A survey is given of physical, chemical and biological events in the water-mass of a productive lake (Esthwaite Water, N. England) during summer stratification. They are interrelated in terms of i) responses to meteorological changes (ii) susceptibility to density stratification (iii) participation in acid-base and redox systems, with interactions of these (iv) modification due to the generation of particulate material from soluble precursors (v) limiting factors in such bio-generation (vi) mechanisms of internal nutrient loading, and (vii) control by the characteristics of individual plankters. The significance is demonstrated of differential inputs of meteorological variables, and especially of biological production and degradation, for determining the overall structure and metabolism of the lake.  相似文献

The attraction of small gadoids to physically disturbed seabed patches was examined experimentally using a modified remote operated vehicle. Video observations were made of simulated disturbances to investigate the influence of tidal state, time of day and current speed on the pattern of exploitation. Fish were more likely to be present in disturbed patches, and when present, occurred in significantly greater numbers than in control areas. However, this response was dependent on tidal state, occurring only on rising tides.  相似文献

Adenylate cyclase activity was assayed in a crude particulate fraction of one benign and one malignant human insulinoma. Adenylate cyclase of both tumours responded to 5'-guanylyl-imidodiphosphate, sodium fluoride, glucagon and prostaglandin E2, and in addition the adenylate cyclase of the benign tumour responded to isoprenaline. Glucose and prostaglandin I2 (prostacyclin) did not stimulate the adenylate cyclase in either tumour, although prostaglandin I2 stimulated insulin secretion in cultures of the benign tumour. The in vitro responsiveness of the adenylate cyclase to glucagon did not correlate closely with the effect of glucagon on insulin secretion in vivo.  相似文献

Activated B cells and T cells express CD154/CD40 ligand in vitro. The in vivo expression and function of B cell CD154 remain unclear and therefore were examined. Tonsillar B and T cells expressed CD154 at a similar density both in situ and immediately ex vivo, whereas a significantly higher percentage of the former expressed CD154. CD154-expressing B cells were most frequent in the CD38positiveIgD+ pre-germinal center (GC)/GC founder, CD38positive GC and CD38-IgD- memory populations, and were also found in the CD38-IgD+ naive and CD38brightIgD+ plasmablast subsets, but not in the CD38brightIgD- plasma cell subset. B cell expression of CD154 was induced by engaging surface Ig or CD40 by signals that predominantly involved activation of AP-1/NF-AT and NF-kappaB, respectively. The functional importance of CD154-mediated homotypic B cell interactions in vivo was indicated by the finding that mAb to CD154 inhibited differentiation of CD38positiveIgD- GC B cells to CD38-IgD- memory cells. In addition, mAb to CD154 inhibited proliferation induced by engaging sIg or CD40, indicating the role of up-regulation of this molecule in facilitating B cell responsiveness. Of note, CD154 itself not only functioned as a ligand but also as a direct signaling molecule as anti-CD154-conjugated Sepharose beads costimulated B cell responses induced by engaging surface Ig. These results indicate that CD154 is expressed by human B cells in vivo and plays an important role in mediating B cell responses.  相似文献

Previous studies have suggested that increases in skin blood flow (SkBF) are the primary physiological mechanism responsible for cardiovascular drift during exercise in the heat. Most of these studies, however, used exercise bouts of 60 min in duration or less. The purpose of this study was to explore the possibility that during prolonged (> 60 min) exercise in the heat, cardiovascular drift can occur without a concomitant increase in SkBF. The subjects were five heat-acclimated female volunteers. Each subject completed a 6-h heat exposure (38oC, 62% RH). Heart rate (HR), stroke volume (SV), and two independent measures of SkBF were obtained each hour. Cardiovascular drift occurred, as evidenced by a significant (p<0.05) 19% increase in HR and a significant 21% decrease in SV. Interestingly, however, SkBF plateaued by hour 2 and showed no further increase. Such results suggest that during prolonged exercise in the heat, when SkBF has reached very high values (>20 ml / 100 ml per min) and plateaued, cardiovascular drift can still occur.  相似文献
