Climate change-integrated conservation strategies   总被引:8,自引:1,他引:7  
Aim Conservation strategies currently include little consideration of climate change. Insights about the biotic impacts of climate change from biogeography and palaeoecology, therefore, have the potential to provide significant improvements in the effectiveness of conservation planning. We suggest a collaboration involving biogeography, ecology and applied conservation. The resulting Climate Change‐integrated Conservation Strategies (CCS) apply available tools to respond to the conservation challenges posed by climate change. Location The focus of this analysis is global, with special reference to high biodiversity areas vulnerable to climate change, particularly tropical montane settings. Methods Current tools from climatology, biogeography and ecology applicable to conservation planning in response to climate change are reviewed. Conservation challenges posed by climate change are summarized. CCS elements are elaborated that use available tools to respond to these challenges. Results Five elements of CCS are described: regional modelling; expanding protected areas; management of the matrix; regional coordination; and transfer of resources. Regional modelling uses regional climate models, biotic response models and sensitivity analysis to identify climate change impacts on biodiversity at a regional scale appropriate for conservation planning. Expansion of protected areas management and systems within the planning region are based on modelling results. Management of the matrix between protected areas provides continuity for processes and species range shifts outside of parks. Regional coordination of park and off‐park efforts allows harmonization of conservation goals across provincial and national boundaries. Finally, implementation of these CCS elements in the most biodiverse regions of the world will require technical and financial transfer of resources on a global scale. Main conclusions Collaboration across disciplines is necessary to plan conservation responses to climate change adequately. Biogeography and ecology provide insights into the effects of climate change on biodiversity that have not yet been fully integrated into conservation biology and applied conservation management. CCS provide a framework in which biogeographers, ecologists and conservation managers can collaborate to address this need. These planning exercises take place on a regional level, driven by regional climate models as well as general circulation models (GCMs), to ensure that regional climate drivers such as land use change and mesoscale topography are adequately represented. Sensitivity analysis can help address the substantial uncertainty inherent in projecting future climates and biodiversity response.  相似文献

Mouse NK cells express Ly-49 receptors specific for classical MHC class I molecules. Several of the Ly-49 receptors have been characterized in terms of function and ligand specificity. However, the only Ly-49 receptor-ligand interaction previously described in detail is that between Ly-49A and H-2D(d), as studied by point mutations in the ligand and the crystal structure of the co-complex of these molecules. It is not known whether other Ly-49 receptors bind MHC class I in a similar manner as Ly-49A. Here we have studied the effect of mutations in Ly-49C on binding to the MHC class I molecules H-2K(b), H-2D(b), and H-2D(d). The MHC class I molecules were used as soluble tetramers to stain transiently transfected 293T cells expressing the mutated Ly-49C receptors. Three of nine mutations in Ly-49C led to loss of MHC class I binding. The three Ly-49C mutations that affected MHC binding correspond to Ly-49A residues that are in contact or close to H-2D(d) in the co-crystal, demonstrating that MHC class I binding by Ly-49C is dependent on residues in the same area as that used by Ly-49A for ligand contacts.  相似文献

Concern for climate change has not yet been integrated in protocols for reserve selection. However if climate changes as projected, there is a possibility that current reserve‐selection methods might provide solutions that are inadequate to ensure species' long‐term persistence within reserves. We assessed, for the first time, the ability of existing reserve‐selection methods to secure species in a climate‐change context. Six methods using a different combination of criteria (representation, suitability and reserve clustering) are compared. The assessment is carried out using European distributions of 1200 plant species and considering two extreme scenarios of response to climate change: no dispersal and universal dispersal. With our data, 6–11% of species modelled would be potentially lost from selected reserves in a 50‐year period. Measured uncertainties varied in 6% being 1–3% attributed to dispersal assumptions and 2–5% to the choice of reserve‐selection method. Suitability approaches to reserve selection performed best, while reserve clustering performed poorly. We also found that 5% of species modelled would lose their entire climatic envelope in the studied area; 2% of the species modelled would have nonoverlapping distributions; 93% of the species modelled would maintain varying levels of overlapping distributions. We conclude there are opportunities to minimize species' extinctions within reserves but new approaches are needed to account for impacts of climate change on species; especially for those projected to have temporally nonoverlapping distributions.  相似文献

The recent discovery of thrombopoietin has enhanced our understanding of both hematopoiesis and platelet production. Thrombopoietin supports hematopoietic stem cell survival and expansion as well as promoting all aspects of megakaryocyte development. The hormone displays many structural similarities to other members of the hematopoietic cytokine family and some notable differences, and regulation of its expression requires both receptor-mediated removal and other mechanisms. Thrombopoietin induces receptor dimerization and tyrosine phosphorylation, and a series of signaling events including activation of JAK/STAT, Shc/Ras/MAPK and PI3K/Akt; these pathways overlap with those induced by other cytokines, but the differences that lead to the unique biological effects of the hormone are gradually being uncovered. Our growing appreciation of how cytokine signaling pathways are translated into megakaryocyte development is discussed.  相似文献

This article is about the beginnings of tissue culture—the culture of living, reproducing cells of complex organisms outside the body. It argues that Ross Harrison’s experiments in nerve culture between 1907 and 1910 should be viewed as part of a larger shift in early twentieth-century laboratory practice from in vivo to in vitro experimentation. Via a focus on the temporality of experiment—contrasting the live object of Harrison’s investigation with the static object of histological representations—this article details the production of a new and surprising form of life, cellular life in vitro. Tissue culture, developed from Harrison’s experiments, was greeted with great surprise and disbelief, despite Harrison’s protestations that he had merely juxtaposed extant techniques. An analysis of these initial reactions to tissue culture illuminates the extent to which cells living visibly outside of the body in glass broke with in vivo practices and assumptions of the hiddenness and interiority of certain processes of growth and change.  相似文献

Environmental enrichment may affect infant care and development in captive primates. We investigated the effects of this factor in laboratory common marmosets (Callithrix jacchus). An enriched physical environment enhanced the social activities of the marmosets and elicited a greater repertoire of behaviors, without negatively affecting the provision of infant care. In addition, infants in enriched cages displayed certain behaviors sooner than infants in non-enriched cages did, which suggests an increased developmental rate. Infants in enriched cages also ate more solid food and engaged in solitary play and exploration more than ones in non-enriched cages did. Play and exploration probably improve spatial cognition and motor skills, which, together with a higher degree of independence, may allow infants to cope better with laboratory routines and general social interactions later in life than their counterparts reared in less complex enclosures. We conclude that laboratories can significantly increase the welfare of marmosets by providing a more complex physical environment.  相似文献

BACKGROUND: The ability of a cell to polarize and move is governed by remodeling of the cellular adhesion/cytoskeletal network that is in turn controlled by the Rho family of small GTPases. However, it is not known what signals lie downstream of Rac1 and Cdc42 during peripheral actin and adhesion remodeling that is required for directional migration. RESULTS: We show here that individual members of the Rho family, RhoA, Rac1, and Cdc42, direct the specific intracellular targeting of c-Src tyrosine kinase to focal adhesions, lamellipodia, or filopodia, respectively, and that the adaptor function of c-Src (the combined SH3/SH2 domains coupled to green fluorescent protein) is sufficient for targeting. Furthermore, Src's catalytic activity is absolutely required at these peripheral cell-matrix attachment sites for remodeling that converts RhoA-dependent focal adhesions into smaller focal complexes along Rac1-induced lamellipodia (or Cdc42-induced filopodia). Consequently, cells in which kinase-deficient c-Src occupies peripheral adhesion sites exhibit impaired polarization toward migratory stimuli and reduced motility. Furthermore, phosphorylation of FAK, an Src adhesion substrate, is suppressed under these conditions. CONCLUSIONS: Our findings demonstrate that individual Rho GTPases specify Src's exact peripheral localization and that Rac1- and Cdc42-induced adhesion remodeling and directed cell migration require Src activity at peripheral adhesion sites.  相似文献

1. This paper presents a framework for the analysis of glacial stream ecosystems based upon the hierarchy of physical processes that operate over timescales varying from diel to millennial. Linked conceptual models propose interrelationships between climate, hydrology and fluvial geomorphology in alpine catchments.
2. These conceptual models are illustrated using data from the Taillon/Gabiétous catchment, French Pyrénées. Secondary sources provide information concerning the long-term sequence of climatic and geomorphological controls on contemporary catchment processes. Detailed hydrogeomorphological field data collected over three consecutive summer melt seasons (1995–97) permit identification of marked changes in shorter-term (diel, seasonal and inter-annual) physical processes.
3. Clear differences in the response of water quality and quantity variables were observed between years as climatic conditions varied. In two of the three study years, a precipitation-driven regime was imposed upon the typical ablation-driven river discharge pattern in alpine streams. Clear changes in water quality and quantity were evident with increasing distance from the glacier: (i) discharge increased although specific discharge decreased markedly, (ii) the mean and variability in water temperature increased, and (iii) base concentrations of suspended solids decreased.
4. The physical processes incorporated in the conceptual models presented in this paper have ecological implications because they underpin a nested suite of disturbance regimes operating over timescales from diel to millennial.  相似文献

RNA hydrolysis presents problems in manufacturing, long-term storage, world-wide delivery and in vivo stability of messenger RNA (mRNA)-based vaccines and therapeutics. A largely unexplored strategy to reduce mRNA hydrolysis is to redesign RNAs to form double-stranded regions, which are protected from in-line cleavage and enzymatic degradation, while coding for the same proteins. The amount of stabilization that this strategy can deliver and the most effective algorithmic approach to achieve stabilization remain poorly understood. Here, we present simple calculations for estimating RNA stability against hydrolysis, and a model that links the average unpaired probability of an mRNA, or AUP, to its overall hydrolysis rate. To characterize the stabilization achievable through structure design, we compare AUP optimization by conventional mRNA design methods to results from more computationally sophisticated algorithms and crowdsourcing through the OpenVaccine challenge on the Eterna platform. We find that rational design on Eterna and the more sophisticated algorithms lead to constructs with low AUP, which we term ‘superfolder’ mRNAs. These designs exhibit a wide diversity of sequence and structure features that may be desirable for translation, biophysical size, and immunogenicity. Furthermore, their folding is robust to temperature, computer modeling method, choice of flanking untranslated regions, and changes in target protein sequence, as illustrated by rapid redesign of superfolder mRNAs for B.1.351, P.1 and B.1.1.7 variants of the prefusion-stabilized SARS-CoV-2 spike protein. Increases in in vitro mRNA half-life by at least two-fold appear immediately achievable.  相似文献
