Mode of formation of cholesta-5,7-dien-3β-ol from cholest-5-en-3β-ol by larvae of Calliphora erythrocephala

1. The conversion of cholest-5-en-3beta-ol (cholesterol) into cholesta-5,7-dien-3beta-ol by axenic Calliphora erythrocephala larvae was demonstrated. 2. The transformation is probably direct (Delta(5)-->Delta(5,7)) and does not involve a Delta(0) intermediate (Delta(5)-->Delta(0)-->Delta(7)--> Delta(5,7)). 3. Delta(7)-bond formation involves the stereospecific elimination of the 7beta hydrogen atom. 4. The relative amounts of free and esterified sterols were determined in larvae grown on cholesterol as sole sterol source and on 5alpha-cholestan-3beta-ol supplemented with minimal amounts of cholesterol. 5. The significance of the results is assessed in relation to the probable role of cholesta-5,7-dien-3beta-ol as an intermediate in the biosynthesis of ecdysones.     

Plasma immunoreactive melanotrophic hormones in patients on maintenance haemodialysis.

Circulating levels of melanotrophic horomones and ACTH were determined in patients treated by maintenance dialysis for chronic renal failure. Plasma melanotrophic hormone levels were greatly increased in all patients studied (125-1100 ng/1 as compared with 12-36 ng/1 in normal adults) and were correlated with the duration of treatment. Skin pigmentation, especially in exposed areas, was notably increased, particularly in those patients with the highest plasma melanotrophic concentrations. Plasma ACTH levels were normal or only slightly raised and circulating corticosteroid concentrations, as determined by a fluorimetric method, all lay within the physiological range. The dissociation between ACTH and melanotrophic hormone levels in these patients may have been the result of a slower metabolic clearance of the latter.     

Immunohistochemical localisation of cyclooxygenase in the human uterus

The distribution in the human uterus of the enzyme cyclooxygenase, involved in prostaglandin synthesis, was studied using the peroxidase-antiperoxidase immunohistochemical technique. Specific staining was observed only during the luteal phase of the menstrual cycle. It was found that cyclooxygenase was localised in the surface and glandular epithelium of the endometrium but could not be demonstrated in the endometrial stroma, myometrium or blood vessels. The location of specific staining for cyclooxygenase and its variation during the menstrual cycle was the same regardless of the amount of menstrual blood loss. Tissue samples obtained from early pregnant and post-menopausal patients failed to exhibit any specific staining.     

Reverse enzyme synthesis in microemulsion-based organo-gels.

Lipase from three different sources has been immobilised in microemulsion-based gels (MBGs) with retention of catalytic activity. Such lipase-containing MBGs prove to be novel solid-phase catalysts for use in apolar organic solvents such as n-heptane. Using these systems, preparative-scale synthesis of a wide variety of esters under mild conditions was possible with products easily isolated and obtained in high yield. Stereoselective esterification of octan-2-ol was observed for all three lipases with Chromobacterium viscosum (CV) lipase yielding product with an enantiomeric excess of 92%. Repeated usage of a CV lipase-containing MBG resulted in a visually unchanged gel whose activity was 75% of the initial value after 30 days. The sectioned MBGs were well suited for use in column flow reactors and were also found to be effective esterification catalysts at temperatures as low as -20 degrees C.     

Multiple sources of evidence for density dependence in the endangered Hawaiian stilt (Himantopus mexicanus knudseni)

Hawaiian stilts (Himantopus mexicanus knudseni) are an endangered subspecies of the Black-necked stilt endemic to the Hawaiian Islands. Despite long-term study, the main drivers of Hawaiian stilt population dynamics are poorly understood. We tested for density dependence using two sources of evidence: a 30-year time series of annual estimated range-wide abundance, and two 15+ year time series of reproductive success. Using separate methods with independent data, sources allowed us to make up for the potentially positive bias of one approach with the more conservative nature of the second. We compared nonlinear density-dependent and density-independent population model fits to our time-series data, using both frequentist and Bayesian state-space approaches. Across both approaches, density-dependent models best fit observed population dynamics, with lower AICc and cross-validation statistics compared to density-independent models. Among density-dependent models, a conditional model in which density-independent dynamics occur below a population size threshold (~850–1,000 birds), and then density-dependent dynamics occur above that threshold, performed best across Bayesian and frequentist model comparisons, with the Ricker model ranked next or equivalently. Our analysis of reproduction data revealed a strong negative effect of local adult density on nest success (proportion of nests hatching at least one chick) at Kealia National Wildlife Refuge on Maui, where few alternative breeding habitats are available, but no such effect at another site where many nearby alternative wetlands are available. These congruent results across independent datasets and analytical approaches support the hypothesis that Hawaiian stilts exhibit density dependence across their range.
