strongly with gluten proteins and to keep the gluten matrix from damage in comparison with the lipids of shortening S20.  相似文献
