It was previously found that cations introduced into a discontinuous sucrose gradient exert a very pronounced effect on microsomal vesicles, and this principle proved to be effective in microsomal subfractionation. The mechanism of the cation effect was investigated. By using the radioactive isotopes 137Cs and 85Sr, it could be calculated that the amount of ions bound to the various subfractions increases their density by 0.14%, thereby enhancing the sedimentation velocity by only ~7%. In the presence of Cs+ the total volume of the microsomal pellet was decreased by ~15%. Assuming this change in volume to be due to a contraction of the individual vesicles, a roughly 2½-fold increase in sedimentation velocity would be expected. It is further demonstrated, on the basis of light scattering and millipore filtration experiments, that monovalent cations cause an extensive aggregation of rough microsomes and a less pronounced aggregation of smooth microsomes. The mean radius of the sedimenting particles of rough microsomes was found to be at least doubled or trebled in the presence of Cs+, which would give a 4- to 9-fold increase in the sedimentation velocity. Aggregation, therefore, appears to be the main factor in the accelerated sedimentation of rough microsomes in the presence of CsCl. Divalent cations exert a similar effect on a subfraction of the smooth microsomes. Isolated smooth microsomes are very unstable and often exhibit spontaneous aggregation. The presence of attached ribosomes, however, appears to impart greater stability to the rough microsomes as well as increasing their ability to bind monovalent cations. Rough and smooth microsomes and Golgi membranes isolated from rat liver were treated with proteolytic enzymes under conditions which removed 30–40% of the surface proteins without seriously disrupting the membrane structure. This treatment also removed 40–60% of protein-bound mannose, galactose and glucosamine. When protease treatment was combined with neuraminidase treatment, 80% of the sialic acid was removed from intact rough microsomal and Golgi vesicles and about half of the sialic acid of smooth microsomes was solubilized. Recurrent myelitis is one of the predominant characteristics in patients with neuromyelitis optica (NMO). While paresis, visual loss, sensory deficits, and bladder dysfunction are well known symptoms in NMO patients, pain has been recognized only recently as another key symptom of the disease. Although spinal cord inflammation is a defining aspect of neuromyelitis, there is an almost complete lack of data on altered somatosensory function, including pain. Therefore, eleven consecutive patients with NMO were investigated regarding the presence and clinical characteristics of pain. All patients were examined clinically as well as by Quantitative Sensory Testing (QST) following the protocol of the German Research Network on Neuropathic Pain (DFNS). Additionally, plasma endocannabinoid levels and signs of chronic stress and depression were determined. Almost all patients (10/11) suffered from NMO-associated neuropathic pain for the last three months, and 8 out of 11 patients indicated relevant pain at the time of examination. Symptoms of neuropathic pain were reported in the vast majority of patients with NMO. Psychological testing revealed signs of marked depression. Compared to age and gender-matched healthy controls, QST revealed pronounced mechanical and thermal sensory loss, strongly correlated to ongoing pain suggesting the presence of deafferentation-induced neuropathic pain. Thermal hyperalgesia correlated to MRI-verified signs of spinal cord lesion. Heat hyperalgesia was highly correlated to the time since last relapse of NMO. Patients with NMO exhibited significant mechanical and thermal dysesthesia, namely dynamic mechanical allodynia and paradoxical heat sensation. Moreover, they presented frequently with either abnormal mechanical hypoalgesia or hyperalgesia, which depended significantly on plasma levels of the endogenous cannabinoid 2-arachidonoylglycerole (2-AG). These data emphasize the high prevalence of neuropathic pain and hyperalgesia in patients with NMO. The degree of mechanical hyperalgesia reflecting central sensitization of nociceptive pathways seems to be controlled by the major brain endocannabinoid 2-AG. 相似文献

Rough and smooth microsomes and Golgi membranes were incubated with UDP[14C]galactose and the incorporation of radioactivity into the lipid extract and into endogenous protein acceptors were measured. Antagonistic pyrophosphatases were inhibited with ATP and interference from β-galactosidase activity was greatly decreased by carrying out the incubation at pH 7.8. After incubation the particles were centrifuged to remove free oligosaccharide residues. Radioactivity was found in the lipid extract from Golgi membranes but not from rough and smooth microsomes. This radioactivity, however, was not associated with dolichol or retinyl phosphates. The incorporation of radioactivity into proteins of the Golgi fraction was more than double than that of the microsomal fractions. In addition, the transferases in these two types of particles exhibited different properties. Trypsin treatment of intact rough microsomal vesicles, smooth vesicles and Golgi membranes removed about 5, 15 and 50%, respectively, of newly incorporated protein-bound galactose, indicating that the proportion of the newly galactosylated proteins, which are localized at the cytoplasmic surface of the membrane, is lowest in rough microsomes, intermediate in smooth, and highest in Golgi membranes. 相似文献
