An investigation into the population status and distribution of the African buffalo (Syncerus caffer Sparrman, 1779) in Chebera Churchura National Park, Ethiopia, was carried out during the wet and dry seasons of 2012–2015. This study tested the hypothesis that buffalo would demonstrate seasonal habitat preferences and changes in population density. Sample counts were carried out in an area of 1215 km2. The estimated buffalo population was 5193 individuals, with the population density of 4.3/km2. The population showed an increase from 2617 to 5194 individuals during 2006–2015. Males comprised 42.6%, while females 46.7% of the population. Age structure was dominated by adults, which constituted 52.5% of the total population. Subadults comprised 24.3% and young 12.4% of the population. Larger herds of up to 30 individuals were observed during the wet season, and smaller herds of a minimum of four individuals were seen during the dry season. The mean herd sizes during the wet and dry seasons were 29.59 and 16.95, respectively. They were observed more in the riverine vegetation types during the dry season. Of the total, 57.6% utilized riverine habitat during the dry season, whereas 39.8% used this habitat during the wet season. Relative abundance of food sources, green vegetation cover and availability of water were the major factors governing their distribution in the present study area.  相似文献

One of the fundamental questions in animal ecology concerns the activity pattern of animals and the environmental and intrinsic factors that influence such dynamics. This study tested the hypotheses that activity time budgets of the African buffalo appeared to vary by season and times of day and predicted that buffalo would express unequal proportion of time for different activity patterns during the wet and dry seasons in Chebera Churchura National Park (CCNP). An investigation on the diurnal activity budget of the African buffalo was carried out during the wet and dry seasons of 2012–2014 in the Chebera Churchura National Park, Ethiopia, using focal‐animal sampling method. Buffalo spent a greater proportion of the time in feeding and resting/ruminating activities in both the wet and dry seasons. Feeding and resting (lying down and standing) were the predominant activities (87.14% of the diurnal active period), 48.95% time spent feeding during the dry season and 44.91% during the wet season. There was a significant decrease in feeding and an increase in resting from dry seasons to wet seasons. Daytime grazing and resting periods during the wet season were estimated to be 5.39 h and 4.98 h, respectively. Morning and the late afternoon activity peaks were more pronounced during the dry season than the wet season. Therefore, feeding and resting time was influenced by the time of day and the seasons. But there were no significant difference in time allocation for other activities in both the wet and dry seasons. The study has implications for understanding animal activity budget across species, particularly relationships between temperature and season.  相似文献

Wheat is one of the most important crops globally, and its grain is mainly used for human food, accounting for 20% of the total dietary calories. It is also used as animal feed and as a raw material for a variety of non-food and non-feed industrial products such as a feedstock for the production of bioethanol. Starch is the major constituent of a wheat grain, as a result, it is considered as a critical determinant of wheat yield and quality. The amount and composition of starch deposited in wheat grains is controlled primarily by sucrose transport from source tissues to the grain and its conversion to starch. Therefore, elucidation of the molecular mechanisms regulating these physiological processes provides important opportunities to improve wheat starch yield and quality through biotechnological approaches. This review comprehensively discusses the current understanding of the molecular aspects of sucrose transport and sucrose-to-starch metabolism in wheat grains. It also highlights the advances and prospects of starch biotechnology in wheat.  相似文献

The extent of seasonal variation in the nutrient profile of Arthrospira biomass harvested from Lake Chitu was investigated to evaluate the variability of the quality of the product over a period of a year. Protein content varied from 47.9 to 55.7% for wet season biomass samples and from 39.2 to 40.8% for dry season samples. Dry season samples were characterized by relatively higher carbohydrate values (38.0–41.3%). Higher proportion of amino acids and unsaturated fatty acids were recorded for biomass harvested in wet season. Similarly, higher contents of phytonutrients (pigments) were recorded for wet season biomass samples: chlorophyll a (8.2–10.3 mg g?1), phycobiliproteins (104.1–120.7 mg g?1), total carotenoids (3.17–4.31 mg g?1), and β-carotene (1.24–1.61 mg g?1). The contents of Na and K were higher for a dry season biomass whereas other major (Ca, P, Mg) and trace (Mn, Fe, Cu, Zn, Se) minerals were found relatively in higher quantities in a wet season biomass. The nutritional composition of Arthrospira from Lake Chitu was found to be relatively comparable to that found in commercial Arthrospira products in the market. The significance of the findings is discussed in relation to potential sustainable production of Arthrospira biomass from this lake.  相似文献

The binding of ZnO nanoparticles (NPs) and caffeic acid (CFA) was investigated using fluorescence quenching, UV/vis absorption spectrscopy, Fourier transform infrared (FTIR) spectroscopy, transmission electron microscopy (TEM) and dynamic light scattering (DLS) at different temperatures. The study results indicated fluorescence quenching between ZnO NPs and CFA rationalized in terms of a static quenching mechanism or the formation of non‐fluorescent CFA–ZnO. From fluorescence quenching spectral analysis, the binding constant (Ka), number of binding sites (n) and thermodynamic properties were determined. Values of the quenching (KSV) and binding (Ka) constants decrease with increasing temperature and the number of binding sites n = 2. The thermodynamic parameters determined using Van't Hoff equation indicated that binding occurs spontaneously involving the hydrogen bond, and van der Waal's forces played a major role in the reaction of ZnO NPs with CFA. The FTIR, TEM and DLS measurements also indicated differences in the structure, morphology and size of CFA, ZnO NPs and their corresponding CFA–ZnO. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

Genetic improvement programme will only be successful when accompanied by a good understanding of the influence of different environmental factors, knowledge of the genetic parameters and the genetic relationships between the traits of interest. This study aimed to evaluate the influence of non-genetic factors on growth traits and Kleiber ratios and to estimate genetic parameters for early growth traits in Dorper × indigenous crossbred sheep. The effects of fixed factors were analysed by the general linear model procedure of statistical analysis system, while the genetic parameters were estimated using a WOMBAT computer program fitted animal model. The overall least-square mean for birth weight (BRW), weaning weight (3MW), six-month weight, nine-month weight, and yearling weight were 3.03 ± 0.02, 14.5 ± 0.18, 20.4 ± 0.26, 24.8 ± 0.31, and 28.3 ± 0.40 kg, respectively. The overall least-square mean for Kleiber ratio from birth to weaning (KR1), weaning to six months, six to nine months and nine months to yearling age were 16.8 ± 0.10, 6.41 ± 0.17, 4.55 ± 0.21 and 3.38 ± 0.20 g/kg of metabolic weight, respectively. The inclusion of maternal genetic effect had a significant influence on BRW, and it explains 20% of the phenotypic variation. The total heritability estimates for BRW, 3MW, birth to weaning average daily weight gain and KR1 were 0.10, 0.14, 0.16 and 0.12, respectively. The phenotypic correlation varied from ?0.11 ± 0.05 to 0.98 ± 0.02, whereas the direct genetic correlation ranged from ?0.32 ± 0.40 to 0.98 ± 0.17. The mean inbreeding coefficient was 0.105% with an annual rate of 0.02%. The heritability estimates for growth traits and Kleiber ratio suggest that slow genetic progress would be expected from the selection. However, the integration of selection with crossbreeding programme can enhance genetic gain. Therefore, selection should be conducted based on breeding values estimated from multiple information sources to increase the selection response.  相似文献

Methanococcus thermolithotrophicus is a methanogenic archaebacterium that can use either H2 or formate as its source of electrons for reduction of CO2 to methane. Growth and suspended-whole-cell experiments show that H2 plus CO2 methanogenesis was constitutive, while formate methanogenesis required adaptation time; selenium was necessary for formate utilization. Cells grown on formate had 20 to 100 times higher methanogenesis rates on formate than cells grown on H2-CO2 and transferred into formate medium. Enzyme assays with crude extracts and with F420 or methyl viologen as the electron acceptor revealed that hydrogenase was constitutive, while formate dehydrogenase was regulated. Cells grown on formate had 10 to 70 times higher formate dehydrogenase activity than cells grown on H2-CO2 with Se present in the medium; when no Se was added to H2-CO2 cultures, even lower activities were observed. Adaptation to and growth on formate were pH dependent, with an optimal pH for both about one pH unit above that optimal for H2-CO2 (pH 5.8 to 6.5). When cells were grown on H2-CO2 in the presence of formate, formate (greater than or equal to 50 mM) inhibited both growth and methanogenesis at pH 5.8 to 6.2, but not at pH greater than 6.6. Both acetate and propionate produced similar inhibition. Formate inhibition was also observed in Methanospirillum hungatei.  相似文献

A variety of sulfur-containing compounds were investigated for use as medium reductants and sulfur sources for growth of four methanogenic bacteria. Sulfide (1 to 2 mM) served all methanogens investigated well. Methanococcus thermolithotrophicus and Methanobacterium thermoautotrophicum Marburg and delta H grew well with S0, SO3(2-), or thiosulfate as the sole sulfur source. Only Methanococcus thermolithotrophicus was able to grow with SO4(2-) as the sole sulfur source. 2-Mercaptoethanol at 20 mM was greatly inhibitory to growth of Methanococcus thermolithotrophicus on SO4(2-) or SO2(2-) and Methanobacterium thermoautotrophicum Marburg on SO3(2-) but not to growth of strain delta H on SO3(2-). Sulfite was metabolized during growth by Methanococcus thermolithotrophicus. Sulfide was produced in cultures of Methanococcus thermolithotrophicus growing on SO4(2-), SO3(2-), thiosulfate, and S0. Methanobacterium thermoautotrophicum Marburg was successfully grown in a 10-liter fermentor with S0, SO3(2-), or thiosulfate as the sole sulfur source.  相似文献

Samples of human dental plaque were examined for the presence of methanogenic bacteria. Of 54 samples from 36 patients, 20 yielded H2/CO2-using methanogenic enrichment cultures. All methanogen-positive samples were from patients with some degree of periodontal disease. The predominant populations in the enrichments had morphologies characteristic of Methanobrevibacter spp. In six enrichments derived from three patients, the common methanogen was antigenically similar to Methanobrevibacter smithii. The same was true for the three methanogenic isolates obtained in axenic culture from a fourth patient. The six enrichments and two of the three isolates were antigenically closer to strain ALI than to PS. Two of the enrichments also had subpopulations with weak antigenic similarity to Methanosphaera stadtmanae. The data indicate that methanogens in the oral cavity of humans are antigenically close to those found in the intestinal tract.  相似文献

Life-threatening disseminated infection withTrichosporon beigelii (trichosporonosis) is a rare mycosis most commonly seen in patients with hematologic malignancies made neutropenic by cytotoxic therapy. This infection is usually resistant to conventional antifungal therapies. Poor correlation between therapeutic outcome of trichosporonosis and in vitro susceptibility of clinical isolates ofT. beigelii to antifungal agents is often reported. To obtain a better understanding of its pathogenesis, and to aid in the future study of the therapy of this disease, a murine model of trichosporonosis was developed. The in vitro growth of clinical isolates ofT. beigelii was first studied. Subsequently, mice made neutropenic with cyclophosphamide were inoculated intravenously with the fungus to produce the disease model. Inoculum size which produced 100% mortality, yet allowed an apparent therapeutic window (6×106) was determined. Tissue distribution and burden of organism during the course of infection was examined by viability and histopathologic studies.T. beigelii disseminated rapidly in this model, involving numerous organs including the heart, brain, kidneys, lungs, and liver. The heart and kidneys of the infected animals showed evidence of infection as early as 6 hours following inoculation. Further understanding of the pathogenesis of trichosporonosis in the neutropenic host was imparted by this study. This will aid in the future study of antibiotic treatment of this disease and its untreated progression.  相似文献
