Autotransporters (ATs) of Gram-negative bacteria contain an N-proximal passenger domain that is transported to the extracellular milieu and a C-terminal β-domain that inserts into the outer membrane (OM) in a β-barrel conformation. This β-domain facilitates translocation of the passenger domain across the OM and has long been considered to be the translocation pore. However, available crystal structures of β-domains show that the β-barrel pore is too narrow for the observed transport of folded elements within the passenger domains. ATs have recently been shown to interact with the β-barrel assembly machinery. These findings questioned a direct involvement of the β-domain in passenger translocation and suggested that it may only target the passenger to the β-barrel assembly machinery pore. To address the function of the β-domain in more detail, we have replaced the β-domain of the Escherichia coli AT hemoglobin protease by β-domains originating from other OM proteins. Furthermore, we have modified the diameter of the β-domain pore. The mutant proteins were analyzed for their capacity to insert into the OM and for surface display of the passenger. Our results show that efficient passenger secretion requires a specific β-domain that not only functions as a targeting device but also is directly involved in the translocation of the passenger to the cell surface.  相似文献

The cAMP-dependent protein kinase [protein kinase A (PKA)] mediates a myriad of cellular signaling events, and its activity is tightly regulated in both space and time. Among these regulatory mechanisms is N-myristoylation, whose biological role has been elusive. Using a combination of thermodynamics, kinetics, and spectroscopic methods, we analyzed the effects of N-myristoylation and phosphorylation at Ser10 on the interactions of PKA with model membranes. We found that, in the absence of lipids, the myristoyl group is tucked into the hydrophobic binding pocket of the enzyme (myr-in state). Upon association with lipid bilayers, the myristoyl group is extruded and inserts into the hydrocarbon region of the lipid bilayer (myr-out state). NMR data indicate that the enzyme undergoes conformational equilibrium between myr-in and myr-out states, which can be shifted byeither interaction with membranes and/or phosphorylation at Ser10. Our results provide evidence that the membrane binding motif of the myristoylated C-subunit of PKA (PKA-C) steers the enzyme toward lipids independent of its regulatory subunit or an A-kinase anchoring protein, providing an additional mechanism to localize the enzyme near membrane-bound substrates.  相似文献

Sterically highly hindered phosphiniminato complexes MCl3(NCP) were prepared from MCl4 and Li[NPC] in toluene [M = Zr or Hf; NPC = 4-ButC6H4C(SiMe3)P(Ph)2NC6H2Me3-2,4,6]. Reaction with methyl lithium readily affords the corresponding zirconium and hafnium trimethyl complexes. The structures of representative zirconium and hafnium complexes MX3(NPC) (X = Cl, M = Zr, Hf; X = Me, M = Hf) were determined by X-ray diffraction. In all cases the NPC ligand acts as C-N chelate, with an additional bonding contribution from the ipso-carbon atom of the C-bound aryl substituent, which results in a η12-coordination mode. The reaction of the hafnium trimethyl complex with salts of perfluoroarylborate anions results either in the diastereoselective formation of the binuclear cation [{(NPC)HfMe2}2(μ-Me)]+ or in the formation of the mononuclear cation [(NPC)HfMe2]+, depending on the molar ratio of reagents.  相似文献

Oxidative stress (OS) and its consequences which promote alterations in biomolecules, to tissue damage and to the development of pathological conditions, continue to attract many investigators. The identification of reliable biomarker is essential for the characterization of OS and possibly for early discovery of OS-associated diseases. The aim of the present study was to offer a new concept in the development of novel probes for OS, based on the design, synthesis, and utilization of exogenous markers, as alternative to the search for endogenous markers. This article describes: (a) the synthesis of such a marker, linoleoyl tyrosine 2-deoxyguanosyl ester (LTG), constructed from three endogenous subunits: linoleic acid, tyrosine, and 2'-deoxyguanosine, representing the three major groups from which the body is composed, unsaturated fatty acids (USFA), proteins, and DNA, respectively, all bound covalently and (b) the development of analytical tools (LC/MS/MS) to enable the identification of the different LTG oxidized products formed under OS by exposure of LTG to different reactive oxygen species (ROS) such as, copper ions and hypochlorous acid.  相似文献

Previous studies about geographic patterns of species diversity of avian malaria parasites and others in the Order Haemosporida did not include the avian biodiversity hotspot Madagascar. Since there are few data available on avian malaria parasites on Madagascar, we conducted the first known large-scale molecular-based study to investigate their biodiversity. Samples (1067) from 55 bird species were examined by a PCR method amplifying nearly the whole haemosporidian cytochrome b gene (1063?bp). The parasite lineages found were further characterized phylogenetically and the degree of specialization was determined with a newly introduced host diversity index (Hd). Our results demonstrate that Madagascar indeed represents a biodiversity hotspot for avian malaria parasites as we detected 71 genetically distinct parasite lineages of the genera Plasmodium and Haemoproteus. Furthermore, by using a phylogenetic approach and including the sequence divergence we suspect that the detected haemosporidian lineages represent at least 29 groups i.e. proposed species. The here presented Hd values for each parasite regarding host species, genus and family strongly support previous works demonstrating the elastic host ranges of some avian parsites of the Order Haemosporida. Representatives of the avian parasite genera Plasmodium and Leucocytozoon tend to more often be generalists than those of the genus Haemoproteus. However, as demonstrated in various examples, there is a large overlap and single parasite lineages frequently deviate from this rule.  相似文献

MicroRNAs (miRNAs) are active regulators of numerous biological and physiological processes including most of the events of mammalian reproduction. Understanding the biological functions of miRNAs in the context of mammalian reproduction will allow a better and comparative understanding of fertility and sterility in male and female mammals. Herein, we summarize recent progress in miRNA‐mediated regulation of mammalian reproduction and highlight the significance of miRNAs in different aspects of mammalian reproduction including the biogenesis of germ cells, the functionality of reproductive organs, and the development of early embryos. Furthermore, we focus on the gene expression regulatory feedback loops involving hormones and miRNA expression to increase our understanding of germ cell commitment and the functioning of reproductive organs. Finally, we discuss the influence of miRNAs on male and female reproductive failure, and provide perspectives for future studies on this topic.  相似文献

Proper trafficking of membrane-bound ion channels and transporters is requisite for normal cardiac function. Endosome-based protein trafficking of membrane-bound ion channels and transporters in the heart is poorly understood, particularly in vivo. In fact, for select cardiac cell types such as atrial myocytes, virtually nothing is known regarding endosomal transport. We previously linked the C-terminal Eps15 homology domain-containing protein 3 (EHD3) with endosome-based protein trafficking in ventricular cardiomyocytes. Here we sought to define the roles and membrane protein targets for EHD3 in atria. We identify the voltage-gated T-type Ca2+ channels (CaV3.1, CaV3.2) as substrates for EHD3-dependent trafficking in atria. Mice selectively lacking EHD3 in heart display reduced expression and targeting of both Cav3.1 and CaV3.2 in the atria. Furthermore, functional experiments identify a significant loss of T-type-mediated Ca2+ current in EHD3-deficient atrial myocytes. Moreover, EHD3 associates with both CaV3.1 and CaV3.2 in co-immunoprecipitation experiments. T-type Ca2+ channel function is critical for proper electrical conduction through the atria. Consistent with these roles, EHD3-deficient mice demonstrate heart rate variability, sinus pause, and atrioventricular conduction block. In summary, our findings identify CaV3.1 and CaV3.2 as substrates for EHD3-dependent protein trafficking in heart, provide in vivo data on endosome-based trafficking pathways in atria, and implicate EHD3 as a key player in the regulation of atrial myocyte excitability and cardiac conduction.  相似文献

The pollen morphology of the ten taxa belonging to Cassubicae Radzhi, Cracca Dumort., Ervilia (Link) Koch, Lentopsis Kupicha, Trigonellopsis Rech. f. and Variegatae Radzhi sections of the genus Vicia L. subgenus Vicilla (Schur) Rouy (Fabeae, Fabaceae) in Turkey has been examined by use of light microscopy and scanning electron microscopy. Twelve morphometric characters are analysed with one-way analysis of variance and Tukey’s honestly significant difference test for multiple comparisons. Polar axis and equatorial diameter of pollen grains range from 24.64 to 44.19 and from 15.90 to 30.42 μm, respectively. The pollen grains are prolate–spheroidal to perprolate (P/E = 1.09–2.07), but the prolate shape occurs in the majority of the taxa. The regular pollen grains of all taxa are trizonocolporate, isopolar, and released in monads. Ornamentation of the mesocolpium is psilate–perforate in V. tenuifolia subsp. dalmatica and V. tenuifolia subsp. tenuifolia (sect. Cracca), reticulate–rugulate in V. villosa subsp. villosa (sect. Cracca), and reticulate–perforate in the remaining taxa. The apocolpium and colpus area are reticulate–perforate in V. hirsuta (sect. Cracca), V. ervilia (sect. Ervilia) and V. lunata subsp. grandiflora (sect. Trigonellopsis), and psilate, psilate–slightly perforate or perforate in the other taxa. The results obtained from analyses of pollen grains from the studied taxa of the subgenus Vicilla reveal significant differences in general morphology and some morphometric attributes, and ornamentation types. However, it is concluded that pollen morphology has proved to be uninformative for differentiating the subgenera Vicilla and Vicia.  相似文献

Compounds, which inhibit tyrosinase, could be effective as depigmenting agents. We have introduced a group of mono-, di-, tri- and tetra-substituted hydroxychalcones as effective tyrosinase inhibitors, showing that the most important factor determining tyrosinase inhibition efficiency is the position of the hydroxyl group(s) rather their number. The aim of the present study was to investigate the contribution of the different functional groups of the tetrahydroxychalcones to their inhibitory potency, with a view to optimizing the design of whitening agents. Four tetrahydroxychalcones were evaluated, the commercially available Butein and other three were synthesized, and their inhibitory effect on tyrosinase was tested. Results showed that a 2,4-substituted resorcinol subunit on ring B contributed the most to inhibitory potency. Changing the resorcinol substitute to position 3,5- or placing it on ring A significantly diminished the inhibitory effect of the compounds. A catechol subunit on ring A acted as a metal chelator (in the presence of copper ions) and as a competitive inhibitor (in the presence of tyrosinase), while a catechol on ring B oxidized to o-quinone (in the presence of both copper ions and tyrosinase). Three of the compounds also demonstrated antioxidant activity, which may contribute to the prevention of pigmentation. An examination of correlations between inhibitory activity and physical properties of the chalcones tested (such as dissociation energy and molecular planarity) showed positive correlation with the moment dipole value in the Y-axis, which may be used as an indicator of the inhibitory potential of new molecules. The present study revealed two very active tyrosinase inhibitors, 2,4,3',4'-hydroxychalcone and 2,4,2',4'-hydroxychalcone (with IC50 of 0.2 and 0.02 microM, respectively). Structure-related activity studies added some understanding of the role and contribution of different functional groups associated with tyrosinase inhibitors.  相似文献
