Roads are detrimental to wildlife populations that require contiguous networks of terrestrial and aquatic habitats. Many species of freshwater turtles are sensitive to habitat fragmentation caused by roads, and are susceptible to road mortality during overland migrations. The common long‐necked turtle (Chelodina longicollis) is an Australian freshwater turtle that frequently moves between wetlands, and so populations may incur negative impacts from road effects. Here, we assessed the relationship between C. longicollis and road density and landscape variables within populations inhabiting 20 wetlands distributed throughout greater Melbourne, Australia. The size frequency distribution of C. longicollis at sites surrounded by high road densities was skewed towards larger individuals, but there was no difference in the frequency of juveniles between high and low road density sites. Regression modelling revealed a clear positive relationship between road density and carapace length (CL) of C. longicollis; the mean CL at a site with the highest road density was predicted to be 23% greater than mean CL at a site surrounded by no roads. Female CL was also positively related to road density. There was a clear positive relationship between wetland age and CL, although this relationship was not as strong. While there was no relationship evident between road density and the proportion of female C. longicollis at a site, more females were captured at smaller ephemeral sites surrounded by a high proportion of green open space and located near drainage lines. We did not find evidence of sex‐related differences in road effects. These results suggest that roads may be affecting C. longicollis in the study area, but the direct cause of any effects is difficult to identify.  相似文献

We describe the cloning and characterization of the mouse MOK-2 gene, a new member of the Krüppel family of zinc finger proteins. Sequencing of both cDNA and genomic clones showed that the predicted MOK-2 protein consists of seven zinc finger domains with only five additional amino acids. The finger domains of MOK-2 are highly homologous to one another but not to those of other zinc finger proteins. MOK-2 is preferentially expressed in transformed cell lines, brain tissue, and testis tissue. Its possible role in cellular transformation is discussed.  相似文献

Most of the data concerning heat shock gene expression reported in the literature are derived from batch culture experiments under substrate and nutrient sufficient conditions. Here, the effects of dilution rate and medium composition on the steady state and heat shock induced htpG gene expression have been investigated in continuous cultures of Escherichia coli, using a chromosomal htpG-lacZ gene fusion. During steady state growth temperature dependent patterns of the relative htpG expression were found to be largely similar, irrespective of the growth condition. However, nitrogen-limited growth resulted in a markedly reduced specific steady state htpG expression as compared to growth under carbon limitation or in complex medium, correlating qualitatively with the total cellular protein content. During heat shock, tight temperature controlled expression was evident. While the relative heat shock induced expression was largely identical at various dilution rates in a given growth medium, significantly different response patterns were observed in the three growth media at any give dilution rate. From these results a clearly temperature regulated htpG expression during both, steady and transient state growth in continuous culture is evident, which is further significantly affected by the growth condition used.  相似文献

We have isolated two metallothionein (MT) cDNA clones copied from the RNA of cadmium-resistant monkey kidney cells. The complete DNA sequences of these clones show that they encode two distinct MTs. One clone appears to represent monkey MT-II, as shown by its close homology to the human MT-II sequence, whereas the second may correspond to monkey MT-I or a related variant metallothionein. Conserved sequences were identified in both the 5′ and 3′ untranslated regions of these clones.  相似文献

We compared three unstructured mathematical models, the master reaction, the square root, and the damage/repair models, for describing the relationship between temperature and the specific growth rates of bacteria. The models were evaluated on the basis of several criteria: applicability, ease of use, simple interpretation of model parameters, problem-free determination of model parameters, statistical evaluation of goodness of fit (chi 2 test), and biological relevance. Best-fit parameters for the master reaction model could be obtained by using two consecutive nonlinear least-square fits. The damage/repair model proved to be unsuited for the data sets considered and was judged markedly overparameterized. The square root model allowed nonproblematical parameter estimation by a nonlinear least-square procedure and, together with the master reaction model, was able to describe the temperature dependence of the specific growth rates of Klebsiella pneumoniae NCIB 418, Escherichia coli NC3, Bacillus sp. strain NCIB 12522, and the thermotolerant coccobacillus strain NA17. The square root and master reaction models were judged to be equally valid and superior to the damage/repair model, even though the square root model is devoid of a conceptual basis.  相似文献

Phyllopod populations were monitored in three temporary pools differing in the amount of submerged, peripheral vegetation present, surface area and duration. The effects of these factors on the life history strategies employed by phyllopods were investigated. Triops granarius, various conchostracan species and the anostracan Branchipodopsis sp. inhabited the periphery of two pools where rooted, submerged vegetation was abundant while three Streptocephalus species dominated the central, unvegetated regions of the pools and the unvegetated pool. This distribution pattern appeared to be related to the animals' morphology and feeding habits. The peripheral regions of the pools were stressful habitats since they were colonized by large numbers of predators and competitors 30–40 days after inundation and they dried out sooner than the centre. The peripheral species exhibited typical r-selected life history strategies; they grew rapidly, reproduced early and had short lifespans and in this way they overcame the threats presented by their habitat. The central species took advantage of their predator-free, more stable habitat and exhibited life history patterns which tended towards the K-end of the r-K continuum. A degree of intraspecific variation in growth and reproduction was obvious and appeared to be related to differences in habitat duration of the three pools.  相似文献

The poor definition of pathotype variation in the rice blast fungus has historically handicapped strategies for reducing blast disease damage to the world's rice crop. We have employed a probe for a dispersed repeated DNA sequence called MGR [Hamer et al. (1989). Proc. Natl. Acad. Sci. USA 86, 9981-9985] to construct genotype-specific, EcoRl restriction fragment length profiles (MGR-DNA fingerprints) from United States field isolates of this fungus. By using a blind-test design, we demonstrated that MGR-DNA fingerprints distinguished the major pathotypes in the United States, accurately identified the pathotypes of isolates collected over a 30-year period, and defined the organization of clonal lineages within and among pathotype groups. These results resolved a lingering controversy regarding rice blast pathotype stability and illustrated new opportunities for tracking the population dynamics and evolution of this important crop pathogen.  相似文献

After a single injection,65Zn is slowly taken up by the brain of the rat to a maximum after 7 d, followed by a turnover phase, with a half-time of about 3 wk. In the brain of rats on a zinc-deficient diet, the65Zn content in the brain continued to increase up to 30 d after the injection. The uptake and turnover phases in six different subcellular fractions of the brain showed a pattern similar to that of the whole brain in both the control and zinc-deficient rats. There was no internal redistribution of65Zn in the brain under conditions of progressive zinc deficiency. The results are discussed in a model for zinc homeostasis in the brain.  相似文献
