Creatine kinase (CK) forms a small family of isoenzymes playing an important role in maintaining the concentration of ATP and ADP in muscle cells. To delineate the impact of a lack of CK activity, we studied contractile performance during a single maximal tetanic contraction and during 12 repeated tetanic contractions of intact dorsal flexors of CK knockout (CK(-/-)) mice. To investigate the effect on ATP regeneration, muscular high-energy phosphate content was determined at rest, immediately after the contraction series, and after a 60-s recovery period. Maximal torque of the dorsal flexors was significantly lower in CK(-/-) mice than in wild-type animals, i.e., 23.7 +/- 5.1 and 33.3 +/- 6.8 mN. m. g(-1) wet wt, respectively. Lower muscle ATP (20.1 +/- 1.4 in CK(-/-) vs. 28.0 +/- 2.1 micromol/g dry wt in controls) and higher IMP (1.2 +/- 0.5 in CK(-/-) vs. 0.3 +/- 0.1 micromol/g dry wt in controls) levels at the onset of contraction may contribute to the declined contractility in CK(-/-) mice. In contrast to wild-type muscles, ATP levels could not be maintained during the series of 12 tetanic contractions of dorsal flexors of CK(-/-) mice and dropped to 15.5 +/- 2.4 micromol/g dry wt. The significant increase in tissue IMP (2.4 +/- 1.1 micromol/g dry wt) content after the contraction series indicates that ATP regeneration through adenylate kinase was not capable of fully compensating for the lack of CK. ATP regeneration via the adenylate kinase pathway is a likely cause of reduced basal adenine nucleotide levels in CK(-/-) mice.  相似文献

We studied the role of the ubiquitin-proteasome system in rat skeletal muscle during sepsis and subsequent recovery. Sepsis was induced with intraperitoneal zymosan injections. This model allows one to study a sustained and reversible catabolic phase and mimics the events that prevail in septic and subsequently recovering patients. In addition, the role of the ubiquitin-proteasome system during muscle recovery is poorly documented. There was a trend for increased ubiquitin-conjugate formation in the muscle wasting phase, which was abolished during the recovery phase. The trypsin- and chymotrypsin-like peptidase activities of the 20S proteasome peaked at day 6 following zymosan injection (i.e. when both muscle mass and muscle fiber cross-sectional area were reduced the most), but remained elevated when muscle mass and muscle fiber cross-sectional area were recovering (11 days). This clearly suggests a role for the ubiquitin-proteasome pathway in the muscle remodeling and/or recovery process. Protein levels of 19S complex and 20S proteasome subunits did not increase throughout the study, pointing to alternative mechanisms regulating proteasome activities. Overall these data support a role for ubiquitin-proteasome dependent proteolysis in the zymosan septic model, in both the catabolic and muscle recovery phases.  相似文献

It is desirable to determine wheter a cow has failed to become pregnant as early as possible, preferably prior to 50 d after insemination. Although palpation per rectum has been the generally accepted method of pregnancy diagnosis in cattle, the procedure may be a significant iatrogenic cause of fetal attrition. In a study conducted at a North Florida dairy from January through June 1982, pregnancy was determined in 192 Holstein-Friesian cows by measuring low milk progesterone (P4) content on day of insemination (Day 0) and elevated P4 on Days 21 and 24. Pregnant cows were randomly assigned to a treatment and a control group. Cows in the treatment group (n = 85) were palpated per rectum twice between Days 42 and 46 after insemination. Cows in the control group (n = 107) were not palpated until both groups were palpated at Day 90. Palpation, done by two experienced clinicians, consisted of palpation of fetal fluid fluctuation, identification of the amniotic vesicle, and slipping of the chorioallantoic membranes. In both groups of cows fetal viability was monitored by milk P4 content. Last milk (5 to 15 ml) was collected from one front quarter on Days 0, 21, and 24 and twice weekly thereafter through Day 63. Milk was defatted by centrifugation and the fat-free milk progesterone content measured by a radioimmunoassay without extraction. The milk P4 test was 80.0% accurate in determining pregnancy in the palpated and nonpalpated cows. In the cows palpated on Days 42 to 46, pregnancy rates declined by 7.5% as determined by palpation at Day 90, or by 11.4% as determined by milk P4 content through Day 63 (both values P < 0.05). Cows that were not palpated on Days 42 to 46 showed a 1.9% increase or a 4.3% decline in pregnancy rates as determined by the same criteria. Before palpation, at Days 42 to 46, pregnancy rates were better in cows that were inseminated in winter (January to March) than in spring (April to June) (82.3% vs 61.6%; P < 0.05); P4 content was higher (winter > spring = 2.13 ng/ml vs 1.38 ng/ml; P < 0.05). First-lactation cows had higher P4 values on Days 21 and 24 than older cows (P < 0.01).  相似文献

Superovulation and embryo transfer records on performance of embryo donor (n = 3,908; beef 99%, dairy 1%) and recipient (n = 19,936; beef 92%, dairy 8%) cattle from a commercial transfer unit were analyzed for environmental effects. Embryos (n = 42,428) were recovered on Days 5 to 8 postestrus from superovulated donors. Numbers of ova, fertilized ova and embryos of transferable quality were recorded. Transferable embryos were classified according to stage of development and morphological quality. Embryos (n = 19,936) were transferred nonsurgically. Responses were modeled with maximum-likelihood procedures using log-linear functions of independent variables and ANOVA. Fluctuations in daily maximum temperature (1 to 43 degrees C), for Days 0 to 7 of embryo development, had no effect on distribution of embryos classified as good (48%), fair (40%) and poor (12%). Temperature did not affect the percentage of donors flushed with recoverable ova (89%), mean number of ova (12.2 +/- 0.3), fertilization rate (76%) or percentage of transferable embryos (57%). Recipient pregnancy rate (56%) was not affected by mean maximum temperature for Days 0 to 10 posttransfer. Interactions between temperature and breed type (dairy vs beef), parity (cow vs heifer), or lactational status (lactating vs dry) on pregnancy rate were recorded. Elevated environmental temperature does not appear to adversely affect reproductive responses of donor and recipient cattle intensely managed in a commercial transfer unit.  相似文献

Forty-one Day 5.0 to Day 5.5 embryos and one unfertilized ovum were recovered nonsurgically from 24 superovulated, parous buffalo (Bubalus bubalis ) and transferred nonsurgically to 28 synchronized recipients by a team of Bulgarian and American scientists. The objective of Experiment I, replicated twice, was to evaluate whether fertility of estrus-synchronized dairy heifers could be improved by postinsemination progesterone supplementation. Estrous cycles were synchronized using two injections of prostaglandin (PG) F(2alpha) adiministered 11 days apart. Heifers displaying estrus were inseminated and assigned to control (n = 155) and treated (n = 159) groups. Treatment consisted of intravaginal insertion of controlled internal drug release (CIDR) devices for Days 7 to 13 (Day 0 = day of estrus). The conception rate for CIDR-treated heifers (57.9%) did not differ significantly from that of the controls (53.6%). The return-to-estrus rate and pattern of return estruses were not affected by treatment, but indicated that early embryonic mortality may have occurred in some of the heifers diagnosed nonpregnant. The objective of Experiment II was to evaluate if used CIDR devices were effective in resynchronizing returns to estrus in previously synchronized inseminated but nonpregnant and noninseminated heifers. Estrous cycles of dairy heifers of breeding age were synchronized with PGF(2alpha). Heifers displaying estrus were assigned to be inseminated (n = 117) or not inseminated (n = 35). All heifers were treated with 9-day used CIDR devices for Days 17 to 22 after synchronized estrus in order to resynchronize returns to estrus. Of the inseminated but nonpregnant heifers (n = 41), 78.1% were detected in estrus after CIDR removal (versus 94.3% of noninseminated heifers [n = 35]; P < 0.05) and 61.0% of the estruses occurred within 4 days of CIDR removal (versus 91.4% of noninseminated; P < 0.05). Estruses of synchronized inseminated nonpregnant heifers occurred over a longer period compared with those of noninseminated heifers (P < 0.025). The results indicate that response to the resynchronization protocol was altered by the outcome (early embryo death or failed fertilization) of the previous unsuccessful insemination, and support the hypothesis that delayed returns to estrus can be attributable to a pregnancy which was initiated but failed to establish itself. Such factors should be considered when evaluating responses of cattle to treatments designed to enhance fertility.  相似文献

The Island Night Lizard (Xantusia riversiana) is endemic to three of the Channel Islands off the coast of California, USA. Introduced species such as goats, sheep, and cats have profoundly affected the fauna and flora of the islands for over 150 years, but most of these non-native species have been recently removed. We measured the distribution of genetic diversity in Island Night Lizards across San Nicolas Island using DNA microsatellites to assess the impacts of historical habitat change on effective population size, gene flow, and population divergence; to provide baseline data for future monitoring of genetic diversity; and to provide recommendations to inform the restoration of degraded habitat. Despite a history of profound anthropogenic habitat disturbance, genetic diversity was high within sites, and there was no evidence of population bottlenecks. Divergence between sites was extraordinarily high, as expected for this sedentary species. Landscape resistance modeling using circuit theory showed that unsuitable habitat is relatively permeable to gene flow compared to suitable habitat, and yet populations separated by very short geographic distances remain genetically distinct. We found no evidence of a need for short-term intervention such as artificial translocations to maintain genetic diversity. Instead, we suggest that management should focus on maintaining, improving, and increasing habitat, especially in creating patches of habitat to link existing sites.  相似文献

Apparent second-order rate constants for complex formation between poly (I) and poly (C) and copolymers of C containing non-complementary I or U residues have been determined spectrophotometrically. The rate constants decrease as the concentration of either I or U in the C strands increases–the effect seems insensitive to the species of residue involved, when differences in the thermal stabilities of the poly (I) poly (C,I) and poly (I). poly (C,U) complexes are taken into account. These results suggest that low concentrations of relatively stable defects can alter the apparent kinetic “complexity” of polynucleotides as determined by hybridization methods (C0t analysis).  相似文献

Fertility in water buffalo (Bubalus bubalis) is considerably lower than that in cattle (Bos taurus and Bos indicus). Poor breeding efficiency is attributed to late onset of puberty, seasonality, poor estrus expression, and long calving intervals. Accurate estrus detection is a prerequisite for efficient reproductive management. Established reproductive management techniques in cattle can be successfully applied to water buffalo because of the similarities in the anatomy, physiology, and endocrinology of reproduction between the two genera.  相似文献
